The Global Polio Eradication Initiative was adopted by Ghana in 1996, and through robust AFP surveillance was able to interrupt the circulation of wild poliovirus in 2008. However, the country suffered vaccine-derived poliovirus type 2 outbreaks in 2019 and 2022. We conducted a retrospective analysis of all AFP surveillance data received by the polio program in Ghana from 2018 to 2022. An analysis of the WHO performance indicators for evaluating a surveillance system was conducted using Epi Info 3.5.4 and Microsoft Excel. Of the 4832 cases investigated, 56.3% were males, 71.1% comprised children aged 5 years and below, and more than half (65.2%) had received a maximum of three doses of OPV. Over 77% (3028) had a fever at the onset of paralysis, and 67.8% had paralysis progression within 3 days. The non-polio AFP rate of ≥2 and the stool adequacy rate exceeded the target of ≥80% in nearly every study year. The proportion of non-polio enteroviruses isolated surpassed the target of ≥10% in all years except 2018. The AFP surveillance system in Ghana is sensitive and representative. Though the surveillance became more intensive and proactive during the outbreak, the system needs to focus on improving the completeness of the data as well as the timeliness of the arrival of stool specimens within 3 days of collection. Enterovirus A71 (EV-A71) is a major cause of hand-foot-and-mouth disease (HFMD), particularly in cases that involve complications affecting the nervous system or cardiopulmonary function. In South America, EV-A71 has primarily been identified through studies of acute flaccid paralysis (AFP) and other neurological disorders. In September 2022, two children from a small city in Uruguay were hospitalized with presumptive rhombencephalitis, exhibiting symptoms of HFMD. EV-A71 was identified through RT-PCR and next-generation sequencing of stool and skin lesion samples. A maximum-likelihood phylogenetic analysis of the P1 coding region classified the Uruguayan strains as part of an emerging lineage, primarily reported in Europe over the past decade, known as the C1-like lineage. The findings presented here represent the first detection of the EV-A71 C1-like lineage in cases of HFMD and encephalitis reported from South America, underscoring the urgent need to enhance surveillance for HFMD, aseptic meningitis, encephalitis, and AFP, in countries facing challenges in establishing effective surveillance programs related to enteroviruses and associated diseases. Background: Liver transplantation (LT) is a method for treating hepatocellular carcinoma (HCC) with satisfactory outcomes. One of the novel methods for predicting LT outcomes is the Metroticket 2.0 model. The disease in patients initially within the Milan criteria (MC) may progress while on a transplantation waitlist; thus, various transplantation bridging therapy (BT) methods are proposed for patients to stay within the MC and optimize the LT outcome. Methods: We performed a retrospective analysis of patients who underwent LT for HCC at an oncological and transplantation center in northern Poland. Patients who underwent (n = 10) or did not undergo (n = 11) BT were included. The primary endpoints of the study were mortality among the patients, HCC recurrence, and Metroticket 2.0 scores based on LT qualification results and explant pathology outcomes. The median follow-up length was 44.03 months. Results: Patients who underwent BT had significantly lower Metroticket 2.0 scores and greater AFP concentrations at baseline. At LT, there was no significant difference in Metroticket 2.0 scores or AFP concentrations between the groups. Explant Metroticket 2.0 scores were significantly lower in patients who received BT. A complete pathologic response was achieved in 30.0% of patients who underwent BT. The recurrence-free survival rates were 100% and 90.91% in patients who underwent and did not undergo BT, respectively. Overall survival was 80.0% and 81.81% in patients who underwent and did not undergo BT, respectively. Conclusions: BT should be considered only as a means of remaining within the LT criteria. Routine BT does not appear to be justified for LT patients. Full article

Slow-release urea, as an environmentally friendly fertiliser, can provide a continuous and uniform supply of nutrients needed by the crop, reduce the amount and frequency of fertiliser application, and promote the uptake and utilisation of nitrogen in crops. The production of E. ferox is often dominated by the application of quick-acting fertilisers, resulting in serious problems of over-fertilisation, inappropriate periods of fertilisation, eutrophication of soil and water due to fertilisation, and difficulties in applying fertilisers. Therefore, in this study, different amounts (CK, T1, T2, T3, T4, T5) of SRU (Slow-release Urea) were first applied, and T3 (18.8 kg·667 m−2) was found to significantly improve both yield and quality. Further, it was found that under different SRU (CK, S1, S2, S3, S4) application period treatments, application of 18.8 kg·667 m−2 at