4-methyl-5-vinylthiazole (MVT) is a significant volatile of caladium (Caladium bicolor) which produces a very high level of thiamin thiazole synthase (THI4) in male flowers. We explored transcriptomes upregulating MVT using RNA-seq during the six developmental stages of the male flower (Day−10 to Day0) in C. bicolor ‘Tapestry’. THI4 was the highest transcript throughout the male flower development. Additionally, the genes showing the high expression associated with floral volatiles of caladium on Day0 were trans-resveratrol di-O-methyltransferase (ROMT), chalcone synthase (CHS), 3-ketoacyl-CoA thiolase 2 (KAT2), and linalool synthase (TPS). These four genes correspond to the following elevated volatiles of caladium: 1,3,5-trimethoxybenzene, MVT, indole, methyl salicylate, and linalool on Day0 compared to Day−10. The upstream THI4 gene was cloned to drive a fluorescent gene (ZsGreen1) in transient and stable transgenic petunia and tobacco plants, showing the gene expression only in the male tissue. The tissue-specific expression of the caladium THI4 promoter could benefit crop production with minimal modification of plants. Investigating transcriptomes associated with caladium fragrance can help provide insight into understanding the regulatory mechanisms of floral volatiles of caladium. Full article

Comprising the largest population cohort on this planet, Gen Z presents a future-oriented consumer segment driven by climate change and food. This study sought to investigate Gen Z’s perceptions toward plant-based foods and diets and explore the relationship that attitude components, meal preparation involvement, personal and lifestyle factors, and perceived barriers in adopting a plant-based diet have with willingness to adopt green-eating practices. Using cross-sectional data from university students in Greece, India, and the UK, various tools were employed to determine the factors influencing youths’ consumer behavior toward animal-protein substitutes. PCA indicated the underlying dimensions of students’ viewpoints on plant-based foods, whereas hierarchical and k-means clustering provided the cluster structure. An ordered probit model was estimated to delineate Gen Z’s willingness to adopt plant-based diets and distinguish among mostly unwilling, somewhat willing, and mostly willing youths. Our findings identified two consumer segments, namely proponents and opponents of plant-based foods and diets, with statistically significant differences in the perceived health benefits of plant-based diets, attachment to animal-based proteins, perceived exclusion of animal-based foods, dissatisfaction with plant-based foods’ attributes, and demand for ensuring adequate protein intake. The ordered probit model estimates showed that there is a “homogeneity” in the factors influencing youths’ intention to adopt plant-based diets, with attitude components, meal preparation indicators, perceived barriers to eating “green”, and personal factors, such as self-assessed knowledge of healthy eating and physical activity, being strongly associated with students’ willingness to switch to plant-based diets in all three countries. Mapping potential obstacles and enablers in terms of shifting to more green-eating behaviors, our findings could add information to better understand the factors affecting food choice and youths’ transition to a more sustainable lifestyle. This study investigates the sustainable purchasing behaviors of Generation Z in Saudi Arabia, providing an insightful exploration by integrating the theory of planned behavior and social identity approach. Through qualitative research, the study uncovers nuanced insights into the motivational factors and social dynamics influencing eco-friendly consumer choices among young adults. It highlights the importance of environmental consciousness, peer influence, and identity-based motivations in shaping sustainable purchasing patterns. The findings reveal that Generation Z’s decisions are significantly influenced by a combination of personal attitudes, perceived social norms, and identity-related aspirations towards sustainability. This research broadens the current understanding of green consumption in a culturally specific context, suggesting that young consumers’ sustainable behaviors are deeply intertwined with their social identities and environmental awareness. The implications of these findings are significant for policymakers, marketers, and manufacturers aiming to encourage eco-friendly practices among younger generations, highlighting the need for tailored strategies that resonate with their values and social identities. This study examined eco-friendly practices (EFPs), green image, and brand attachment to identify antecedents that affect coffee shop consumers’ brand advocacy and brand loyalty, targeting Generation Z, who are emerging as the primary agents of eco-friendly consumption. In particular, we applied only the tangible EFPs that consumers can observe to identify a clear relationship between perceptions of EFPs and consumer behavior. This is the most significant difference from previous studies. In this study, 287 people who responded that they had visited Starbucks, which was selected as the coffee shop brand to be investigated through a pilot test, were used as a sample. As a result of the verification, EFPs demonstrated a positive influence on green image and brand attachment, and green image appeared to have a positive effect on brand attachment. Brand attachment was confirmed to be an antecedent that strengthens brand advocacy and brand loyalty. This study contributes to the literature on