Osteomyelitis is a significant cause of total carcass condemnation in pigs at the slaughterhouse. The decision for total condemnation of a pig carcass for osteomyelitis is often based on traditional perceptions of the risk of pyaemia, leading to controversy among Official Veterinarians (OV) in the industry. This review aims to provide a more comprehensive understanding of the etiopathogenesis of osteomyelitis in pigs, the microorganisms involved, and the risk factors. It also highlights the urgent need for a more uniform method to evaluate osteomyelitis cases, which could significantly reduce economic losses in the industry. Lesions originating from tail-biting, tail docking, castration, teeth resection, and raw management are described as risk factors for osteomyelitis. Osteomyelitis is caused by the entry of pathogens into the animal’s bloodstream through an open wound. Trueperella monocytogenes, Staphylococcus aureus, and Streptococcus spp. are the most described pathogens. At slaughter, OVs condemn carcasses with osteomyelitis due to pyaemia. Signs of acute disease are essential to identify pyaemia cases. In chronic cases, total carcass condemnation can be avoided depending on the number of lesions and vertebrae affected. A clear overall image of the problem would help authorities in various countries adopt a more homogenous approach. Full article

Trichinellosis is a major foodborne zoonotic disease responsible for 41 human cases, according to the European Union One Health Zoonoses Report. In southern Spain, a quality management system (QMS) was applied to satellite laboratories (SLs) that conduct meat inspections of Trichinella spp. ensuring excellence practices. This study aimed to determine how eventual deviations from standard procedures may influence risk levels using Canonical Discriminant Analysis (CDA). Data were collected during slaughterhouses and game handling establishments’ official audits in 18 SLs located in the provinces of Cordoba and Seville during a 6-year period. Technical requirement deviations regarding technique and trial information, such as performing tests or calculations incorrectly or not following technical procedures, significantly increased risk level differences. Imminent risk levels were detected if the above-mentioned deviations arose. Quality assurance compromising deviations were responsible for 1150 times risk level differences, suggesting finding such may be critical for risk determination. A lack of significant influence of records and documents compromising deviations (incomplete forms or missing-erroneous or illegible data) was found. These results strengthen Trichinella spp. control strategies by pinpointing crucial aspects within QMS that require improvement, particularly in addressing deviations related to technique, trial information, and quality assurance procedures to mitigate associated risks effectively. Bovine leukemia virus (BLV) is prevalent worldwide, causing serious problems in the cattle industry. The BLV proviral load (PVL) is a useful index for estimating disease progression and transmission risk. We previously developed a quantitative real-time PCR (qPCR) assay to measure the PVL using the coordination of common motif (CoCoMo) degenerate primers. Here, we constructed a novel duplex BLV-CoCoMo qPCR assay that can amplify two genes simultaneously using a FAM-labeled MGB probe for the BLV LTR gene and a VIC-labeled MGB probe for the BoLA-DRA gene. This liquid duplex assay maintained its original sensitivity and reproducibility in field samples. Furthermore, we developed a dry duplex assay composed of PCR reagents necessary for the optimized liquid duplex assay. We observed a strong positive correlation between the PVLs measured using the dry and liquid duplex assays. Validation analyses showed that the sensitivity of the dry duplex assay was slightly lower than that of the other methods for the detection of a BLV molecular clone, but it showed similar sensitivity to the singleplex assay and slightly higher sensitivity than the liquid duplex assay for the PVL quantification of 82 field samples. Thus, our liquid and dry duplex assays are useful for measuring the BLV PVL in field samples, similar to the original singleplex assay. Full article