AFP20 (S2) period significantly increased the yield and quality of E. ferox. In the seed kernels of E. ferox, the total yield, soluble sugar content, total starch, and flavonoid content increased significantly by 10.35%, 36.40%, 5.91%, and 22.80%, respectively, compared with CK. In addition, the expression of key sugar transporter genes (EfSWEETs), flavonoid synthesis-related genes (EfPAL, EfDFR, etc.), and starch synthesis-related enzyme activities (SBE, SSS, GBSS) were significantly increased. By exploring the quantity of application and application period of SRU, this study was carried out to investigate the in-depth effect of SRU on the growth and development of E. ferox and to provide technical references for the increase in E. ferox yield, the improvement in E. ferox quality, and the simplification of fertiliser application. The construction level of artificial grassland is an important index of the development degree of grassland animal husbandry. Therefore, improving the productivity level of artificial grassland and promoting the sustainable utilization of artificial grassland have become important tasks that need to be urgently addressed. There have been numerous studies on the effects of monoculture on the soil microbial community structure in artificial grassland, but there is limited research on the effects of mixed sowing on the soil microbial community structure and the related patterns. In this study, Elymus nutans (En), Festuca sinensis (Fs), Avena sativa (As), and Poa pratensis (Pp) were used as common herbage materials in an alpine grassland pastoral area of the eastern Tibetan Plateau. Multi-density monoculture and mixed seeding were employed to establish artificial grassland communities with varying structures. By comparing the soil microbial community structure of the differently treated artificial planting grass, degraded grassland with bald spots, and natural grassland, it was confirmed that plant community diversity significantly influences the microbial community structure. The high-density planting treatment of multiple forage grasses had a more pronounced impact on the soil microbial community structure compared to that of the high-density planting treatment of a single variety of forage grass. The soil microbial community diversity index of the four mixed-planting treatments was higher than those of the other artificial grassland treatments and the natural grassland treatments, and the soil microbial community structure was most similar to that of the natural grassland. Avena sativa planting increased the abundance of Actinobacteria and Basidiomycota and decreased the number of Acidobacteria by increasing the soil pH value. The AFP (As+Fs+Pp) treatment reduced the proportion of Mortierellomycota in the soil by decreasing the content of available phosphorus. The AEFP (As+En+Fs+Pp) treatment increased the number of Proteobacteria by raising the soil total phosphorus content and reduced the abundance of Acidobacteria by lowering the soil pH value. Additionally, a machine learning method was used to evaluate the comprehensive performance of 21 artificial grassland treatments on nine soil physical and chemical properties. It was found that the AEFP mixed-planting and high-density planting treatments had the greatest improvement effect on the nine soil physical and chemical properties, which was conducive to sustainable land use. Background/Objectives: We evaluated the relationship between the neoangiogenic transcriptomic signature (nTS) and clinical symptoms, treatment outcomes, and survival in hepatocellular carcinoma (HCC) patients. Methods: This study prospectively followed 328 patients in the derivation and 256 in the validation cohort (with a median follow-up of 31 and 22 months, respectively). The nTS was associated with disease presentation, treatments administered, and overall survival rates. Additionally, this study investigated how multiple treatments influenced changes in nTS status and alterations in microRNA expression. Results: The nTS was identified in 27.4% of patients, linked to aggressive features like multifocality and elevated alpha-fetoprotein (AFP), a pattern consistent with that of the validation cohort. Most patients in both cohorts received treatment for HCC. nTS+ patients had limited access to, and benefited less from, liver transplantation or radiofrequency ablation (RFA) compared to nTS− patients. By the end, 78.9% had died, with nTS− patients showing better median survival and response to treatments than their nTS+ counterparts, who had lower survival across all treatment types. Among those who received transarterial chemoembolization (TACE), 31.2% (21/80 patients after the initial treatment and another four following a second TACE) transitioned from an nTS− to an nTS+ status. This shift was associated with lower survival and alterations in microRNA expressions related to oncogenic pathways. Conclusions: The nTS markedly influences treatment eligibility and survival in patients with HCC. Notably, the nTS can develop after repeated TACE procedures, significantly impacting patient survival and altering oncogenic microRNA expression patterns. These findings highlight the critical role of the nTS in guiding treatment decisions and prognostication in HCC management. Full article

