Ochratoxin A (OTA) is a nephrotoxic mycotoxin that commonly contaminates maize, posing significant health risks to both poultry and humans. In this study, a rapid and sensitive method utilizing ultra-performance liquid chromatography coupled with fluorescence detection (UPLC-FLD) was developed for the quantification of OTA levels in maize. The method utilizes immunoaffinity column purification for improved specificity. Accuracy and precision were validated in line with European Union Reference Laboratory (EURL-MP) guidelines, meeting regulatory standards for linearity, trueness, detection and quantification limits, precision, and uncertainty, as per European Commission Regulation (EC) No. 401/2006 and its amendments. The method demonstrated an average recovery rate of 116.78% for maize, with RSDwR values (within-laboratory reproducibility) of 12.72%. Furthermore, OTA occurrence and its possible effects were investigated in several farms in South Romania, where necropsy and histopathological analyses of poultry revealed severe kidney damage, including renal tubular degeneration. We present a case of fatal disseminated neosporosis in a 3-month-old Cane Corso puppy. The puppy presented to the Purdue Veterinary Hospital emergency service with a 1-week history of progressive left hindlimb lameness. On presentation, the puppy was nonambulatory paraparetic. Signs progressed over two days to nonambulatory tetraparesis, and decreased to absent limb reflexes, cranial nerve deficits, and dull mentation. Blood work showed eosinophilia, elevated ALT, and extremely elevated creatine kinase at 36,000 IU/L (reference interval 22–491 IU/L). Two days after diagnostics were performed, the puppy experienced acute cardiac arrest, and the body was submitted for necropsy. At necropsy, skeletal muscle in all four limbs was diffusely pale tan. Tan streaks were disseminated through the diaphragm, abdominal wall, and myocardium. Histologically, Skeletal myocytes and cardiomyocytes were frequently degenerative, with abundant lymphohistiocytic inflammation and fibrosis. White matter within the brain and spinal cord was inflamed and contained frequent dilated myelin sheaths and spheroids. A few protozoal cysts were within the brain and skeletal myocytes. Antemortem antibody titers were positive for IgG against Neospora caninum (dilution titer of 1:4096, baseline 1:32), confirming protozoal cysts as Neospora caninum. Disseminated neosporosis is an uncommon but important clinical differential for ascending paresis in young dogs. Full article

The growth rate of in situ-induced hepatic lesions in an Oncopig large animal model is quantitatively assessed. Oncopigs (n = 9) received baseline triple-phase CT scans prior to lesion induction. Lesions were subsequently induced by delivering the Ad-Cre vector to four locations in the liver. Triple-phase CT scans were obtained weekly to track the growth of the lesions. Animals were sacrificed at 14, 21, or 28 days (n = 3 in each group). The overall success rate of lesion generation was ~78%. Histopathology sections consistently revealed lesions that were highly inflammatory and consisted of a large leukocyte population without clear evidence of carcinomas. Lesions presented within imaging as hypovascular, low attenuating masses with slight contrast enhancement around the margins but little to no enhancement within the lesions themselves. The observed lesions were manually segmented on the venous phase image. Segmentation volumes were fitted to a logistic growth and decay model. Several lesions observed at earlier time points in the 28-day group had fully regressed by the time of the necropsy. The overall trend of rapid growth for the first 21 days, with spontaneous regression of the lesions being observed from day 21 to 28, suggests that the optimal window for experimental studies may be from days 14 to 21. The data and mathematical models generated from this study may be used for future computational models; however, the current model presented has moderate clinical relevance because many induced tumors resolved spontaneously within a few weeks. Awareness and careful consideration of the modest relevance and limitations of the model are advisable for each specific use case. Full article

Red wolves (Canis rufus) are the most critically endangered wolf globally, with over 95% of individuals living under human care. Gastrointestinal (GI) disease is one of the leading causes of death among adult red wolves under human care, yet links between management practices and this condition remain unclear. By integrating studbook data, survey-based husbandry information, veterinary records, and necropsy reports, our study explored the relationships between management practices and post-mortem diagnoses of GI mucosal disease among 36 adult red wolves between 2004 and 2022. For final diets fed for a minimum of 4 months before death, we report that wolves fed diets limited to only kibble were more likely to exhibit a greater severity of GI mucosal disease than those fed diets that integrated whole prey or other meat items. In addition, while living at their final facilities for a minimum of 9 months before death, wolves treated with a nonsteroidal anti-inflammatory drug exhibited more severe GI issues than those who were not. Conversely, we found no effect of housing and life history traits on GI health. This study highlights the importance of evaluating management and veterinary protocols on red wolf GI health. The nasal mucosa is a crucial filtering organ to prevent attachment and invasion of pathogens. To assess nasal health in relation to lung health, transverse cross sections of the nasal turbinates of 121 pigs suffering from respiratory disease and sent for diagnostic necropsy were scored visually and by an artificial intelligence (AI) medical diagnostic application (AI DIAGNOS), resulting in a high correlation of both scores (p < 0.001). Nasal samples of the diseased pigs were examined only for Bordetella (B.) bronchiseptica (PCR and bacteriological culture) and Pasteurella (P.) multocida (bacteriological culture). All pigs showed various degrees of inflammatory lung tissue alterations, and 35.5% of the pigs had atrophy of the nasal turbinates with no relation to detection rates of B. bronchiseptica (54.5%) and P. multocida (29.0%) in the nose. All P. multocida strains from nose samples were negative for the toxA gene so non-progressive atrophic rhinitis was diagnosed. Pigs positive for B. bronchiseptica in the nose were more often positive for B. bronchiseptica in the lung (p < 0.001) and for other bacterial species in the lower respiratory tract (p = 0.005). The new diagnostic application for scoring cross sections of nasal turbinates is a valuable tool for a fast and reproducible diagnostic. Full article