Osteomyelitis is the leading cause of total carcass condemnation in finishing pigs in Portugal, causing significant economic losses in swine production. The present study sought to determine a possible link between osteomyelitis in pig carcasses, pre-slaughter factors, and concomitant post-mortem inspection findings. For this purpose, meat inspection data were collected from 100,489 finishing pigs slaughtered in a northern Portuguese abattoir. Information regarding total carcass condemnation, slaughter season, origin, sex, tail-biting lesions, and husbandry invasive procedures (tail docking and teeth resection) was collected. The main cause of total carcass condemnation was osteomyelitis (61.03%). A total of 36.16% of osteomyelitis cases were present in the anterior region and 52.20% in the posterior region. In the anterior region, 94.78% of osteomyelitis cases were in the mandibular bone. Pigs with clipped teeth and carcasses with pleurisies were associated with a higher occurrence of osteomyelitis (p = 0.00262 and p < 0.0001, respectively). Second- and third-grade tail-biting lesions were also linked to a higher occurrence of osteomyelitis (p = 0.00128 and p < 0.0001, respectively). Slaughter inspection and monitoring procedures should be revised to better assess welfare factors and correlate management practices with the occurrence of osteomyelitis in pig carcasses. The moisture content of immersion vacuum-cooled sausages with modified casings containing citrus fruit extracts under different storage conditions was studied using hyperspectral imaging (HSI) associated with chemometrics. Different pre-processing combinations were applied to improve the robustness of the model. The partial least squares regression model, employing the full reflectance spectrum with pre-treatment of the standard normal variate, showed calibration coefficients of determination (Rc2) of 0.6160 and a root mean square error of calibration (RMSEC) of 2.8130%. For the first time, prediction maps developed via HSI visualized the distribution of moisture content in the immersion vacuum-cooled sausages with unique modified casings in response to fluctuating storage conditions. The prediction maps showed exact parts with high water content, which will help us to monitor and prevent mold growth. The combination of HSI with multivariate analysis not only quantifies changes in moisture content but also visually represents them in response to various casing treatments under different storage conditions, illustrating the significant potential for real-time inspection and early mold detection in sausages within the processed meat industry. The detection of the storage state of frozen meat, especially meat frozen–thawed several times, has always been important for food safety inspections. Hyperspectral imaging (HSI) is widely applied to detect the freshness and quality of meat or meat products. This study investigated the feasibility of the low-cost HSI system, combined with the chemometrics method, to classify beef cuts among fresh (F), frozen–stored (F–S), frozen–thawed three times (F–T–3) and frozen–thawed five times (F–T–5). A compact, low-cost HSI system was designed and calibrated for beef sample measurement. The classification model was developed for meat analysis with a method to distinguish fat and muscle, a CARS algorithm to extract the optimal wavelength subset and three classifiers to identify each beef cut among different freezing processes. The results demonstrated that classification models based on feature variables extracted from differentiated tissue spectra achieved better performances, with ACCs of 92.75% for PLS-DA, 97.83% for SVM and 95.03% for BP-ANN. A visualization map was proposed to provide detailed information about the changes in freshness of beef cuts after freeze–thawing. Furthermore, this study demonstrated the potential of implementing a reasonably priced HSI system in the food industry. Full article

Food Safety and Management System Audits in Food Retail Chain Stores in Greece

The present study aimed to assess the performance of food safety management systems in food retail stores via audits to reveal potential areas of improvement and to find out possible corrective actions to suggest to the top management. Two cycles of on-site audits took place in 106 stores to assess the requirements and hygiene conditions. After the first cycle of audits, improvements were suggested to the top management, and a second cycle of audits took place after a reasonable time. In the checklist, we recorded the temperatures of retail refrigerators and the scores from the inspection of hygiene and HACCP documentation. In the A′ audit, the percentage of stores that had higher temperatures than the critical limits was equal to 51%, and those temperatures occurred in the refrigerators for salads, followed by the refrigerators for deli meat, yogurts and desserts. In the B′ audit, only the refrigerators for salads exhibited percentages that were statistically significant lower (p-value < 0.05), and the stores were improved after the audit. High percentages of high-scoring stores were observed in the A′ and B′ audit in the inspection of HACCP documentation, although there was not a statistically significant improvement observed (p-value > 0.05). In the hygiene inspection, statistically significant improvement with 95% confidence