Innate immunity, the body’s initial defense against bacteria, fungi, and viruses, heavily depends on antimicrobial peptides (AMPs), which are small molecules produced by all living organisms. Insects, with their vast biodiversity, are one of the most abundant and innovative sources of AMPs. In this study, AMPs from the red palm weevil (RPW) Rhynchophorus ferrugineus (Coleoptera: Curculionidae), a known invasive pest of palm species, were examined. The AMPs were identified in the transcriptomes from different body parts of male and female adults, under different experimental conditions, including specimens collected from the field and those reared in the laboratory. The RPW transcriptomes were examined to predict antimicrobial activity, and all sequences putatively encoding AMPs were analyzed using several machine learning algorithms available in the CAMPR3 database. Additionally, anticancer, antiviral, and antifungal activity of the peptides were predicted using iACP, AVPpred, and Antifp server tools, respectively. Physicochemical parameters were assessed using the Antimicrobial Peptide Database Calculator and Predictor. From these analyses, 198 putatively active peptides were identified, which can be tested in future studies to validate the in silico predictions. Genome-wide analysis revealed that several AMPs have predominantly emerged through gene duplication. Noticeably, we detect a newly originated defensin allele from an ancestral defensin via the deletion of two amino acids following gene duplication in RPW, which may confer an enhanced resilience to microbial infection. Our study shed light on AMP gene families and shows that high duplication and deletion rates are essential to achieve a diversity of antimicrobial mechanisms; hence, we propose the RPW AMPs as a model for exploring gene duplication and functional variations against microbial infection. Antifreeze agents play a critical role in various fields including tissue engineering, gene therapy, therapeutic protein production, and transplantation. Commonly used antifreeze agents such as DMSO and other organic substances are known to have cytotoxic effects. Antifreeze proteins sourced from cold-adapted organisms offer a promising solution by inhibiting ice crystal formation; however, their effectiveness is hindered by a dynamic ice-shaping (DIS) effect and thermal hysteresis (TH) properties. In response to these limitations, antifreeze peptides (AFPs) have been developed as alternatives to antifreeze proteins, providing similar antifreeze properties without the associated drawbacks. This review explores the methods for acquiring AFPs, with a particular emphasis on chemical synthesis. It aims to offer valuable insights and practical implications to drive the realm of sub-zero storage. Full article

Circulating miR-18a and miR-532 Levels in Extrahepatic Cholangiocarcinoma

Background: Cholangiocarcinoma (CCA) is a highly aggressive cancer of the bile ducts with a poor prognosis and limited diagnostic markers. This study aims to investigate the potential of miR-18a and miR-532 as biomarkers for CCA by exploring their correlations with clinical parameters and traditional tumor markers such as CA19.9, CEA, and AFP. Methods: This study involved a cohort of patients diagnosed with CCA. Serum levels of miR-18a and miR-532 were measured and analyzed in relation to various clinical parameters, including age, tumor markers, and histological features. Results: Serum levels of miR-18a and miR-532 were upregulated in patients with extrahepatic cholangiocarcinoma (eCCA) compared to healthy controls (p < 0.05). MiR-18a and miR-532 levels were correlated with each other (p = 0.011, Spearman’s rho = 0.482) but showed no significant correlation with age or traditional tumor markers (CA19.9, CEA, AFP). No significant differences in miR-18a and miR-532 levels were observed concerning tumor localization or histological grading. For predicting tumor resectability, miR-532 at a cut-off point of 2.12 showed a sensitivity of 72.73%, specificity of 81.25%, and an AUC of 71.3%, while miR-18a, at a cut-off of 1.83, had a sensitivity of 63.64%, specificity of 75%, and an AUC of 59.7%. ROC curve analysis suggested moderate diagnostic potential for miR-18a and miR-532, with AUC values of 0.64 and 0.689, respectively. Conclusions: Although miR-18a and miR-532 showed significant upregulation in eCCA patients compared to healthy controls, they did not demonstrate significant associations with key clinical parameters, limiting their effectiveness as standalone diagnostic biomarkers. Further research involving larger, multi-center cohorts and additional molecular markers is necessary to validate these findings and explore the broader diagnostic potential of miRNAs in CCA. Aim: To investigate the characteristics and