The Expression Level of SOX Family Transcription Factors’ mRNA as a Diagnostic Marker for Osteoarthritis

Background/Objectives: Osteoarthritis (OA) is the most common degenerative and chronic joint disease and is a leading cause of pain and disability in adults worldwide. The SRY-related HMG box (SOX) family transcription factors (TFs) play a crucial role during the pathogenesis of OA; however, their exact mechanisms remain unexplored. The aim of our study was to conduct a bioinformatics analysis of the common interactions of SOX-5, SOX-9, and SOX-11 with other proteins, as well as their role in OA pathogenesis. Methods:SOX5, SOX9, and SOX11 mRNA expression levels in articular cartilage with subchondral bone and synovium from knee OA patients were assessed using the qPCR method. The study group consisted of thirty-one patients (n = 31). Total RNA was isolated from the articular cartilage with subchondral bone and synovium from the affected and unaffected area of the knee joint. Results: Our results revealed a regulatory network between SOX-5, SOX-9, and SOX-11, and various proteins involved in the pathogenesis of knee OA and their collective interactions, which are involved in the regulation of cartilage extracellular matrix (ECM) organization, response to stimulus, regulation of gene expression, inflammatory response, cartilage condensation, and ossification in chondrocytes. Higher expression levels of SOX5, SOX9, and SOX11 mRNA were noted in OA-affected articular cartilage with subchondral bone compared to control tissue (p = 0.00015, p = 0.0024 and p > 0.05, respectively, Mann–Whitney U-test). All studied genes demonstrated elevated mRNA expression levels in the articular cartilage with subchondral bone from stage 4 patients than those with stage 3 (p > 0.05; Mann–Whitney U-test). Lower SOX5, SOX9, and SOX11 mRNA expression levels were found in OA-affected synovium compared to the control tissue (p = 0.0003, p > 0.05 and p = 0.0007, respectively, Mann–Whitney U-test). Decreased SOX9 mRNA expression levels in synovium were noted in patients with stage 4 disease than those with stage 3; however, SOX5 and SOX11 mRNA expression levels were higher in patients with stage 4 (p > 0.05; Mann–Whitney U-test). Conclusions: The results of our research show that the studied SOX TFs play a role in the development of OA, contributing to the formation of pathological changes not only in the articular cartilage, but also in the synovial membrane. The changes in the SOX5, SOX9, and SOX11 mRNA expression levels in the articular cartilage with subchondral bone and synovium may serve as potential molecular diagnostic biomarkers for detecting OA and could indicate the progression of this disease; however, our observations require further investigation. Effect of n-3 Polyunsaturated Fatty Acids Enriched Chicken Meat Consumption in Relation to Oxidative Stress Marker Levels in Young Healthy Individuals: A Randomized Double-Blind Study

Oxidative stress and inflammation are considered important risk contributors for various diseases. Over the last few decades, increasing attention has been focused on the role of n-3 polyunsaturated fatty acids (n-3 PUFAs) in human health and disease. We aimed to evaluate the effect of n-3 PUFA-enriched chicken meat consumption (~1500 mg of n-3 PUFAs intake per day) for three weeks on oxidative status and antioxidative capacity in young healthy individuals. This was a randomized, double-blinded, controlled trial, in which thirty-nine young healthy people were randomly allocated to eating 500 g/day of regular chicken meat (Control group) or n-3 PUFA-enriched chicken meat (n-3 PUFAs group) over 3 weeks. Subjects’ biochemical parameters, including serum lipids level, liver enzymes, serum activities of antioxidant enzymes (glutathione peroxidase (GPx), superoxide dismutase (SOD)), serum oxidative stress markers (thiobarbituric acid reactive substances (TBARS) and ferric-reducing ability (FRAP)), as well as intracellular production of reactive oxygen species (ROS) in peripheral blood mononuclear cells, were assessed before and after completing the three-week dietary protocol. N-3-enriched chicken meat consumption significantly reduced high-sensitivity C reactive protein (hsCRP) serum level and increased the level of the antioxidant defense marker, FRAP. Furthermore, GPx and SOD enzyme activities significantly increased in the n-3 PUFAs group compared to