A 24/7 AI sound-based coughing monitoring system was applied in combination with oral fluids (OFs) and bioaerosol (AS)-based screening for respiratory pathogens in a conventional pig nursery. The objective was to assess the additional value of the AI to identify disease patterns in association with molecular diagnostics to gain information on the etiology of respiratory distress in a multimicrobially infected pig population. Respiratory distress was measured 24/7 by the AI and compared to human observations. Screening for swine influenza A virus (swIAV), porcine reproductive and respiratory disease virus (PRRSV), Mycoplasma (M.) hyopneumoniae, Actinobacillus (A.) pleuropneumoniae, and porcine circovirus 2 (PCV2) was conducted using qPCR. Except for M. hyopneumoniae, all of the investigated pathogens were detected within the study period. High swIAV-RNA loads in OFs and AS were significantly associated with a decrease in respiratory health, expressed by a respiratory health score calculated by the AI The odds of detecting PRRSV or A. pleuropneumoniae were significantly higher for OFs compared to AS. qPCR examinations of OFs revealed significantly lower Ct-values for swIAV and A. pleuropneumoniae compared to AS. In addition to acting as an early warning system, AI gained respiratory health data combined with laboratory diagnostics, can indicate the etiology of respiratory distress. Full article

Objectives: The aim of this study was to determine and compare the capability of several B-mode ultrasound (US) and shear wave elastography (SWE) metrics to differentiate subjects with chronic non-specific neck pain from asymptomatic subjects. Methods: A diagnostic accuracy study recruiting a sample of patients with chronic neck pain and asymptomatic controls was conducted. Data collection included sociodemographic information (i.e., gender, age, height, weight and body mass index), clinical information (pain intensity assessed using the Visual Analogue Scale and pain-related disability using the Neck Disability Index) and B-mode ultrasound and shear wave elastography features of the cervical multifidus muscle (cross-sectional area, perimeter, mean echo intensity, fat infiltration, shear wave speed and Young’s modulus). After analyzing between-group differences for left/right sides, cases and controls, and males and females, the area under the receiver operating characteristic (ROC) curve, the optimal cut-off point, the sensitivity, the specificity, the positive likelihood ratio (LR) and negative LR for each metric were calculated. A total of 316 individuals were recruited in this study (n = 174 cases with neck pain and n = 142 asymptomatic controls). Results: No significant differences (p > 0.05) were found between cases and controls for most variables, except for fatty infiltration, which was significantly higher in chronic neck pain cases (p < 0.001). Gender differences were significant across all US and SWE metrics (all, p < 0.001 except p = 0.015 for fatty infiltrates). A slight asymmetry was observed between the left and right sides for area (p = 0.038). No significant interactions between group, gender and side (all metrics, p > 0.008) were identified. Fatty infiltration was the most effective discriminator, with a ROC value of 0.723, indicating acceptable discrimination. The optimal cut-off point for fatty infiltration was 25.77, with a moderate balance between sensitivity (59.8%) and specificity (20.5%). However, its positive likelihood ratio (LR) of 0.75 suggests limited usefulness in confirming the condition. Conclusions: Fatty infiltration was significantly higher in individuals with chronic idiopathic neck pain compared to those without symptoms, while other muscle metrics were similar between both groups. However, since fat infiltration had moderate diagnostic accuracy and the other metrics showed poor discriminatory power, US cannot be used solely to discriminate patients with idiopathic neck pain. American plum line pattern virus (APLPV) is the most infrequently reported Ilarvirus infecting stone fruit trees and is of sufficient severity to be classified as an EPPO quarantine A1 pathogen. In late spring, yellow line pattern symptoms were observed on leaves in a few flowering cherries (Prunus serrulata Lindl.) grown in a public garden in Northwest Italy. RNA extracts from twenty flowering cherries were submitted to Ilarvirus multiplex and APLPV-specific RT-PCR assays already reported or developed in this study. One flowering cherry (T22) with mixed prunus necrotic ringspot virus (PNRSV) and prune dwarf virus (PDV) infection also showed infection with APLPV. Blastn analysis of PCR products of the full coat protein (CP) and movement protein (MP) genes obtained from flowering cherry T22 showed 98.23% and 98.34% nucleotide identity with reference APLPV isolate NC_003453.1 from the USA. Then, a LAMP-specific assay was designed to facilitate the fast and low-cost identification of this virus either in the laboratory or directly in the field. The developed assay allowed not only the confirmation of APLPV (PSer22IT isolate) infection in the T22 flowering cherry but also the identification of APLPV in an asymptomatic flowering cherry tree (TL1). The LAMP assay successfully worked with crude flowering cherry extracts, obtained after manually shaking a single plant extract in the ELISA extraction buffer for 3–5 min. The developed rapid, specific and economic LAMP assay was able to detect APLPV using crude plant extracts rather that RNA preparation in less than 20 min, making it suitable for in-field detection. Moreover, the LAMP assay proved to be more sensitive in APLPV detection in flowering cherry compared to the specific one-step RT-PCR assay. The new LAMP assay will permit the estimation of APLPV geographic spread in the territory, paying particular attention to surrounding gardens and propagated flowering cherries in ornamental nurseries. Nursery material intended for establishing intensive apple orchards should be characterised by a dominant and straight leader with an appropriate number of shoots that develop at the right height and are regularly spaced along the leader. The use of well-branched trees can lead to fruiting in the first year after planting. However, many apple varieties have difficulty forming lateral shoots due to strong apical dominance. The aim of the study was to assess the effectiveness of treatments stimulating the branching of maiden apple trees of the ‘Gloster’ variety. The research was carried out in 2017–2019 at a private nursery farm located in eastern Poland. The studied trees were subjected to a mechanical branching stimulation treatment, which consisted of pinching off 4–5 of the youngest leaves located below the growth cone, and chemical branching stimulation treatments, which consisted of applying growth regulator mixtures in the form of an aqueous solution, i.e., BA+GA3 and BA+GA4+7. The conducted studies showed that the type of branching-stimulating treatment had a significant effect on the height and trunk diameter of the maiden trees, the number of lateral shoots, the average length of one shoot and the sum of the lengths of all sylleptic shoots. The maiden trees treated with BA+GA3 were characterised by the best quality among the analysed combinations. Maiden apple trees treated with BA+GA3 were the tallest (2017—167.7 cm; 2018—175.3 cm; 2019—164.4 cm), produced the largest number of shoots (2017—6.5 pcs; 2018—6.8 pcs; 2019—6.3 pcs) and had the largest sum of lateral shoot lengths (2017—148.0 cm; 2018—155.4 cm; 2019—140.6 cm) among the evaluated combinations. The number of treatments and the concentration of applied growth regulators had a significant effect on the structure of the crown of the maiden apple trees of the ‘Gloster’ cultivar. Full article

