Zinc oxide (ZnO) nanoparticles are widely recognized for their distinctive properties and versatile applications across diverse technological domains. However, traditional methods of synthesizing ZnO nanoparticles are characterized by environmental incompatibility, high costs, and the necessity for precise process control to attain the intended particle dimensions and morphology. The present study utilized a chives extract as a sustainable and eco-friendly fuel in the solution combustion synthesized (SCS) process to produce ZnO nanoparticles. The investigation encompassed an analysis of the impact of the fuel-to-oxidizer (F/O) ratio on the synthesized ZnO nanoparticles’ size, morphology, and crystallinity. X-ray diffraction (XRD) results showed that the particle’s crystallite size increased significantly from 12 nm to 42 nm after decreasing the F/O ratio. Furthermore, electron microscopic imagery and FTIR spectroscopy outcomes indicated that modifications in the F/O ratio significantly influenced the SCS process parameters, forming particles with diverse morphologies, including spherical, pyramid-like, hexagonal, and hexagonal plate-like shapes. This research presents a straightforward, cost-efficient, and environmentally sustainable approach for producing ZnO nanoparticles with diverse morphologies, presenting a broad potential for various applications. Immunonutrition, which focuses on specific nutrients in breast milk and post-weaning diets, plays a crucial role in supporting infants’ immune system development. This study explored the impact of maternal supplementation with Bifidobacterium breve M-16V and a combination of short-chain galacto-oligosaccharide (scGOS) and long-chain fructo-oligosaccharide (lcFOS) from pregnancy through lactation, extending into the early childhood of the offspring. The synbiotic supplementation’s effects were examined at both mucosal and systemic levels. While the supplementation did not influence their overall growth, water intake, or food consumption, a trophic effect was observed in the small intestine, enhancing its weight, length, width, and microscopic structures. A gene expression analysis indicated a reduction in FcRn and Blimp1 and an increase in Zo1 and Tlr9, suggesting enhanced maturation and barrier function. Intestinal immunoglobulin (Ig) A levels remained unaffected, while cecal IgA levels decreased. The synbiotic supplementation led to an increased abundance of total bacteria and Ig-coated bacteria in the cecum. The abundance of Bifidobacterium increased in both the intestine and cecum. Short-chain fatty acid production decreased in the intestine but increased in the cecum due to the synbiotic supplementation. Systemically, the Ig profiles remained unaffected. In conclusion, maternal synbiotic supplementation during gestation, lactation, and early life is established as a new strategy to improve the maturation and functionality of the gastrointestinal barrier. Additionally, it participates in the microbiota colonization of the gut, leading to a healthier composition. Full article

Immune system development during gestation and suckling is significantly modulated by maternal environmental and dietary factors. Breastfeeding is widely recognized as the optimal source of nutrition for infant growth and immune maturation, and its composition can be modulated by the maternal diet. In the present work, we investigated whether oral supplementation with Bifidobacterium breve M-16V and short-chain galacto-oligosaccharide (scGOS) and long-chain fructo-oligosaccharide (lcFOS) to rat dams during gestation and lactation has an impact on the immune system and microbiota composition of the offspring at day 21 of life. On that day, blood, adipose tissue, small intestine (SI), mesenteric lymph nodes (MLN), salivary gland (SG), cecum, and spleen were collected. Synbiotic supplementation did not affect the overall body or organ growth of the pups. The gene expression of Tlr9, Muc2, IgA, and Blimp1 were upregulated in the SI, and the increase in IgA gene expression was further confirmed at the protein level in the gut wash. Synbiotic supplementation also positively impacted the microbiota composition in both the small and large intestines, resulting in higher proportions of Bifidobacterium genus, among others. In addition, there was an increase in butanoic, isobutanoic, and acetic acid concentrations in the cecum but a reduction in the