baseline, which was accompanied by significantly decreased ROS production. In healthy young individuals, the 3-week dietary intake of n-3 PUFA-enriched chicken meat significantly increased the serum total antioxidant and anti-inflammatory potential, indicating that n-3 PUFAs may be protective in resting health condition without inflammatory processes. Nitrogen source is a necessary nutrient factor in the process of mycelial growth and metabolite synthesis. To improve the yield of ergosterol, in the previous study, we used transcriptome technology to explore the difference in gene expression of Cordyceps cicadae by CO(NH2)2 (urea) in the process of synthesizing ergosterol. In the present study, CO(NH2)2 was used to examine its effects on cell membrane permeability and metabolic flow in the process of C. cicadae fermentation and ergosterol synthesis. Metabonomic results showed that CO(NH2)2 supplementation caused significant changes in five aspects: (1) CO(NH2)2 notably increased biomass growth and extracellular ergosterol, and decreased intracellular ergosterol concentration; (2) CO(NH2)2 boosted the level of inositol 1,4-bisphosphate, which implied the acceleration of cell membrane decomposition, the weakening of integrity, and the increase in permeability and change in metabolic regionalization; (3) CO(NH2)2 changed the metabolic fluxes and metabolic speed, including increasing the levels of amino acids, vitamins, hormones, and nucleotides, which exhibited an elevated biomass growth, promoting the synthesis of intracellular flavonoids, alkaloids, and terpenes, facilitating extracellular ergosterol synthesis and decreasing the degradation of ergosterol; (4) in the fermentation anaphase, CO(NH2)2 caused the inhibition of 6-phosphogluconate dehydrogenase and α-ketoglutarate dehydrogenase activities, decreased NADPH, NADH, and ATP synthesis, and finally inhibited biomass growth and ergosterol synthesis. Collectively, metabonomics was a valuable strategy to study the regulatory effects of medium composition and incubation conditions on ergosterol synthesis by C. cicadae. Full article

This study examined the impact of three pre-season fitness training protocols with different training frequencies (heavy, moderate and light) on the physical abilities and the biochemical indicators of muscle damage and oxidative stress in female basketball athletes as expressed through physiological and biochemical assessments. Also, a comparison and a search for correlations were made between the physiological and biochemical changes that were caused by each protocol. The sample consisted of 28 professional female basketball athletes, who competed in three different teams, and each training protocol was assigned to each of the three teams. The somatometric and performance markers measured were as follows: weight, fat percentage, aerobic capacity, anaerobic capacity, vertical jumping ability, speed, acceleration, explosiveness and maximum lower body part strength. The biochemical measurements consisted of exercise-induced muscle damage (creatine kinase—CK, lactate dehydrogenase—LDH) and oxidative stress markers (protein carbonyls—PCs, glutathione peroxidase—GPx). The three fitness training protocols that were used improved the fitness status of high-level female basketball athletes after 6 weeks during the pre-season period, but they were also accompanied by some significant differences between them (fat percentage, anaerobic capacity, speed and acceleration). The results were also accompanied by an increase in CK and LDH for the three teams and by an increase in GPx for Teams 1 and 3. Also, we concluded that even 4 more or 4 less fitness training units (TUs) during a 6-week basketball preparation period can be considered important to differentiate the outcomes of physiological and biochemical markers. The purpose of this study was to provide more theoretical and practical knowledge to basketball coaches and trainers, so they can optimize the training process during the preparation period and thus maximize the performance of their athletes. Nevertheless, the present study was created with certain limitations, such as the small sample size for each team, the absence of a long-term follow-up or the lack of a control group. Future studies may need to turn their focus on fitness protocols with longer durations, try to differentiate the types of training protocols and attempt to achieve a better