Copper is an essential trace element but becomes toxic in overexposed animals. Sheep are the domestic species most prone to chronic copper poisoning, as a slight increase in the dietary concentration can lead to liver accumulation and the development of clinical signs in this species. Common sources of copper in the diet are feed additives and mineral supplements, which are commonly used in pigs and poultry. Recently, new copper supplements were registered for animal nutrition, including copper bilysinate. This study describes an episode of presumed chronic copper poisoning in Five Texel sheep, which were exposed to a compound feed containing copper bilysinate. Four weeks after the introduction of the compound feed into the diet, the first animal started to show typical clinical signs of chronic copper poisoning and died, followed by another animal a week later. Despite removing the compound feed from the diet, a third sheep died 3 weeks later. Two animals survived and fully recovered. Necropsy and histology showed characteristic gross and microscopical lesions typical of copper poisoning. The case report highlights the potential toxic effect of copper bilysinate in sheep. Infectious Agents Associated with Abortion Outbreaks in Italian Pig Farms from 2011 to 2021

The present study retrospectively analyzed the infectious agents associated with 829 abortion outbreaks occurring from 2011 to 2021 in northern Italy. Foetuses were subjected to necropsies, and organ samples were analyzed by direct PCR to screen for six swine pathogens. In 42.0% of the examined outbreaks, at least one infectious agent was found. Porcine reproductive and respiratory syndrome virus (PRRSV) (24.9%) and porcine circovirus-2 (PCV2) (11.5%) were the most frequently detected among the known abortion-inducing pathogens. Chlamydia spp. (5.6%), porcine parvovirus (PPV) (4.0%), and Leptospira spp. (2.6%) were less common. Although its role in swine reproductive disorders is still unclear, PCV3 was detected in 19.6% of the cases. Coinfections were detected in 25.0% of positive outbreaks, and the most frequent coinfection was represented by PRRSV and PCV2 (32.2%), followed by PRRSV and PCV3 (23%). PCV2 prevalence showed a slight but consistent reduction during the study period, while PCV3 increased in frequency. Our data suggest an overall reduction in abortion outbreaks during the study period. PRRSV was confirmed as the main abortion agent detected in the examined area, while PCV2 prevalence showed a decline. Conversely, PCV3 detection has been increasing, supporting its potential role as an abortion agent. Our results highlight the importance of implementing a consistent and standardized sampling procedure, as well as a thorough diagnostic protocol, to reduce the incidence of inconclusive diagnoses. Several studies with kaempferol (KP) and linearolactone (LL) have demonstrated their antiparasitic activity. However, the toxicity of these treatments is unknown. Therefore, this study aimed to evaluate the possible toxicological effects of intraperitoneal (i.p.) administration of KP or LL on the amoebic liver abscess model (ALA) in Mesocricetus auratus. An ALA was induced in male hamsters with 1.5 × 105Entamoeba histolytica (E. histolytica) trophozoites inoculated in the left hepatic lobe. The lesion evolved for 4 days, and then KP (5 mg/kg body weight/day) or LL (10 mg/kg body weight/day) was administered for 4 consecutive days. Then, magnetic resonance imaging (MRI), paraclinical analyses, and necropsy for histopathological evaluation were performed. There was similar ALA inhibition by KP (19.42%), LL (28.16%), and metronidazole, the antiamoebic control (20.87%) (p ≤ 0.05, analysis of variance [ANOVA]). There were hepatic and renal biochemical alterations in all treatment groups, mainly for KP (aspartate aminotransferase: 347.5 ± 37.5 U/L; blood urea nitrogen: 19.4 ± 1.9 g/dL; p ≤ 0.05, ANOVA). Lesions found in the organs were directly linked to the pathology. In conclusion, KP and LL decreased ALA development and exerted fewer toxicological effects compared with metronidazole. Therefore, both compounds exhibit therapeutic potential as an alternative treatment of amoebiasis caused by E. histolytica. However, additional clinical studies in different contexts are required to reaffirm this assertion. Full article