environmental friendliness in the hospitality industry and can be used to establish sustainable, eco-friendly marketing strategies in the food service business. This study aims to investigate the induction effect of LncRNA-CIR6 on MSC differentiation into cardiogenic cells in vitro and in vivo. In addition to pretreatment with Ro-3306 (a CDK1 inhibitor), LncRNA-CIR6 was transfected into BMSCs and hUCMSCs using jetPRIME. LncRNA-CIR6 was further transfected into the hearts of C57BL/6 mice via 100 μL of AAV9-cTnT-LncRNA-CIR6-ZsGreen intravenous injection. After three weeks of transfection followed by AMI surgery, hUCMSCs (5 × 105/100 μL) were injected intravenously one week later. Cardiac function was evaluated using VEVO 2100 and electric mapping nine days after cell injection. Immunofluorescence, Evans blue-TTC, Masson staining, FACS, and Western blotting were employed to determine relevant indicators. LncRNA-CIR6 induced a significant percentage of differentiation in BMSCs (83.00 ± 0.58)% and hUCMSCs (95.43 ± 2.13)% into cardiogenic cells, as determined by the expression of cTnT using immunofluorescence and FACS. High cTNT expression was observed in MSCs after transfection with LncRNA-CIR6 by Western blotting. Compared with the MI group, cardiac contraction and conduction function in MI hearts treated with LncRNA-CIR6 or combined with MSCs injection groups were significantly increased, and the areas of MI and fibrosis were significantly lower. The transcriptional expression region of LncRNA-CIR6 was on Chr17 from 80209290 to 80209536. The functional region of LncRNA-CIR6 was located at nucleotides 0–50/190–255 in the sequence. CDK1, a protein found to be related to the proliferation and differentiation of cardiomyocytes, was located in the functional region of the LncRNA-CIR6 secondary structure (from 0 to 17). Ro-3306 impeded the differentiation of MSCs into cardiogenic cells, while MSCs transfected with LncRNA-CIR6 showed a high expression of CDK1. LncRNA-CIR6 mediates the repair of infarcted hearts by inducing MSC differentiation into cardiogenic cells through CDK1. Full article

The COVID-19 pandemic and the consequent emergence of new SARS-CoV-2 variants of concern necessitates the determination of populational serum potency against the virus. Here, we standardized and validated an imaging-based method to quantify neutralizing antibodies against lentiviral particles expressing the spike glycoprotein (pseudovirus). This method was found to efficiently quantify viral titers based on ZsGreen-positive cells and detect changes in human serum neutralization capacity induced by vaccination with up to two doses of CoronaVac, Comirnaty, or Covishield vaccines. The imaging-based protocol was also used to quantify serum potency against pseudoviruses expressing spikes from Delta, Omicron BA.1.1.529, and BA.4/5. Our results revealed increases in serum potency after one and two doses of the vaccines evaluated and demonstrated that Delta and Omicron variants escape from antibody neutralization. The method presented herein represents a valuable tool for the screening of antibodies and small molecules capable of blocking viral entry and could be used to evaluate humoral immunity developed by different populations and for vaccine development. Financial technology is quickly developing, making the financial industry more accessible and encouraging individual investor engagement in the investing process. Generation Z, characterised by a high level of digital literacy, curiosity, and receptivity to innovation, tends to very quickly make decisions and rapidly consume. Since 2007, there has been an increase in the number of articles analysing investor behaviour, drawing on insights from financial and psychological theories. The purpose of this exploratory study is to categorise the behaviour of students surveyed by the type of their investments, while at the same time assessing their willingness to choose green investments. The survey used in the analysis not only aims at collecting data but also educates students on the importance of critical self-awareness and the identification of their emotions to make rational, responsible investment decisions and, at the same time, to form a responsible investor who understands that investing is not only a way to earn a return but also can make a positive impact on the world when green investments are chosen. This study shows that studying students tend to be very rational and interested in contributing to greening the world; however, they are still hesitant to put their theoretical skills into practise and are more likely to provide theoretical support for green investments rather than actually invest. Respondents are grouped according to their potential investment behaviour. The proportions of groups are assessed using statistical inference with a precision of 95% that allowed to propose the method of deriving confidence intervals for each group estimation and, thus, making estimates both reliable and available as statistical estimations. This study addresses the motivations behind Generation Z’s attendance at conferences and aims to contribute to a better understanding of their conference-participation behavior. The study identified six latent factors that influence conference-attendance motivation: green and digital conference experience, educational and professional opportunities, conference costs, destination and conference stimuli, stimulating factors, and conference accessibility. These factors align with Generation Z’s unique characteristics and preferences, such as their environmental consciousness, their reliance on digital