balance between maximizing performance and mitigating the muscle damage and oxidative stress levels. Oxidative stress is considered a common underlying mechanism in many retinal degenerative diseases and is often associated with inflammation. The use of dietary supplements containing Saffron has beneficial effects in ocular diseases, though the molecular mechanisms are still unclear. In this study, we investigated how Saffron can exert protective effects against oxidative damage in retinal pigment epithelial cells (ARPE-19) and whether its combination with Elderberry and Melilotus may have additive beneficial effects. ARPE-19 cells were pretreated with Saffron alone or in a mix containing Saffron, Elderberry and Melilotus, then exposed to hydrogen peroxide (H2O2) for 3 h. Afterwards, we evaluated cell viability, oxidative stress and inflammatory status. Our results showed that H2O2 reduced cell viability and total glutathione levels, while increasing caspase-3, caspase-1 and LDH activity. Moreover, H2O2 triggered ROS production, glutathione oxidation and IL-1β secretion. Pretreatments with Saffron alone or with the mix counteract these damaging effects by improving cell viability, reducing oxidative stress and enhancing SOD2 expression. Pretreatment with the mix activated the NRF2 pathway and was more effective than Saffron alone in preventing caspase-1 activation. These findings suggest that the combination of Saffron, Elderberry and Melilotus could have therapeutic potential in the prevention and treatment of retinal degenerative diseases. Full article

(1) Liver injury caused by an overdose of acetaminophen (APAP) represents a major public health concern. Paeoniflorin (PF) has been reported to have anti-inflammatory and liver-protective effects, but the underlying mechanisms remain unclear. This study aimed to investigate the effect of PF on the crosstalk between pyroptosis and NETs in AILI. (2) APAP-treated C57BL/6J mice were used to demonstrate the protective effect of PF on liver injury. HepG2 and dHL-60 cells were cultured to study the effects of PF on hepatocyte pyroptosis and neutrophil extracellular traps (NETs) in vitro. Moreover, cell co-culture experiments were performed, and mice were treated with a NETs-depleting agent and hepatocyte pyroptosis inhibitor to investigate the improvement of AILI induced by PF through regulating the crosstalk between hepatocyte pyroptosis and NETs. (3) PF significantly alleviated AILI. Additionally, PF inhibited the expression of pyroptosis-related proteins, high-mobility group box 1 (HMGB1), and NETs-associated proteins in vitro and in vivo. The co-culture experiments demonstrated that PF not only inhibited the NETs triggered by hepatocyte pyroptosis, but also suppressed the hepatocyte pyroptosis induced by NETs. In mice with depleted neutrophils, the level of hepatocyte pyroptosis notably decreased, indicating a diminished impact of PF. Similarly, NETs formation was reduced in mice receiving a pyroptosis inhibitor compared to the APAP group. Compared with DNase I alone, the reduction effect of PF combined with DNase I on serum ALT and AST levels decreased from 46.857% and 39.927% to 44.347% and 33.419%, respectively. Compared with DSF alone, PF combined with DSF reduced the ALT and AST levels from 46.857% and 39.927% to 45.347% and 36.419%, respectively. (4) PF demonstrated therapeutic effects on AILI. Its mechanism involves the regulation of the crosstalk between hepatocyte pyroptosis and NETs. This research substantiates the pharmacological promise of PF as a therapeutic intervention for acute AILI. Full article

Background: Ultrasound has become more popular and useful over the last few years in improving healthcare. While handheld devices offer portability and convenience, their diagnostic accuracy and clinical utility require further scrutiny. This study attempted to evaluate the non-inferiority of handheld portable ultrasound devices compared to standard ultrasound devices for common lung pathologies. Methods: Videos of various common lung pathologies from 20 patients were recorded by a single operator using both portable handheld and standard ultrasound devices in a single setting. These videos were then assessed via online questionnaires by clinicians of various levels of experience from respiratory and non-respiratory departments. A Likert scale was used, ranging from strongly disagree to strongly agree (ranging