The platypus (Ornithorhynchus anatinus) is an amphibious, egg-laying mammal of high conservation value that is found only in Australia. The zoonotic bacterium Leptospira interrogans serovar Hardjo was discovered in platypuses in prior studies, but little is known about its epidemiology. Samples in the Platypus Serum Bank were tested in 2023 and the results were combined with historical records. Antibodies against L. interrogans serovar Hardjo were found in 50% of 464 serum samples from 411 platypuses collected from 14 river basins in southeastern Australia between 1981 and 2012; prevalence remained high over three decades in the Shoalhaven River population. Seroprevalence increased with age, suggesting environmental exposure. Individual platypuses had persistent titres, some for six years. Seropositive females lactated, juveniles were recruited into the population, and there were no reports of clinical leptospirosis. Three necropsied platypuses were seropositive and had mild nephritis with leptospires in the renal tubules. The high seroprevalence, persistent titres, lack of disease, mild renal lesions, and renal colonisation suggest the platypus may be a maintenance host. Sympatric cattle had L. interrogans serovar Hardjo titres, but the spatial association with seropositive platypuses was statistically weak. Other mammalian wildlife species and sheep also have L. interrogans serovar Hardjo titres; therefore, a complex ecological network must be considered. A landscape-wide study is recommended to properly assess transmission pathways and confirm who is infecting whom. Full article

Neonatal and pediatric mortality in kittens could be associated with bacterial infections, complications from inadequate management, congenital malformations, neonatal isoerythrolysis, parasitic diseases, and viral diseases. The complexity of causes, coupled with kittens’ physiological and immunological immaturity, complicates the diagnosis and treatment of disease, highlighting the necessity for preventive measures. This study aimed to identify the leading causes of death and the occurrence of congenital malformations in stray kittens. Necropsies were performed on 68 kittens, all aged under two months. Results indicated that respiratory lesions were the leading cause of death in the youngest group, while gastrointestinal problems were more prevalent in older groups. Infectious causes were predominant across all age groups. Congenital malformations were observed in 40% of the animals, with megaesophagus, cardiovascular anomalies, bone defects, and kidney defects being the most prevalent. The findings underscore the critical importance of hygiene in preventing infections and related complications. Promoting sterilization and sanitary control in stray cats is essential to reduce overpopulation and improve living conditions. Full article

Subclinical necrotic enteritis (NE) is an enteric disease that inflicts significant economic losses in the poultry industry, primarily by reducing performance in commercial flocks but without significant mortality. This study evaluated the effects of a variety of phytogenic blends on broilers’ performance and carcass composition during an induced NE challenge. In this study, 1120 day (d)-old male broilers were allocated to four treatments groups (14 replicate floor pens, 20 birds/pen): the control (CONT) group, fed a basal corn-soybean diet, and three phytogenic blend dietary additives (PHYTO1, PHYTO2, and PHYTO3) added to the basal diet at 150, 250, and 500 mg/kg feed, respectively. Subclinical NE was induced by spraying a concentrated coccidiosis vaccine onto the feed and litter 24 h post-placement. On day 8, two birds/pen were necropsied for NE lesions. On days 8, 14, 28, and 42, the average daily gain (ADG), feed intake (ADFI), and feed conversion ratio (FCR) were calculated. On day 42, two birds/pen were euthanized to assess carcass composition using dual-energy X-ray absorptiometry (DXA). Statistical analyses for all data were performed using the ANOVA procedure (JMP, Pro 16) and significance (p ≤ 0.05) between treatments was determined by the LSD test. There was no effect of treatment on NE lesions. PHYTO1, PHYTO2, and PHYTO3 significantly improved FCR from days 9 to 14, 0 to 14, and 0 to 42 and resulted in greater ADG from days 9 to 14, 29 to 42 and cumulatively on days 0 to 42. Carcass composition data revealed a numerically higher lean-to-fat ratio in the PHYTO groups compared to the CONT group. These results indicate that the dietary supplementation of phytogenic blends could alleviate the adverse effects of NE challenge on broilers’ performance and carcass composition. Full article