Perceived Infant Discomfort Linked to Lower Maternal Oral Health Quality of Life: Results from a Cross-Sectional Study

Aim: We aimed to explore whether there is an association between maternal perceived infant discomfort due to suggestive gastrointestinal alterations and oral-health-related quality of life (OHRQoL) through a survey. Materials and Methods: The present study included two main phases involving Portuguese-speaking parents with full-term infants aged 2–12 weeks old who were not previously hospitalized in a neonatal nursery. First, the original French Infant Colic Questionnaire (ColiQ) was translated, cross-culturally adapted and validated to Portuguese (ColiQ-PT). Then, a survey was distributed, and included sociodemographics, the ColiQ-PT, an oral health value scale, OHRQoL, self-perceived periodontal status, and smoking and oral health habits. Data were analyzed through inferential, correlation and multivariate logistic models in this cross-sectional study. Results: The ColiQ-PT revealed reliability and validity. From a total of 421 responses, higher infant discomfort was correlated with less maternal professional dental care prioritization (ρ = −0.096, p < 0.05). Self-perceived periodontitis correlated with all items of OHRQoL (p < 0.001), all seven OHIP-14 domains, and with the physical (p < 0.001), psychological (p = 0.006), and social (p = 0.011) super-domains. While the infant-related score was associated with baby age (p = 0.023) and physical pain (p = 0.040) related to OHRQoL, the parent score was associated with education (p = 0.005), unemployment (p = 0.035), and physical pain (p = 0.017). The total ColiQ-PT score was significantly associated with more deteriorated social disability related to maternal OHRQoL (ρ = −0.130, p < 0.05). Conclusions: Perceived infant discomfort seems to be linked to maternal deteriorated OHRQoL. This finding highlights the importance of prioritizing oral health in postpartum care. Further research is needed to explore the mechanisms underlying this association and to develop targeted interventions. Full article