from 1 to 5) in terms of overall image quality, clear anatomical visualization, similar clinical interpretations/decisions, and the perception of non-inferiority. Median values with interquartile ranges were reported; a rating of 3 or above was defined as indicating non-inferiority. Results: Thirty participants completed the questionnaires, of which the majority were at trainee level (n = 20, 73%) and from a respiratory department (n = 20, 67%). The participants had mixed levels of experience in terms of the years and frequency of use of the ultrasound. Overall median ratings were 4.0 for overall image quality, clear anatomical visualization, and similar clinical interpretations/decisions, with slight variations in interquartile ranges. No significant differences were observed between subgroups. The portable ultrasound device was rated similarly for the overall perception of non-inferiority, but clinicians from respiratory departments and clinicians with less experience showed statistically significant variability in their assessments. Conclusions: The portable handheld device demonstrated potential as a reliable alternative to standard models in standard clinical settings without compromising clinical decision. Further evaluation is needed that includes a direct comparison of various types of handheld ultrasound devices, across different operators’ levels of experience, to further solidify their suitability in patient care. As global income levels and population increase, meat consumption and production are rising, raising concerns about climate change, animal welfare, and environmental sustainability. Offal offers a sustainable, nutrient-rich, and economical alternative to muscle meat, providing proteins, vitamins, and minerals. This study was triggered by the scarcity of information on the microbiological quality of edible bovine offal, which is often associated with poor hygienic conditions during handling and processing at slaughterhouse. This study assessed the superficial microbiota present on cattle liver and tongue in a vertical slaughterhouse across various stages of the procurement process and on days with different slaughter volumes, with three sampling points: immediately after post-mortem inspection (Point 1); after the arrival of the viscera in the preparation room (Point 2); and in the refrigeration chamber at 3 °C (Point 3). Samples were analysed according to ISO methods for enumeration of mesophilic aerobic bacteria, Enterobacteriaceae, generic Escherichia coli, presumptive Escherichia coli O157, and Staphylococcus aureus and detection of presumptive Escherichia coli O157, Staphylococcus aureus, and Salmonella spp. Microbiological counts were within the acceptability values stipulated in the amendment of Regulation EC 1441/2007 to Regulation EC 2073/2005 for cattle carcasses. In general, both viscera showed higher microbial counts on days with higher slaughter volumes, with the exception of Enterobacteriaceae and Staphylococcus counts on the tongue. There was also an increase in microorganism levels across the offal processing stages. Out of 144 samples, 20.83% were contaminated with presumptive Escherichia coli O157, 3.47% Salmonella spp., and 25% S. aureus. Therefore, although 20.83% of the samples tested were presumptive for E. coli O157, it is important to note that a considerable number of these presumptive positives may in fact have been negative after confirmation by molecular methods. However, the presence of presumptive E. coli O157 at high levels is one reason that the prevention of offal contamination requires more stringent measures. Preventing offal contamination requires stricter measures during slaughter procedures to ensure safety, reduce food losses, and enhance sustainability in meat production. Full article