technologies, and their desire for continuous learning, career advancement, and experiences. The study also found that destination characteristics and educational and professional opportunities are significant motivators for conference attendance by members of Generation Z across different cultural contexts. Additionally, their strong interest in sustainability and financial challenges highlights the need for eco-friendly practices and affordable registration fees to make conferences more appealing and accessible. The study further explores the impact of socio-demographic characteristics, revealing gender and past-conference-attendance differences in motivation. Finally, the study discusses the implications of its findings for the conference industry and suggests areas for future research to enhance the understanding of conference-attendance motivation among members of Generation Z. We studied the relationship between neuronal NO synthase (nNOS) expression and capillarity in the tibialis anterior (TA) muscle of mice subjected to treadmill training. The mRNA (+131%) and protein (+63%) levels of nNOS were higher (p ≤ 0.05) in the TA muscle of C57BL/6 mice undergoing treadmill training for 28 days than in those of littermates remaining sedentary, indicating an up-regulation of nNOS by endurance exercise. Both TA muscles of 16 C57BL/6 mice were subjected to gene electroporation with either the pIRES2-ZsGreen1 plasmid (control plasmid) or the pIRES2-ZsGreen1-nNOS gene-inserted plasmid (nNOS plasmid). Subsequently, one group of mice (n = 8) underwent treadmill training for seven days, while the second group of mice (n = 8) remained sedentary. At study end, 12–18% of TA muscle fibers expressed the fluorescent reporter gene ZsGreen1. Immunofluorescence for nNOS was 23% higher (p ≤ 0.05) in ZsGreen1-positive fibers than ZsGreen1-negative fibers from the nNOS-transfected TA muscle of mice subjected to treadmill training. Capillary contacts around myosin heavy-chain (MHC)-IIb immunoreactive fibers (14.2%; p ≤ 0.05) were only higher in ZsGreen1-positive fibers than ZsGreen1-negative fibers in the nNOS-plasmid-transfected TA muscles of trained mice. Our observations are in line with an angiogenic effect of quantitative increases in nNOS expression, specifically in type-IIb muscle fibers after treadmill training. Full article

Using PLS-SEM, this article proposes and verifies a model among Gen Z that captures the relationship between attitudes towards the environmental ecosystem and green tourism, personal and social norms regarding pro-environmental behaviour, perceived behavioural control, perceived green image of destinations (PGID), behavioural intentions regarding green holiday destinations (GHD), and willingness to pay (WTP) more for visiting them. The paper also verifies whether intercultural differences exist in the relationships between these variables. The most important results indicate that (1) for Gen Z, the perceived green image of destinations has the strongest impact on intention to travel to green holiday destinations; (2) the proposed variables explain the willingness to visit green holiday destinations to a much greater extent than the WTP a higher price for such trips. This study contributes to the literature concerning generational changes in tourism, pro-environmental (transition) planning, and the growing green economy and marketing. Full article

Generation Z represents the young people of today. They are considered as “digitally literate” and were born between mid-to-late 1990s to early 2000s. Generation Z pays more attention to popular environmental issues such as global warming, high energy consumption, overgrazing, and university social responsibility (USR), which are present around the world. We formed a double moderated mediation exam from 910 college students in southeast China, used a new notion “green psychological capital”, and proposed it as a vital mediator. In addition, we found that green organizational ambidexterity and environmental attitude are both boundary conditions in the green shared vision organizational citizenship behavior for the environment (OCBE) link. These findings have unlocked a deeper insight into Generation Z’s green conception and offered a more comprehensive investigation on USR research. Furthermore, the amazing findings can provide a worldwide blueprint for USR studies in the long term. Full article

Leaf senescence is the final stage of leaf development and is essential for storage properties and crop productivity. WRKY transcription factors have been revealed to play crucial roles in several biological processes during plant growth and development, especially in leaf senescence. However, the functions of Brassica napus WRKY transcription factors in leaf senescence remain unclear. In the present study, Bna.A07.WRKY70, one paralogue of Brassica napus WRKY70, was cloned from the B. napus cultivar “Zhongshuang11 (ZS11)”. We found that Bna.A07.WRKY70 contains a highly conserved WRKY domain and is most closely related to Arabidopsis thaliana WRKY70. The subcellular localization and transcriptional self-activation assays indicated that Bna.A07.WRKY70 functions as a transcription factor. Meanwhile, RT-qPCR and promoter-GUS analysis showed that Bna.A07.WRKY70 is predominantly expressed in the leaves of B. napus and rosette leaves of A. thaliana. In addition, our results demonstrated that ectopic expression of Bna.A07.WRKY70 in A. thaliana wrky70 mutants could restore the senescence phenotypes to wild-type levels. Consistently, the expression levels of three senescence-related marker genes of wrky70 mutants were restored to wild-type levels by ectopic expression of Bna.A07.WRKY70. These findings improve our understanding of the function of Bna.A07.WRKY70 in B. napus and provide a novel strategy for breeding the new stay-green cultivars in rapeseed through genetic manipulation. Abstract