At necropsy, a firm rounded nodule was noted on the left ventricle wall. Histological examination revealed a cystic formation lined by stratified squamous epithelium. The cyst was filled with lamellar keratin, while hair shafts were not present. The adjacent epicardium was infiltrated by lymphoplasmacytic cells in reaction to the rupture of the cyst with the spilling of keratinaceous debris. The lesion was diagnosed as a ruptured epicardial epidermoid cyst. EC are most commonly found in the skin, both in human and animal patients, though infrequently, they can occur in any internal organ. Cardiac EC has not been reported in domestic animals, and this is the first report of EC in a wild animal species. Clinical veterinarians should consider the possibility of similar cardiac lesions in captive subjects since the long lifespan of these rodents could allow the growth of the cyst with the compression of the adjacent tissues. Oxidative stress and chronic inflammation create a perpetual cycle in the elderly, where impaired immune function amplifies susceptibility to oxidative damage, and oxidative stress further weakens the immune response. This cycle is particularly detrimental to the respiratory system of the elderly, which is an easy target for constant exogenous harmful attacks during cold/flu season or under heavy air pollution. Herbal medicines that protect respiratory function are seen as safer alternatives to conventional therapies; however, there is limited availability of scientifically validated, safe, and effective natural supplements for these conditions. In this study, we evaluated a standardized bioflavonoid composition, UP446, that contains bioactives from the roots of Scutellaria baicalensis and the heartwoods of Acacia catechu as a natural and nutritional supplement for its antioxidative and immunoregulatory effects in oxidative stress-accelerated aging and chemically induced immune suppression mouse models. Immunosenescence was induced through the repeated subcutaneous inoculation of D-galactose (D-Gal) at a dose of 500 mg/kg/day in CD-1 mice. UP446 was administered orally at doses of 100 mg/kg and 200 mg/kg starting in the fifth week of immunosenescence induction. This study lasted a total of ten weeks. All mice received a quadrivalent influenza vaccine 2 weeks before termination. Whole blood, serum, spleen homogenate, and thymus tissues were processed for analysis. Cyclophosphamide (Cy)-induced immunosuppression was triggered by three consecutive injections of cyclophosphamide at 80 mg/kg/day, followed by the oral administration of UP446 for 18 days at doses of 100 mg/kg and 200 mg/kg. Blood was collected from each animal at necropsy, and serum was isolated for IgA and IgG ELISA analysis. UP446 was found to improve immune response, as evidenced by the stimulation of innate (NK cells) and adaptive immune responses (T cells and cytotoxic T cells), an increase in antioxidant capacity (glutathione peroxidase), the preservation of vital immune organs (the thymus), and a reduction in NFκB. UP446 also increased serum levels of IgA and IgG. The findings presented in this report demonstrate the antioxidative, anti-inflammatory, and immune-regulatory activities of UP446, suggesting its potential use in respiratory conditions involving immune stress due to aging, oxidative stress, and/or pathogenic challenges. Goats are natural hosts of Mycobacterium (M.) bovis, and affected herds can be the cause of significant economic losses. Similarites in disease course and lesions of M. bovis infections in goats and M. tuberculosis in humans make goats good models for human tuberculosis. The aim of this investigation was to characterize M. bovis challenge models in goats. For this, goats were endobronchially inoculated with three doses of M. bovis or culture medium. Clinical signs, shedding, and immune responses were monitored until 146 days post inoculation (dpi). At necropsy, lesions were examined by computed tomography, histology, and bacteriological culture. Infected goats did not develop clinical signs. M. bovis was cultured from feces, but never from nasal swabs. IGRAs were positive from 28 dpi onwards, antibodies at 140 dpi, and SICCT at 146 dpi. The increase in CD25+, IFN-γ+, and IFN-γ-releasing T-cell subpopulations was time-related, but not dose-dependent. All infected goats developed paucibacillary granulomas in the lungs and regional lymph nodes. M. bovis was regularly cultured. Dose-dependent effects included the size of pulmonary lesions, caverns, intestinal lesions, and early generalization in the high-dose group. In summary, reproducible challenge models with dose-dependent differences in lesions were established, which may serve for testing vaccines for veterinary or medical use. Full article

Recently, hypertrophic cardiomyopathy (HCM) in Sphynx cats has been associated with a variant in the gene encoding Alström syndrome protein 1 (ALMS1). The primary aims of this study were to describe the prevalence of HCM in Sphynx cats in New Zealand, and to assess the association between HCM and the ALMS1 variant in this population. In this prospective study, 55 apparently healthy Sphynx cats from registered Sphynx breeders and pet owners in New Zealand were screened by a cardiologist. A total of 42 of these cats had a repeat cardiac examination after median 1.8 years (range: 1.6–2.2). The frequency of the ALMS1 variant was 70.9% (11 homozygous and 28 heterozygous). At the median age of 5.8 years (range: 2.4–13.1), the prevalence of HCM was 40% (20 out of 55 cats). Three cats with HCM died during the study with congestive heart failure. All three cats had focal but extensive myocardial ischemia or infarction at necropsy. The ALMS1 variant was not associated with the HCM diagnosis. In summary, HCM was common in the studied cohort, suggesting Sphynx cats are predisposed to this disease.