prognosis of patients with advanced hepatocellular carcinoma (HCC) treated with atezolizumab and bevacizumab (Atz/Bev) who achieved a complete response (CR) according to the modified Response Evaluation Criteria in Solid Tumors (mRECIST). Methods: A total of 120 patients with Eastern Cooperative Oncology Group performance status (PS) 0 or 1 and Child–Pugh A at the start of Atz/Bev treatment were included. Barcelona Clinic Liver Cancer stage C was recorded in 59 patients. Results: The CR rate with Atz/Bev alone was 15.0%. The median time to CR was 3.4 months, and the median duration of CR was 15.6 months. A significant factor associated with achieving CR with Atz/Bev alone was an AFP ratio of 0.34 or less at 3 weeks. Adding transarterial chemoembolization (TACE) in the six patients who achieved a partial response increased the overall CR rate to 20%. Among the 24 patients who achieved CR, the median progression-free survival was 19.3 months, the median overall survival was not reached, and 14 patients (58.3%) were able to discontinue Atz/Bev and achieve a drug-free status. Twelve of these patients developed progressive disease (PD), but eleven successfully received post-PD treatments and responded well. Conclusions: Achieving CR by mRECIST using Atz/Bev alone or with additional TACE can be expected to offer an extremely favorable prognosis. Full article

We conducted a comprehensive literature review analyzing protein, circulating nucleic acid, metabolite, and quantitative proteomics-based biomarkers, evaluating the advantages and limitations of each approach. While established markers like alpha-fetoprotein (AFP), des-gamma-carboxy prothrombin, and AFP-L3 remain relevant, promising candidates include circulating tumor DNA, microRNAs, long noncoding RNAs, extracellular vesicle, and metabolomic biomarkers. Multi-biomarker panels like the GALAD score, Oncoguard, and Helio liver test show promise for improved diagnostic accuracy. Non-invasive approaches like urine and gut microbiome analysis are also emerging possibilities. Integrating these novel biomarkers with current screening protocols holds significant potential for earlier HCC detection and improved patient outcomes. Future research should explore multi-biomarker panels, omics technologies, and artificial intelligence to further enhance early HCC diagnosis and management. Full article

Forest fires are increasingly destructive, contributing to significant ecological damage, carbon emissions, and economic losses. Monitoring these fires promptly and accurately, particularly by delineating fire perimeters, is critical for mitigating their impact. Satellite-based remote sensing, especially using active fire products from VIIRS and MODIS, has proven indispensable for real-time forest fire monitoring. Despite advancements, challenges remain in accurately clustering and delineating fire perimeters in a timely manner, as many existing methods rely on manual processing, resulting in delays. Active fire perimeter (AFP) and Timely Active Fire Progression (TAFP) models were developed which aim to be an automated approach for clustering active fire data points and delineating perimeters. The results demonstrated that the combined dataset achieved the highest matching rate of 85.13% for fire perimeters across all size classes, with a 95.95% clustering accuracy for fires ≥100 ha. However, the accuracy decreased for smaller fires. Overall, 1500 m radii with alpha values of 0.1 were found to be the most effective for fire perimeter delineation, particularly when applied at larger radii. The proposed models can play a critical role in improving operational responses by fire management agencies, helping to mitigate the destructive impact of forest fires more effectively. Full article

Primary liver cancer is among the most common cancers globally. It is the sixth-most common malignancy encountered and the third-most common cause of cancer-related death. Hepatocellular carcinoma (HCC) is the most common primary liver malignancy, accounting for about 90% of primary liver cancers. The majority of HCCs occur in patients with underlying cirrhosis, which results from chronic liver diseases such as fatty liver, hepatitis B and hepatitis C infections, and chronic alcohol use, which are the leading causes. The obesity pandemic has led to an increased prevalence of nonalcoholic fatty liver disease (NAFLD), which leads to nonalcoholic steatohepatitis and could progress to cirrhosis. As HCC is among the most common cancers and occurs in the setting of chronic liver disease in most patients, screening the population at risk could help in early diagnosis and management, leading to improved survival. Screening for HCC is performed using biochemical marker testing such as α-fetoprotein (AFP) and cross-sectional imaging. It is critical to emphasize that HCC could potentially occur in patients without cirrhosis (non-cirrhotic HCC), which can account for almost 20% of all HCCs. The lack of cirrhosis can cause a delay in surveillance, which could potentially lead to