small intestine. At the systemic level, synbiotic supplementation resulted in higher levels of immunoglobulin IgG2c in plasma, SG, and MLN, but it did not modify the main lymphocyte subsets in the spleen and MLN. Overall, synbiotic maternal supplementation is able to positively influence the immune system development and microbiota of the suckling offspring, particularly at the gastrointestinal level. The Weilasituo Li–polymetallic deposit, located on the western slope of the southern Great Xing’an Range in the eastern Central Asian Orogenic Belt, is hosted by quartz porphyry with crypto-explosive breccia-type Li mineralisation atop and vein-type Sn-Mo-W-Zn polymetallic mineralisation throughout the breccia pipe. This study introduces new data on multistage quartz and mica in situ trace elements; the study was conducted using laser ablation inductively coupled plasma mass spectrometry and 40Ar/39Ar dating of zinnwaldite to delineate the metallogenic age and genesis of Li mineralisation. Zinnwaldite yields a plateau age of 132.45 ± 1.3 Ma (MSWD = 0.77), representing Early Cretaceous Li mineralisation. Throughout the magmatic–hydrothermal process, quartz trace elements showed Ge enrichment. Li, Al, and Ti contents decreased, with Al/Ti and Ge/Ti ratios increasing, indicating increased magmatic differentiation, slight acidification, and cooling. Mica’s rising Li, Rb, Cs, Mg, and Ti contents and Nb/Ta ratio, alongside its falling K/Rb ratio, indicate the magma’s ongoing crystallisation differentiation. Fractional crystallisation primarily enriched Li, Rb, and Cs in the late melt. Mica’s high Sc, V, and W contents indicate a high fO2 setting, with a slightly lower fO2 during zinnwaldite formation. Greisenisation observed Zn, Mg, and Fe influx from the host rock, broadening zinnwaldite distribution and forming minor Zn vein orebodies later. Late-stage fluorite precipitation highlights a rise in F levels, with fluid Sn and W levels tied to magma evolution and F content. In summary, the Weilasituo Li–polymetallic deposit was formed in an Early Cretaceous extensional environment and is closely related to a nearby highly differentiated Li-F granite. During magma differentiation, rare metal elements such as Li and Rb were enriched in residual melts. The decrease in temperature and the acidic environment led to the precipitation of Li-, Rb-, and W-bearing minerals, and the increased F content in the late stage led to Sn enrichment and mineralisation. Fluid metasomatism causes Zn, Mg, and Fe in the surrounding rock to enter the fluid, and Zn is enriched and mineralised in the later period. Full article

Pediatric T-cell Acute Lymphoblastic Leukemia (T-ALL) relapses are still associated with a dismal outcome, justifying the search for new therapeutic targets and relapse biomarkers. Using single-cell RNA sequencing (scRNAseq) data from three paired samples of pediatric T-ALL at diagnosis and relapse, we first conducted a high-dimensional weighted gene co-expression network analysis (hdWGCNA). This analysis highlighted several gene co-expression networks (GCNs) and identified relapse-associated hub genes, which are considered potential driver genes. Shared relapse-expressed genes were found to be related to antigen presentation (HLA, B2M), cytoskeleton remodeling (TUBB, TUBA1B), translation (ribosomal proteins, EIF1, EEF1B2), immune responses (MIF, EMP3), stress responses (UBC, HSP90AB1/AA1), metabolism (FTH1, NME1/2, ARCL4C), and transcriptional remodeling (NF-κB family genes, FOS-JUN, KLF2, or KLF6). We then utilized sparse partial least squares discriminant analysis to select from a pool of 481 unique leukemic hub genes, which are the genes most discriminant between diagnosis and relapse states (comprising 44, 35, and 31 genes, respectively, for each patient). Applying a Cox regression method to these patient-specific genes, along with transcriptomic and clinical data from the TARGET-ALL AALL0434 cohort, we generated three model gene signatures that efficiently identified relapsed patients within the cohort. Overall, our approach identified new potential relapse-associated genes and proposed three model gene signatures associated with lower survival rates for high-score patients. Full article