appeared for “Refrigerator’s products appearance”, “Storage cleanliness”, and “Grocery shelf cleanliness”. The highest number of non-conformities without statistically significant improvement was found for “Checking temperatures of the receiving products” and “Labeling of fruit store products”, with the percentages being lower than 15% in both of the audit cycles. Many employees of the stores did not check and record the temperatures of receiving products from suppliers. In addition, the storage of spoiled products beneath fresh products for selling in the same refrigerator is not a good practice. Greater efforts must be made by top management and employees to maintain and distribute food products in the best and safest possible hygiene conditions. Full article

Bovine cysticercosis is categorized as a serious parasitic zoonotic infestation. The infection is mainly caused by the tapeworm Taenia saginata, which infects cattle and humans. The larval stage, Cysticercus bovis (C. bovis), is found in the skeletal and cardiac muscles of infected cattle. Despite its potential public health concern, few studies have been conducted on cardiac cysticercosis in Upper Egypt. This study investigates the prevalence, epidemiology, and impact of cardiac cysticercosis in Upper Egypt, emphasizing how histopathological changes in cardiac muscle and physiological parameters might be associated with the infection. From December 2022 to October 2023, a total of 941 animals from Assiut province, Upper Egypt, were slaughtered and their cardiac muscles were examined for the presence of C. bovis. Cysts were classified as viable or degenerated through macroscopic inspection. The overall prevalence of C. bovis infected hearts made up 10.8% of the total examined. The highest prevalence rate was in the summer season followed by spring; winter had the lowest infections. The histopathological examination of infected tissues revealed immune cell infiltration around Cysticercus-infected areas. Additionally, Bax immunostaining demonstrated the apoptotic effect of cysticercosis. Regarding the measured physiological parameters, there were non-significant changes in plasma levels of total protein and albumin in cattle infected with cysticercosis compared with control animals. Moreover, there was a significant decrease in total antioxidant capacity (TAC) combined with a significant increase in lipid peroxide (Malondialdehyde) (MDA), troponin T, and lactate dehydrogenase (LDH) activity in infected animals. The present work documented a set of epidemiological and pathological findings, revealing that C. bovis is a potentially harmful parasite and can cause significant health problems in both cattle and humans. In the food industry, quality and safety issues are associated with consumers’ health condition. There is a growing interest in applying various noninvasive sensorial techniques to obtain quickly quality attributes. One of them, hyperspectral/multispectral imaging technique has been extensively used for inspection of various food products. In this paper, a stacking-based ensemble prediction system has been developed for the prediction of total viable counts of microorganisms in beef fillet samples, an essential cause to meat spoilage, utilizing multispectral imaging information. As the selection of important wavelengths from the multispectral imaging system is considered as an essential stage to the prediction scheme, a features fusion approach has been also explored, by combining wavelengths extracted from various feature selection techniques. Ensemble sub-components include two advanced clustering-based neuro-fuzzy network prediction models, one utilizing information from average reflectance values, while the other one from the standard deviation of the pixels’ intensity per wavelength. The performances of neurofuzzy models were compared against established regression algorithms such as multilayer perceptron, support vector machines and partial least squares. Obtained results confirmed the validity of the proposed hypothesis to utilize a combination of feature selection methods with neurofuzzy models in order to assess the microbiological quality of meat products. Sarcocystis aucheniae is a coccidian parasite that produces macroscopic sarcocysts in South American camelid (SAC) muscles and causes a disease known as SAC sarcocystosis. This parasitosis hampers the commercialization of llama and alpaca meat, a vital economic activity in the Andean regions. No control or prevention methods are available, and diagnosis is based on postmortem visual inspection of carcasses. The aim of this study was to identify S. aucheniae B-cell epitopes suitable for the development of diagnostic methods for SAC sarcocystosis. To this end, sarcocyst immunoreactive protein bands were analyzed via mass