Codium tomentosum holds a variety of bioactive compounds, high nutritional value and health benefits, which makes it a valuable natural resource for the food, cosmetic and pharmaceutical industries. Currently, C. tomentosum is farmed at a small-scale targeting niche markets, and further expansion of production is limited by a lack of optimised propagation and cultivation methods. This study aims to identify the conditions required to control key production parameters including gametogenesis, gamete release and suitable culture conditions for the early stages of development of C. tomentosum. Wild specimens of C. tomentosum were collected on the Aguçadoura shore, north of Portugal. Gametogenesis was successfully induced in infertile specimens cultured under a short-day photoperiod (8 h:16 h; L:D). Gamete release was optimised through a combination of hydric shock and ultrasounds, with the highest gamete yield obtained after a 2 h 30 min desiccation period, followed by re-hydration and a series of three ultrasounds. Germlings, precursors of the adult C. tomentosum, grew faster when cultured under a lower light intensity (20 μmol m−2 s−1) compared to higher intensities (40 and 60 μmol m−2 s−1) in every light spectrum; additionally, the growth of germlings exposed to the lowest light intensity was significantly higher under white, red and green light spectra compared to blue light. The results on key production parameters constitute an important contribution to the establishment of nursery protocols based on sexual reproduction for aquaculture of the species. Full article

Conservation tillage (CT) is an agricultural technology for sustainable soil use, and clearing the root–stubble remaining in the seedbed and nursery bed is a core aspect of CT. In this paper, the characteristics and the testing methods of roots stubble and their growth environments were reviewed, which could provide a reference for the setting of parameters in numerical simulations and the design of stubble treatment devices. The methods for the restoration of the environment in CT are described. Moreover, the root–stubble crushing and clearing technologies and the methods for the evaluation of device performance are summarized. Furthermore, the prospects for the physical properties of the roots stubble soil, the reduction of soil adhesion when a cutter crushes the roots, the tracking of the long-term effects of different methods on soil, intelligent devices for the crushing of roots stubble, and challenges and strategies regarding the application of new root–stubble crushing and clearing technologies are discussed. This paper provides a reference for the development of devices for the crushing of roots stubble from the perspective of CT. Full article

Background and Objectives: Familial hypercholesterolemia (FH) is a genetic disease that is massively underdiagnosed worldwide. Affected patients are at high risk of cardiovascular events at young ages. Early intervention in childhood could help prevent heart attacks and cerebral strokes in these patients. Materials and Methods: We conducted an interventional study including 10 patients that previously underwent genetic testing for familial hypercholesterolemia. These patients received lifestyle and diet recommendations that they followed for a year before being reevaluated. Results: Patients with negative genetic testing were able to achieve lower levels in their lipid panel values compared to the patients with positive genetic testing, with lifestyle changes alone. LDL-cholesterol levels decreased by 18.5% in patients without FH while patients genetically confirmed with FH failed to achieve lower LDL-cholesterol levels without medication. Conclusions: Genetic testing for FH is not always part of screening algorithms for FH. Some studies even advise against it. Our study proved the importance of genetic testing for FH when suspecting this disorder and choosing the treatment course for patients. Full article

Nitrogen (N) fertilization promotes morphofunctional attributes that enhance plant performance under stress conditions, but the amount and form supplied modify the magnitude of plant responses. We assessed several morpho-physiological and biochemical responses of Prosopis laevigata seedlings to a high supply of N, provided as either inorganic (NH4NO3) or organic (amino acids). Such N treatments were applied on four-month-old seedlings as a supplement of 90 mg N to a regular supply of 274 mg N plant−1. Nitrogen supply modified biomass allocation patterns between leaves and roots regardless of N form. Increased N input decreased photosynthetic capacity, even when plants had high internal N reserves. Organic N fertilization reduced the N use efficiency, but increased leaf and root amino acid concentrations. Proteins accumulated in stems in plants receiving inorganic N, while the organic N increased leaf proteins. High N supply promoted root starch accumulation irrespective of N form. Nitrogen supply did not directly influence plants’ regrowth capacity. Still, resprouting was correlated to initial root-to-shoot ratios and root starch, confirming the importance of roots as storage reserves of starch for recovering biomass after browsing. These findings have practical implications for designing nutritional management strategies in nurseries to improve seedling performance in afforestation efforts. Full article