This paper presents a study about the integration of Piezoelectric Energy Harvesting Systems (PE-EHSs) into building envelopes for powering Fiber Bragg Grating (FBG) sensors, enabling efficient and low-consumption monitoring with the objective of leveraging structural health monitoring (SHM). The research includes preliminary tests conducted in a real environment to validate the PE-EHS when fully integrated into a ventilated façade, capturing mechanical vibrations generated mainly by wind loads. Based on these activities, the final configuration of PE-EHSs is defined to provide a complete system for façade monitoring. This integrated system includes the piezoelectric generator (PEG), supercapacitor (SC), Power Conditioner Circuit (PCC), Fiber Optic Sensing (FOS) interrogator, and the IoT gateway transmitting measurement data within an Internet of Things (IoT) monitoring platform. This configuration is tailored to address the challenges related to the structural integrity of building envelopes. Results demonstrate a potential for a stand-alone solution in the façade sector but raise issues for certain limitations, requiring further investigation. In particular, the study emphasizes constraints related to the energy production of PE-EHSs for façade integration. It highlights the necessity to carefully consider these limitations within the broader context of their applicability, providing insights for the informed deployment of piezoelectric energy harvesting technology in building envelope monitoring. This multi-center prospective randomized controlled trial was a tolerance and safety study investigating the thickener locust bean gum (LBG) in infants with regurgitation, to support the re-evaluation of the safety of LBG in infant formula. The primary objective was to demonstrate that after an 8-week intervention, stool consistency was not inferior (i.e., was not looser or more watery) in infants fed an anti-regurgitation (AR) formula containing LBG vs. the stool consistency of infants fed with an unthickened control formula. A total of 103 full-term infants with regurgitation were randomized to the test or control formula. The test formula contained LBG (0.4 g/100 mL), short-chain galacto-oligosaccharides, and long-chain fructo-oligosaccharides (scGOS/lcFOS; 9:1; 0.4 g/100 mL) and postbiotics and the control formula contained scGOS/lcFOS (0.8 g/100 mL), the same amount of postbiotics, and did not contain LBG. The average stool consistency score at the 8th intervention week was the primary outcome parameter. Secondary outcome parameters were stool consistency at other timepoints, stool frequency, Infant Gastrointestinal Symptom Questionnaire (IGSQ) score, growth, (serious) adverse events ([S]AEs), regurgitation severity, and infant well-being. Overall, the infants were 36.9 ± 12.9 [mean ± SD] days old, 62.7% girls in the test, and 50.0% girls in the control group. The primary analysis showed that the test group did not have looser or more watery stools than the control group. IGSQ sum scores decreased comparably in both groups. The frequency of regurgitation was significantly lower in the test group compared to the control group (mixed model repeated measurement, p ≤ 0.028) and parent-reported well-being scores were favorable. Adequate growth was observed in both groups. Both products were well-tolerated and safe and the AR formula with LBG was efficacious in reducing regurgitation frequency. This study provides further evidence for the dietary management of regurgitation by LBG-containing formulae in infants who are not exclusively breastfed, and the reassurance it can bring to parents. Nucleotides, short-chain fructooligosaccharides (scFOS), xylooligosaccharides (XOS), β-carotene and vitamin E are reported to enhance immune function; however, the evidence of this in cats is limited. The aim of this study was to determine the immunomodulatory effects of these ingredients in kittens. Forty domestic short hair kittens were designated in litters to control or test diet for 28 weeks. Test diet was fortified with 0.33 g nucleotides, 0.45 g scFOS, 0.3 g XOS, 0.7 mg β-carotene and 66.5 mg vitamin E per 100 g diet. Kittens were vaccinated against feline parvovirus (FPV) and herpesvirus (FHV) at 10, 14 and 18 weeks. Kittens remained healthy, with no measured evidence of adverse health. Serum FPV and FHV antibody titres were significantly (p < 0.05) higher in the test diet group at week 23 and 27, respectively. A significantly (p < 0.05) higher proportion of test diet group kittens demonstrated an adequate response (four-fold titre increase) to FHV vaccination and a significantly (p < 0.05) higher proportion reached a protective antibody titre for FHV. Serum IgM was significantly (p < 0.05) higher in the test diet group. The test diet group demonstrated a stronger humoral immune response to vaccination, suggesting the diet supports immune defence, enabling a greater response to immune challenges. Full article