The purpose of the study is to investigate how environmental concern, eco-labelling, influencers and user-generated content affect Generation Z’s green purchase intention. The purpose of the study is to investigate how environmental concern, eco-labelling, influencers and user-generated content affect Generation Z’s green purchase intention. The objective of this study is to contribute with a new scope that combines influencers and user-generated content on digital platforms with environmental concern for Generation Z. The study also aims to add new value in predicting Generation Z’s green purchase intention and results that can be implemented in future marketing strategies. To test the framework, a quantitative research approach, with an online survey, was applied to collect data from Generation Z. The sample size consisted of 393 individuals from Generation Z. Structural Equation Modelling was applied to test the hypothesized framework. All hypotheses were accepted, and hence, this research has identified key variables to predict Generation Z’s green purchase intention. Additionally, this paper found that environmental concern has a significant positive impact on Generation Z’s user-generated content and eco-labelling, and influencers positively affect Generation Z’s user-generated content. This study can aid companies that employ an influencer marketing approach to comprehend how they can motivate customers to buy sustainable products more frequently. This study provides crucial and valuable insights into further understanding how the sustainable consumption behavior of Generation Z can be impacted by the utilization of influencer marketing and their concern for the environment. It also provides a deeper understanding of how influencers and their perceived concerns for the environment can be combined with user-generated content and eco-labelling, as well as subsequent effects on the green purchase intention of members of Generation Z. AC loads may demand a fixed or variable voltage at their input terminals. When using inverters to power such loads, the response of the inverter must be precisely controlled to suit the demands of the AC loads. Inverters with higher efficiency and sensitivity will play an increasingly essential role as the need for solar PV applications in prospective green technology grows. To increase power quality and provide a reliable power source, an inverter architecture with harmonic reduction approaches is proposed. The multilevel inverter (MLI), unlike conventional inverters, is developed by cascaded single inverter units and is often used to connect renewable energy sources. As a result, they can be utilized to efficiently reduce harmonics. Among the three topologies, the most widely used in industries is the neutral-point clamped MLI. When the levels are raised, however, they demand a larger number of diodes. When the level of the flying capacitor exceeds three, several capacitors are necessary. As a result, the optimum option for synthesizing the right output voltage from several DC sources is a cascaded multilevel inverter (CMLI). Each link in a CMLI is connected by a single DC source; therefore, there is no voltage imbalance. However, getting equal DC voltages at the input of each unit is once again a limitation. In this work, various existing multilevel inverter topologies including hybrid topologies with different switching strategies are investigated and reported. The performance of a solar PV-based seven-level quasi-Z-source cascaded H-Bridge MLI (qZS-CHBMLI) has been thoroughly examined with the best switching scheme and best topology of multilevel inverters using MATLAB/Simulink. Inteins (intervening proteins) are polypeptides that interrupt the sequence of other proteins and remove themselves through protein splicing. In this intein-catalyzed reaction, the two peptide bonds surrounding the intein are rearranged to release the intein from the flanking protein sequences, termed N- and C-exteins, which are concurrently joined by a peptide bond. Because of this unique functionality, inteins have proven exceptionally useful in protein engineering. Previous work has demonstrated that heterologous proteins can be inserted within an intein, with both the intein and inserted protein retaining function, allowing for intein-containing genes to coexpress additional coding sequences. Here, we show that a fluorescent protein (ZsGreen) can be inserted within the Pyrococcus horikoshii RadA intein, with the hybrid protein (ZsG-Int) maintaining fluorescence and splicing capability. We used this system to create a recombinant Ebola virus expressing a fluorescent protein. We first tested multiple potential insertion sites for ZsG-Int within individual Ebola virus proteins, identifying a site within the VP30 gene that facilitated efficient intein splicing in mammalian cells while also preserving VP30 function. Next, we successfully rescued a virus containing the ZsG-Int-VP30 fusion protein, which displayed fluorescence in the infected cells. We thus report a new intein-based application for adding reporters to systems without the need to add additional genes. Further, this work highlights a novel reporter design, whereby the reporter is only made if the protein of interest is translated and does not remain fused to the protein of interest. Full article