Seagrass meadows consist of angiosperms that thrive fully submerged in marine environments and form distinct ecosystems. They provide essential support for many organisms, acting as nursery grounds for species of economic importance. Beyond their ecological roles, seagrasses and their associated microbiomes are rich sources of bioactive compounds with the potential to address numerous human healthcare challenges. Seagrasses produce bioactive molecules responding to physical, chemical, and biological environmental changes. These activities can treat microbe-borne diseases, skin diseases, diabetes, muscle pain, helminthic diseases, and wounds. Seagrasses also offer potential secondary metabolites that can be used for societal benefits. Despite numerous results on their presence and bioactive derivatives, only a few studies have explored the functional and therapeutic properties of secondary metabolites from seagrass. With the increasing spread of epidemics and pandemics worldwide, the demand for alternative drug sources and drug discovery has become an indispensable area of research. Seagrasses present a reliable natural source, making this an opportune moment for further exploration of their pharmacological activities with minimal side effects. This review provides a comprehensive overview of the biochemical, phytochemical, and biomedical applications of seagrasses globally over the last two decades, highlighting the prospective areas of future research for identifying biomedical applications. The construction sector is a significant contributor to energy consumption and emissions. With the steady increase in the cost of energy carriers and the costs of energy production, the cost for consumers is also increasing. Therefore, the search for solutions capable of reducing energy consumption by increasing the energy efficiency of building structures, in particular the use of prefabricated timber-frame technology, is ongoing. Recent energy supply uncertainties and high costs necessitate the pursuit of green solutions. Timber construction, especially prefabricated timber-frame technology, holds promise due to its renewability and energy efficiency. However, housing estates built using this technology often lack service infrastructure, like shops, crèches, kindergartens, and offices, affecting resident comfort. This study proposes a methodology to select the optimal utility function for a residential building in such an estate, thus enhancing living conditions. The building’s potential new functions—a shop, nursery, or office—were evaluated based on economic criteria, thermal comfort, building airtightness, energy efficiency, and vibration comfort. The analysis indicates that converting the building into a shop requires the least capital investment, making it the most economically beneficial option. Full article

Water deficiency and potential drought periods could be important ecological factors influencing cultivation areas and productivity once different crops are established. The principal supply of vegetable oil for oil crops is oil palm, and new challenges are emerging in the face of climatic changes. This study investigated the photosynthetic performance of 12 genotypes of Elaeis exposed to drought stress under controlled conditions. The assay included genotypes of Elaeis guineensis, Elaeis oleifera, and the interspecific O×G hybrid (E. oleifera × E. guineensis). The principal results showed that the E. guineensis genotype was the most efficient at achieving photosynthesis under drought stress conditions, followed by the hybrid and E. oleifera genotypes. The physiological parameters showed good prospects for vegetal breeding with different O×G hybrids, mainly because of their ability to maintain the equilibrium between CO2 assimilation and stomatal aperture. We validated 11 genes associated with drought tolerance, but no differences were detected. These results indicate that no allelic variants were represented in the RNA during sampling for the validated genotypes. In conclusion, this study helps to define genotypes that can be used as parental lines for oil palm improvement. The gas exchange data showed that drought stress tolerance could define guidelines to incorporate the available genetic resources in breeding programs across the early selection in nursery stages. Potential Changes in Distribution of Major Conifers and Their Seed Mass across Siberia by the Mid-Twenty-First Century in a Warming Climate