The impact of prebiotics on human health is associated with their capacity to modulate microbiota, improving beneficial microbiota–host interactions. Herein, the prebiotic potential of microbial-fructo-oligosaccharides (microbial-FOSs) produced by a co-culture of Aspergillus ibericus plus Saccharomyces cerevisiae was evaluated on seven- and nine-strain bacterial consortia (7SC and 9SC, respectively), designed to represent the human gut microbiota. The 7SC was composed of Bacteroides dorei, Bacteroides vulgatus, Bifidobacterium adolescentis, Bifidobacterium longum, Escherichia coli, Lactobacillus acidophilus, and Lactobacillus rhamnosus. The 9SC also comprised the aforementioned bacteria, with the addition of Bacteroides thetaiotaomicron and Roseburia faecis. The effect of microbial-FOSs on the metabolic activity of intestinal Caco-2/HT29-MTX-E12 co-culture was also assessed. The results showed that microbial-FOS selectively promoted the growth of probiotic bacteria and completely suppressed the growth of E. coli. The microbial-FOSs promoted the highest production rates of lactate and total short-chain fatty acids (SCFA) as compared to the commercial prebiotic Frutalose® OFP. Butyrate was only produced in the 9SC consortium, which included the R. faecis—a butyrate-producing bacteria. The inclusion of this bacteria plus another Bacteroides in the 9SC promoted a greater metabolic activity in the Caco-2/HT29-MTX-E12 co-culture. The microbial-FOSs showed potential as promising prebiotics as they selectively promote the growth of probiotic bacteria, producing high concentrations of SCFA, and stimulating the metabolic activity of gut cells. The Sukinda ultramafic complex in India comprises precisely two areas: Kaliapani (KLPN) and Katpal (KTPL). These areas consist of a sequence of lithotypes, including orthopyroxenite, dunite, serpentinite, and chromitite, displaying a rhythmic layering of rocks. These rocks exhibit a cumulate texture and stand out due to their elevated Mg# (78.43–93.20), Cr (905.40–58,799 ppm), Ni (193.81–2790 ppm), Al2O3/TiO2 (27.01–74.06), and Zr/Hf (39.81–55.24) ratios, while possessing lower TiO2 contents (0.01–0.12 wt%). These ultramafics, characterized by low Ti/V (0.83–19.23) and Ti/Sc (7.14–83.72) ratios, negative anomalies of Zr, Hf, Nb, and Ti in a primitive mantle-normalized spider diagram, indicate that the ultramafics originate from a depleted mantle source. Furthermore, the presence of enriched LREE compared to HREE, a negative Eu anomaly, and enrichment of Th, U, and negative Nb anomalies suggest a subduction setting. The whole-rock geochemical data reveal high levels of MgO, Cr, and Ni, as well as low TiO2 and CaO/Al2O3 ratios and high Al2O3/TiO2 ratios. Moreover, the mineral chemistry data of the ultramafic rocks show high-Mg olivine (Fo 90.9−94.1) in dunite, high-Mg orthopyroxene (En 90.4–90.7) in orthopyroxenite, and high Cr# (0.68–0.82) and low Mg# (0.40–0.54) in chromite, alongside significant Al2O3 (9.93–12.86 wt%) and TiO2 (0.20–0.44 wt%) contents in the melt. Such geochemical characteristics strongly suggest that the Sukinda ultramafic originates from the fractional crystallization of a boninitic parental magma, which is derived from the second-stage melting in a depleted metasomatized mantle source within a supra-subduction zone tectonic setting. Short-chain fructooligosaccharides (sc-FOS) are prebiotics beneficial to human health, which can be synthesized from raw material containing a high sucrose content. Sugarcane syrup (SS) without molasses removal contains sucrose as a major sugar and is a rich source of several bioactive compounds. The aim of this study is to investigate factors affecting sc-FOS synthesis from SS using commercial enzyme preparations containing fructosyltransferase activity as biocatalysts. sc-FOS content increased significantly as the sucrose concentration of SS in the reaction mixture increased up to 40% (w/v). Changes in carbohydrate compositions during the transfructosylating reaction of a pure sucrose solution and SS prepared from the two sugarcane