spectrometry, and proteins in each band were identified in silico by searching in the parasite transcriptome. Five highly antigenic, hydrophilic B-cell epitopes, predicted not to cross-react with antibodies against other coccidia, were selected for future development of peptide-based serological tests. In addition, conserved domains present in the identified proteins allowed us to unravel metabolic pathways and mechanisms active in the parasitic stages present in sarcocysts, including aerobic respiration, antioxidant activity, signal transduction, protein synthesis and processing, and host–pathogen interactions. This study provides novel information on the biology of S. aucheniae, as well as new protein sequences that can be used for the development of diagnostic tests and chemotherapeutic approaches for SAC sarcocystosis. Trichinellosis is a parasitic foodborne zoonotic disease transmitted by ingestion of raw or undercooked meat containing the first larval stage (L1) of the nematode. To ensure the quality and safety of food intended for human consumption, meat inspection for detection of Trichinella spp. larvae is a mandatory procedure according to EU regulations. The implementation of quality assurance practices in laboratories that are responsible for Trichinella spp. detection is essential given that the detection of this parasite is still a pivotal threat to public health, and it is included in list A of Annex I, Directive 2003/99/EC, which determines the agents to be monitored on a mandatory basis. A Quality Management System (QMS) was applied to slaughterhouses and game handling establishments conducting Trichinella spp. testing without official accreditation but under the supervision of the relevant authority. This study aims to retrospectively analyze the outcomes of implementing the QMS in slaughterhouses and game handling establishments involved in Trichinella testing in southern Spain. Canonical discriminant analyses (CDAs) were performed to design a tool enabling the classification of SLs while determining whether linear combinations of measures of quality-assurance-related traits describe within- and between-SL clustering patterns. The participation of two or more auditors improves the homogeneity of the results deriving from audits. However, when training expertise ensures that such levels of inter-/intralaboratory homogeneity are reached, auditors can perform single audits and act as potential trainers for other auditors. Additionally, technical procedure issues were the primary risk factors identified during audits, which suggests that they should be considered a critical control point within the QMS. Full article

The aim of this work was to describe the pathotypes of Escherichia coli strains isolated from one-day-old chickens, as well as the occurrence of resistance and multidrug resistance (MDR) in these strains. A total of 429 mixed swabs from 4290 one-day-old chicks were examined between August 2021 and July 2023 (24 months) during routine point-of-destination inspections at 12 poultry farms in the Czech Republic. All samples were processed via cultivation methods using meat-peptone blood agar and Mc Conkey agar under aerobic conditions at 37 ± 1 °C for 18–24 h. The identification of the strains was performed using MALDI-TOF mass spectrometry. All confirmed strains of E. coli were screened via single or multiplex PCRs for the presence of genes encoding the virulence-associated factors iroN, cvaC, iss, felA, iutA, frz and tsh. Antimicrobial susceptibility tests were performed using the minimal inhibitory concentration (MIC) method, focusing on ampicillin, cefotaxime, tetracycline, doxycycline, enrofloxacin, florfenicol, amoxicillin with clavulanic acid and trimethoprim with sulfamethoxazole. A total of 321 E. coli strains (prevalence of 74.8%) were isolated, and 300 isolates were defined as avian pathogenic strains of E. coli (APEC) via multiplex PCR. Based on the defined virulence genes, the isolates were classified into 31 pathotypes. A total of 15.9% of the tested isolates were susceptible to all the tested antimicrobials. On the other hand, 20.5% of the isolates were identified as multidrug-resistant (8.7% of isolates were resistant to three antimicrobials, 7.3% to four antimicrobials, 3.6% to five antimicrobials and 0.9% to six antimicrobials). Monitoring pathogenic strains of E. coli in different animals and in the environment makes it possible to understand their spread in animal and human populations and, at the same time, reveal the sources of virulence and resistance genes. Beta-agonists (β-agonists) in meat products in one’s diet raise concerns about the possibility of foodborne illness. It may also lead to discomfort, such as headaches and occasional irregular heartbeats, which might be linked to a heightened concern