Research highlights: At the turn of the 21st century, there were more forest territories found disturbed by both natural processes (climate change, wildfires, insect outbreaks, permafrost thawing, etc.) and anthropogenic interferences (air pollution, clearcuts, etc.). Seed collecting, then growing seedlings in forest nurseries, and then planting seedlings over lost forest areas are the forestry measures needed to restore the forest after disturbances. Goals were to construct bioclimatic models of ranges and seed mass of major Siberian conifers (Siberian pine (Pinus sibirica Du Tour), Siberian fir (Abies sibirica Ledeb.), Siberian spruce (Picea obovata Ledeb.), Siberian larches (Larix sibirica Ledeb., L. gmelini (Rupr) Rupr, and L. cajanderi Mayr.) and Pinus sylvestris L.) and predict their potential change in a warming climate by the mid-century. Methods: Multi-year seed mass data were derived from the literature, seed station data, and were collected in the field. Climate data (January and July data and annual precipitation) were derived from published Russian reference books and websites on climate. Bioclimatic indices (growing degree-days > 5 C, negative degree-days < 0 C, and annual moisture index) were calculated from January and July temperatures and annual precipitation for both contemporary and the 2050s (2040–2060) climates using the general circulation model INM-CM5-0 and two climate change scenarios, ssp126 and ssp585, from CMIP6. Our bioclimatic range models (envelope and MaxEnt models) and regression seed mass models for major conifers were built based on these bioclimatic indices. Additionally, their ranges were limited by the permafrost border, which divided the forest area into the permafrost-free zone, where five conifers are able to grow, and the permafrost zone, where only one conifer, Dahurian larch, is able to survive. Results: Under warmed climates, the ranges of all Siberian conifers would expand 1.5-fold due to the decrease in the permafrost zone, except Dahurian larch, which would lose 5–20% of its coverage due to permafrost retreat. Conifers shifting northward would be slower than predicted only by warmed climates because permafrost would thaw slower than climates would warm. Scots pine may expand by up to 60%, covering dryer lands in the south. Future climates were found to favor seed mass increase for major Siberian conifers and for heavier seed to shift northward. Our major conifers differ by the type of seed dispersal mode: zoochoric, animal (Siberian pine) and anemochoric, and wind-dispersed (other five trees). The seed masses of the five anemochoric conifers varied within the range of 1.5–15 g of 1000 seeds, which is about 40–50-fold less than that of zoochoric Siberian pine. Site climate explained about 28–65% of the seed mass variation for the five anemochoric trees and only 11% for Siberian pine (zoochoric tree). This finding needs additional research to explain the reasons. Conclusions: Warmed climates would favor the expansion of the ranges of major Siberian conifers and their seed mass to be heavier, which would support the high-quality seed production for forest well-being and its restoration in Siberia. With the rapid development of ecological agriculture and organic products, there is an urgent need to reduce the use of fertilizers and pesticides by producing bio-nursery substrates containing multifunctional microbial communities. In this study, beneficial Pseudomonas strains were screened from the rhizosphere of muskmelon (Cucumis melo L.) pre-inoculated with Bacillus velezensis R1-3. The ability of the strain to dissolve phosphorus and produce indole-3-acetic acid (IAA), the effect of the strain on seed germination rate, and the antagonism with R1-3 were determined. Four strains of beneficial Pseudomonas strains that had no antagonistic effect against R1-3 were obtained and formed a BacillusPseudomonas community. The seedling effect of biological substrates containing the BacillusPseudomonas community was evaluated using a seedling pot experiment and a pot experiment. The results showed that the phosphorus solubilization range of all Pseudomonas strains was 86.32–459.48 mg L−1 and the IAA production range was 2.98–11.86 mg L−1. There was a significant negative correlation between the amount of phosphorus dissolved in the fermentation solution and pH. Combined with the results of the seed germination rate and antagonism test, the strains R1-3 + HY-S7, R1-3 + HY-S25, R1-3 + HY-S36, and R1-3 + HY-S70 were selected for the seedling pot experiment and the pot experiment. The results of the two-season seedling culture and two-season pot experiments showed that the bio-nursery substrates containing the bacterial community R1-3 + HY-S70 significantly promoted the growth of muskmelon seedlings, improved plant height, maximum leaf length, and fresh weight, and were significantly better than single bacterial and control treatments at increasing plant height and fresh weight. Finally, the bacterial community R1-3 + HY-S70 was established as the optimal combination for developing biological seedling substrates. Based on 16S rDNA gene sequence analysis, the strain HY-S70 was preliminarily identified as Pseudomonas moraviensis. Full article

The avocado cv. Hass requires a suitable rootstock for optimal development under water stress. This study evaluated the performance of two avocado rootstocks (ANRR88 and ANGI52) grafted onto cv. Hass under four water stress conditions, 50% and 25% deficit, and 50% and 25% excess during the nursery stage. Plant height, leaf area (LA), dry matter (DM), and Carbon (OC) content in the roots, stems, and leaves were measured. Root traits were evaluated using digital imaging, and three vegetation indices (NDVI, CIRE, and MTCI) were used to quantify stress. The results showed that genotype significantly influenced the response to water stress. ANRR88 exhibited adaptation to moderate to high water deficits. ANGI52 adapted better to both water deficit and excess, and showed greater root exploration. LA and DM reductions of up to 60% were observed in ANRR88, suggesting a higher sensitivity to extreme changes in water availability. More than 90% of the total OC accumulation was observed in the stem and roots. The NDVI and the MTCI quantified the presence and levels of stress applied, and the 720 nm band provided high precision and speed for detecting stress. These insights are crucial for selecting rootstocks that ensure optimal performance under varying water availability, enhancing productivity and sustainability. Full article