Aim: This cross-sectional study examines sociodemographic, nutritional, and lifestyle factors affecting Body Mass Index (BMI) in the Spanish population, with a particular emphasis on obesity. Methods: A sample of 22,181 Spanish residents aged 18 years and older was recruited through digital and physical channels from August 2020 to November 2021. Data were collected using the validated NutSo-HH questionnaire, which includes sections on sociodemographic information, health perceptions, eating habits, and lifestyle factors. Results: Among respondents, 661 (3%) were underweight (BMI < 18.5 kg/m2), 14,562 (65.7%) were normal weight (18.5 kg/m2 ≤ BMI ≤ 25 kg/m2), 4825 respondents (21.8%) were overweight (25 kg/m2 < BMI ≤ 30 kg/m2), and 2133 (9.6%) were obese (BMI > 30 kg/m2), with significant differences across these groups in relation to diet and lifestyle behaviors. Principal Component Analysis (PCA) was employed to identify the primary variables influencing obesity, revealing that poor dietary habits (frequent consumption of fast food, fried foods, and ultra-processed items) were negatively correlated with healthy behaviors such as regular fish consumption and physical activity. The PCA plot indicated notable distinctions based on educational attainment and age, with individuals with lower educational levels displaying poorer nutritional patterns and younger participants exhibiting higher fast food consumption and poorer sleep quality. Statistical analyses confirmed that sociodemographic factors, including age, education, and income level, significantly influenced BMI. Some differences were also found according to the place of residence. Conclusions: These findings underscore the need for targeted interventions that address both sociodemographic and lifestyle factors to mitigate obesity risk in Spain. Full article

Background: COVID-19 can cause acute pulmonary hypertension (PH), worsening outcomes in critically ill elderly patients. Point-of-care ultrasound (POCUS), assessing right ventricular hemodynamics, predicts short-term outcomes. This study examines the long-term impact of acute PH on mortality in elderly COVID-19 patients. Methods: This retrospective long-term study analyzed data from patients over 70 years old with severe COVID-19 pneumonia admitted to a mixed 25-bed, level 3 intensive care unit (ICU). POCUS focused on systolic pulmonary artery pressure (sPAP) at admission. Mortality was evaluated 1000 days post-admission. Results: The study included 130 patients, comprising 30 long-term survivors and 100 non-survivors, with a cumulative long-term mortality rate of 77%. Non-survivors had significantly higher sPAP values (39.1 ± 12.8 vs. 30.4 ± 9.2, p = 0.04), which were associated with long-term mortality in survival analysis. Conclusion: Acute pulmonary hypertension (PH), reflected by elevated systolic pulmonary artery pressure (sPAP), is strongly associated with long-term mortality in elderly critically ill COVID-19 patients. Early assessment of sPAP via POCUS may help identify high-risk patients and guide management strategies to improve outcomes. Full article

Lung cancer is a leading cause of mortality worldwide, and early detection is crucial in improving treatment outcomes and reducing death rates. However, diagnosing medical images, such as Computed Tomography scans (CT scans), is complex and requires a high level of expertise. This study focuses on developing and evaluating the performance of Convolutional Neural Network (CNN) models, specifically the Visual Geometry Group 16 (VGG16) architecture, to classify lung cancer CT scan images into three categories: Normal, Benign, and Malignant. The dataset used consists of 1097 CT images from 110 patients, categorized according to these severity levels. The research methodology began with data collection and preparation, followed by training and testing the VGG16 model and comparing its performance with other CNN architectures, including Residual Network with 50 layers (ResNet50), Inception Version 3 (InceptionV3), and Mobile Neural Network Version 2 (MobileNetV2). The experimental results indicate that VGG16 achieved the highest classification performance, with a Test Accuracy of 98.18%, surpassing the other models. This accuracy highlights VGG16’s strong potential as a supportive diagnostic tool in medical imaging. However, a limitation of this study is the dataset size, which may reduce model accuracy when applied to new data. Future studies should consider increasing the dataset size, using Data Augmentation techniques, fine-tuning model parameters, and employing advanced models such as 3D CNN or Vision Transformers. Additionally, incorporating Gradient-weighted Class Activation Mapping (Grad-CAM) to interpret model decisions would enhance transparency and reliability. This study confirms the potential of CNNs, particularly VGG16, for classifying lung cancer CT images and provides a foundation for further development in medical applications. Full article