for cardiovascular issues. Taiwan’s high demand for meat and reliance on imported meat products from certain countries where β-agonists are permitted has raised concerns. Recent import border checks and monitoring of meat products in the market have revealed the concentration of non-compliance with β-agonist residue regulations, which is ten ppb. This study aims to analyze the concentration of β-agonist residues in meat products sold in Taiwan and assess the current levels of exposure and dietary risk for consumers. The study analyzed 1415 samples of domestically produced and imported livestock products from supermarkets, traditional markets, and bulk stores in New Taipei City between 2019 and 2023. The samples were analyzed using the method for detecting 21 β-agonists based on the Taiwan Food and Drug Administration’s specifications. Estimated daily intake (EDI) of β-agonists for different age groups and the target hazard quotient (THQ) were used to assess dietary exposure and risk. The results showed that all 1415 samples were compliant with regulations. Among them, 43 beef samples showed residues of ractopamine originating from the United States, with residue concentrations ranging from 1 to 10 μg/kg and an average residue concentration of 3.3 ± 1.9 μg/kg. Under average consumption, the highest EDI for the exposed population was observed in the 6–12 age group, with values of 0.1469 μg/kg/day, 0.0734 μg/kg/day, and 0.0242 μg/kg/day for the three residue concentrations (maximum detected residue, maximum allowable residue, and average detected residue, respectively). The THQs for ractopamine in imported beef samples were all less than 1, indicating no health hazards at the current intake levels of each age group and the residue concentrations in commercially available beef. Despite the findings, traders need to acknowledge regulatory variations between Taiwan and exporting countries when importing meat products. Traders should provide inspection reports to monitor β-agonist residue levels in imports or explore sourcing beef from countries with β-agonist bans. Full article

Risk classification assessment and early warning systems are indispensable tools and technologies in the realm of regulatory control. Evaluating and issuing early warnings regarding heavy metal contaminants in meat products play a pivotal role in ensuring public safety and maintaining societal stability. In this study, we focused on heavy metal pollutants such as lead, cadmium, chromium, and arsenic. We collected national inspection data for meat products from 20 provinces in 2020. Combining dietary structure data, toxicology information, and dietary exposure assessment methods, we constructed a risk assessment model for heavy metal contaminants in food. Furthermore, we employed an entropy weight-based analytic hierarchy process (AHP-EW) to classify the results of the risk assessment for heavy metal contaminants in food. This involved determining risk rating levels and thresholds. Finally, we constructed a multi-step food contaminant risk prediction model based on the Transformer framework. To validate the model’s performance, comparative assessments were conducted across 20 datasets using various models. The results clearly indicate that the Transformer model outperformed the others in 14 datasets, excelling in its ability to provide advanced warnings for heavy metal risks in meat products. This empowers relevant authorities to strengthen their regulatory oversight of meat products based on the procedures and models proposed in this study, ultimately enhancing the efficiency of food safety risk management. The microbiological contamination of wild boar meat depends on the hygiene practices that hunters apply during its preparation, from the point of collection to its refrigeration. This study assesses Portuguese hunters’ knowledge of hygiene practices when handling wild boar carcasses that can jeopardise meat safety. A general structured survey entitled “Private consumption of game meat and good hygiene practices” was distributed to Portuguese hunters. Of the 206 respondents, 95% use wild boar meat for private consumption without prior inspection or initial examination by a veterinarian. This study also revealed that the vast majority of respondents have several risky practices that can compromise the safety of wild boar meat consumed (evisceration, handling, refrigeration, and transport). It is also evident that there is a lack of knowledge related to recognising lesions compatible with zoonotic infections in the hunted animals. These inappropriate knowledge and practices can pose a risk to hunters (occupational zoonotic health) and consumers (foodborne diseases). To reduce this risk, hunters need to be trained and informed about proper game meat handling practices. Full article