cultivars Khon Kaen 3 and Suphanburi 50, catalyzed by Pectinex Ultra SP-L and Viscozyme L, showed similar profiles. Both enzymes showed a high ability to transfer fructosyl moieties to produce sc-FOS and a plateau of the total sc-FOS concentration was observed after 4 h of reaction time. For the pure sucrose solution and SS (Suphanburi 50), Viscozyme showed a superior ability to convert sucrose to Pectinex, with a higher sc-FOS yield (g FOS/100 g of initial sucrose), GF2 or 1-kestose yield (g GF2/g of initial sucrose) and GF3 or nystose yield (g GF3/g of initial sucrose). sc-FOS syrup (FOS SS) and the foam-mat-dried syrup powder prepared from SS and FOS SS, respectively, contained a high total phenolic content and possessed higher antioxidant activities than those prepared from pure sucrose, but contained lower calories. Salmonella enterica serovar Derby (S. Derby) ranks fifth among nontyphoidal Salmonella serovars causing human infections in the European Union. S. Derby isolates (36) collected between 2006 and 2018 in a Spanish region (Asturias) from human clinical samples (20) as well as from pig carcasses, pork- or pork and beef-derived products, or wild boar (16) were phenotypically characterized with regard to resistance, and 22 (12 derived from humans and 10 from food-related samples) were also subjected to whole genome sequence analysis. The sequenced isolates belonged to ST40, a common S. Derby sequence type, and were positive for SPI-23, a Salmonella pathogenicity island involved in adherence and invasion of the porcine jejune enterocytes. Isolates were either susceptible (30.6%), or resistant to one or more of the 19 antibiotics tested for (69.4%). Resistances to tetracycline [tet(A), tet(B) and tet(C)], streptomycin (aadA2), sulfonamides (sul1), nalidixic acid [gyrA (Asp87 to Asn)] and ampicillin (blaTEM-1-like) were detected, with frequencies ranging from 8.3% to 66.7%, and were higher in clinical than in food-borne isolates. The fosA7.3 gene was present in all sequenced isolates. The most common phenotype was that conferred by the tet(A), aadA2 and sul1 genes, located within identical or closely related variants of Salmonella Genomic Island 1 (SGI1), where mercury resistance genes were also present. Diverse IncI1-I(α) plasmids belonging to distinct STs provided antibiotic [blaTEM-1, tet(A) and/or tet(B)] and heavy metal resistance genes (copper and silver), while small pSC101-like plasmids carried tet(C). Regardless of their location, most resistance genes were associated with genetic elements involved in DNA mobility, including a class one integron, multiple insertion sequences and several intact or truncated transposons. By phylogenetic analysis, the isolates were distributed into two distinct clades, both including food-borne and clinical isolates. One of these clades included all SGI1-like positive isolates, which were found in both kinds of samples throughout the entire period of study. Although the frequency of S. Derby in Asturias was very low (0.5% and 3.1% of the total clinical and food isolates of S. enterica recovered along the period of study), it still represents a burden to human health linked to transmission across the food chain. The information generated in the present study can support further epidemiological surveillance aimed to control this zoonotic pathogen. Full article

Validation and Evaluation of GRACE-FO Estimates with In Situ Bottom Pressure Array Measurements in the South China Sea

The Gravity Recovery and Climate Experiment (GRACE), and its follow-on mission (GRACE-FO), provides a novel measurement of the variations in ocean bottom pressure (OBP) at global and basin scales, including those in marginal seas. However, these measurements have not yet been validated rigorously for the South China Sea (SCS). In this study, the accuracy in the monthly GRACE-FO mascon solutions in the SCS from the Jet Propulsion Laboratory (JPL), Center for Space Research (CSR), and Goddard Space Flight Center (GSFC) was validated with the results of the comparison with the in situ OBP records from an array of 25 pressure-recording inverted echo sounders (PIESs) that are located west of the Luzon Strait (LS). The correlation coefficient (Cor) and root mean square difference (RMSD) between the 10-month period of GSFC and PIES, spanning from July 2018 to June 2019 (with missing satellite data for August and September 2018), were 0.77 (p-value = 0.005) and 0.41 mbar (1 mbar = 100 Pa), respectively. These values suggest that the accuracy of GSFC in the SCS in this period was substantially better than that of JPL (Cor = 0.35, p-value = 0.16; RMSD = 0.74 mbar) and CSR (Cor = 0.25, p-value = 0.24; RMSD = 0.89 mbar). Moreover, the volume transport anomaly of the SCS abyssal circulation was estimated and compared based on the OBP records from GSFC and PIES observations, indicating that the GRACE-FO OBP (GSFC) can be used to monitor seasonal or longer-period variations in the SCS abyssal volume transport. Additionally, the variations in OBP from GRACE-FO were significantly overestimated on the continental shelf of the SCS, which may be attributed to signal leakage. Our findings provide reliable evidence for the application of long-term, fully covered OBP records from GRACE-FO in the SCS, and also offer a valuable reference for the application of GRACE-FO in other regions. A microwave hot pressing machine (MHPM) was used to heat the colander to produce fixed oils from each of castor, sunflower, rapeseed, and moringa seed and compared them to those obtained using an ordinary electric hot pressing machine (EHPM). The physical properties, namely the moisture content of seed (MCs), the seed content of fixed oil (Scfo), the yield of the main fixed oil (Ymfo), the yield of recovered fixed oil (Yrfo), extraction loss (EL), six Efficiency of fixed oil extraction (Efoe), specific gravity (SGfo), refractive index (RI) as well as chemical properties, namely iodine number (IN), saponification value (SV), acid value (AV), and the yield of fatty acid (Yfa) of the four oils extracted by the MHPM and EHPM were determined. Chemical constituents of the resultant oil were identified using GC/MS after saponification and methylation processes. The Ymfo and SV obtained using the MHPM were higher than those for the EHPM for all four fixed oils studied. On the other hand, each of the SGfo, RI, IN, AV, and pH of the fixed oils did not alter statistically due to changing the heating tool from electric band heaters into a microwave beam. The qualities of the four fixed oils extracted by the MHPM were very encouraging as a pivot of the industrial fixed oil projects compared to the EHPM. The prominent fatty acid of the castor fixed oil was found to be ricinoleic acid, making up 76.41% and 71.99% contents of oils extracted using the MHPM and EHPM, respectively. In addition, the oleic acid was the prominent fatty acid in each of the fixed oils of sunflower, rapeseed, and moringa species, and its yield by using the MHPM was higher than that for the EHPM. The role of microwave irradiation in facilitating fixed oil extrusion from the biopolymeric structured organelles (lipid bodies) was protruded. Since it was confirmed by the present study that using microwave irradiation is simple, facile, more eco-friendly, cost-effective, retains parent quality of oils, and allows for the warming of bigger machines and spaces, we think it will make an industrial revolution in oil extraction field. The application of biopolymers and feasible technologies to obtain sheets is crucial for the large-scale production of food packages and for reducing plastic pollution. Additionally, the inclusion of additives in sheets can affect and improve their properties. This work aimed to incorporate tucumã oil (TO) and TO microparticles produced by spray drying (SD), spray chilling (SC), and their combination (SDC) into extruded cassava starch sheets and to evaluate the effect of such addition on their physical, optical, and mechanical properties. Gum Arabic and vegetable fat were used as wall materials for SD and SC/SDC, respectively. The sheets enriched with tucumã oil (FO) and the microparticles produced by SD, SC and SDC (FSD, FSC, and FSDC, respectively) presented yellow color (hue angle around 90°) and higher opacity (11.6–25.3%) when compared to the control (6.3%). All sheets showed high thickness (1.3–1.8 mm), and the additives reduced the water solubility of the materials (from 27.11% in the control to 24.67–25.54% in enriched samples). The presence of large SDC particles, as evidenced by Scanning Electron Microscopy (SEM), caused discontinuity of the sheet structure and decreased mechanical strength of the FSDC. One may conclude that potential active packages were obtained by extrusion of cassava starch sheets added with pure and encapsulated TO. Full article