diagnosis at a later stage, worsening the prognosis for such patients. In this article, we discuss the diagnosis of cirrhosis in at-risk populations with details on the different modalities available for screening HCC in patients with cirrhosis, emphasizing the role of abdominal ultrasounds, the primary imaging modality in HCC screening. In the present study, the feasibility to achieve localized induction heating and debonding of multi-material composite structures is assessed in testing coupons prepared by Automated Fiber Placement (AFP) and extrusion-based additive manufacturing (AM) technologies. Nano-compounds of Polyether-ketone-ketone (PEKK) with iron oxide nanoparticles acting as electromagnetic susceptors have been processed in a parallel co-rotating twin-screw extruder to produce filament feedstock for extrusion-based AM. The integration of nanocomposite interlayers as discrete debonding zones (DZ) by AFP-AM manufacturing has been investigated for two types of sandwich-structured laminate composites, i.e., laminate-DZ-laminate panels (Type I) and laminate-DZ-AM gyroid structures (Type II). Specimens were exposed to an alternating magnetic field generated by a radio frequency generator and a flat spiral copper induction coil, and induction heating parameters (frequency, power, heating time, sample standoff distance from coil) have been investigated in correlation with real-time thermal imaging to define the debonding process window without compromising laminate quality. For the optimized process parameters, i.e., 2–3 kW generator power and 20–25 mm standoff distance, corresponding to magnetic field intensities in the range of 3–5 kA m−1, specimens were effectively heated above PEKK melting temperature, exhibiting high heating rates within the range of 5.3–9.4 °C/s (Type I) and 8.0–17.5 °C/s (Type II). The results demonstrated that localized induction heating successfully facilitated debonding, leading to full unzipping of the debonding zones in both laminate structures. Further insight on PEKK nanocomposites debonding performance was provided by thermal, morphological characterization and non-destructive inspection via X-ray micro-computed tomography at different processing stages. The developed framework aims to contribute to the development of rapid, on-demand joining, repair and disassembly technologies for thermoplastic composites, towards more efficient maintenance, repair and overhaul operations in the aviation sector and beyond. Composite materials, particularly carbon fiber-reinforced polymers (CFRPs), have become a cornerstone in industries requiring high-performance materials due to their exceptional mechanical properties, such as high strength-to-weight ratios, and their inherent lightweight nature. These attributes make CFRPs highly desirable in aerospace, automotive, and other advanced engineering applications. However, the compressive behavior of CFRP structures remains a challenge, primarily due to the material sensitivity to structural instability, leading to matrix cracking and premature failure under compressive loads. Isogrid structures, characterized by their unique geometric patterns, have shown promise in enhancing the compressive behavior of CFRP panels by providing additional support that mitigates these issues. Traditionally, these structures are manufactured using automated techniques like automated fiber placement (AFP) and automated tape laying (ATL), which, despite their efficacy, are often cost-prohibitive for small-scale or custom applications. Recent advancements in 3D-printing technology, particularly those involving continuous fiber reinforcement, present a cost-effective and flexible alternative for producing complex CFRP structures. This study investigates the compressive behavior of 3D-printed isogrid structures, fabricated using continuous carbon fiber reinforcement via an Anisoprint Composer A3 printer equipped with towpreg coextrusion technology. A total of eight isogrid panels with varying infill percentages were produced and subjected to buckling tests to assess their performance. The experimental results indicate a direct correlation between infill density and buckling resistance, with higher infill densities leading to increased buckling loads. Additionally, the failure modes were observed to shift from local to global buckling as the infill density increased, suggesting a more uniform distribution of compressive stresses. Post-test analyses using optical microscopy and scanning electron microscopy (SEM) revealed the presence of voids within the 3D-printed structures, which were found to negatively impact the mechanical performance of the isogrid panels. The findings of this study demonstrate that 3D-printed isogrid CFRP structures can achieve significant buckling resistance, making them a viable option for high-performance applications. However, the presence of voids remains a critical issue, highlighting the need for process optimizations in 3D-printing techniques to enhance the overall performance and reliability of these structures.