The aim of this study was to evaluate the effectiveness of different anesthetic protocols for semen collection from healthy tomcats using Urethral Catheterization after Pharmacological Induction (UrCaPI). Twenty sexually mature tomcats (Felis catus) were included in the study, in which high and low dosages of medetomidine or dexmedetomidine were administrated to collect semen. Ten healthy mature tomcats (Group A) received high dosages of the drugs, and, in particular, five of them were i.m. injected with 120 μg/kg of medetomidine (HMED) on day 1 and with 60 μg/kg of dexmedetomidine (HDEX) after 24 h (day 2). The remaining five tomcats of this group were i.m. injected with 60 μg/kg of dexmedetomidine on day 1 and with 120 μg/kg of medetomidine on day 2. The other ten healthy mature tomcats (Group B) received a low dosage of the same drugs; the first five tomcats were i.m. injected with 50 μg/kg of medetomidine (LMED) on day 1 and with 25 μg/kg of dexmedetomidine (LDEX) on day 2; the others were i.m. injected with 25 μg/kg of dexmedetomidine on day 1 and with 50 μg/kg of medetomidine after 24 h. In both groups, semen collection was performed as soon as the pharmacological effect of the drug was reached. All protocols permitted sperm collection, even if with different results in quality for volume, concentration, total number of spermatozoa, and movement score. Results suggest that both a high dosage of medetomidine and a high dosage of dexmedetomidine could be used for the collection of good-quality semen. Semen volume μL: HMED = 32 μL (25.75–37.5), HDEX = 23 μL (15.25–28). Concentration HMED = 670 × 106/mL (576–990.5), HDEX = 670.5 × 106/mL (536–790). Total number of spermatozoa = 23.24 × 106 (18.37–32.05). Total number of spermatozoa = 13.121 × 106 (10.116–16.83). However, the protocol with dexmedetomidine could not always guarantee an adequate sedation for urethral catheterization, so using a high dosage of medetomidine is still the better way to collect high quality semen with the UrCaPI technique. The outcome of the LMED and LDEX protocols was generally unsatisfactory regarding both level of sedation and semen collection in terms of volume (LMED = 3 μL (4–7.5); LDEX = 6 μL (4–7)), concentration (LMED = 215 × 106/mL (157–248); LDEX = 27.05 × 106/mL (0.040–110), total number of spermatozoa (LMED = 0.88 × 106 (0.581–1.38); LDEX = 0.16 × 106 (0.0001–0.80), and movement score (LMED = 3 (2–3); LDEX = 2 (0–2.75)). With the rapid development of digital networks and communication technologies, traditional word-of-mouth (WOM) has transformed into electronic word-of-mouth (eWOM), which plays a pivotal role in improving the management and service quality of ancient town tourism. This study uses Python web scraping techniques to gather eWOM data from the top ten ancient towns in southern China. Using IPA analysis, the analytic hierarchy process (AHP), Term Frequency–Inverse Document Frequency (TF-IDF), and cluster analysis, we developed a comprehensive eWOM evaluation framework. This framework was employed to perform word frequency analysis, sentiment analysis, topic modeling, and rating analysis, providing deeper insights into tourists’ perceptions. The results reveal several key findings: (1) Transportation infrastructure varies significantly across the towns. Heshun and Huangyao suffer from poor accessibility, while the remaining towns benefit from the developed transportation network of the Yangtze River Delta. (2) The volume of eWOM is strongly influenced by seasonal patterns and was notably impacted by the COVID-19 pandemic. (3) The majority of tourists express positive sentiments toward the ancient towns, with a focus on the available facilities. Their highest levels of satisfaction, however, are associated with the scenic landscapes. (4) A comprehensive eWOM analysis suggests that Wuzhen and Xidi–Hongcun are the most popular tourist destinations, while Zhujiajiao, Huangyao, Zhouzhuang, and Nanxun exhibit lower levels of both attention and visitor satisfaction. Background: This study investigated the humoral responses to SARS-CoV-2 in hemodialysis (HD) patients. The clearance of molecules in the blood during hemodialysis is influenced by factors such as filter pore size, flow rate, operating pressure, and treatment duration. Chronic kidney disease patients often show low antibody titers for pathogens like pneumococcus, influenza virus, and hepatitis B virus. Methods: In this study, the surrogate virus neutralization test (sVNT) for the wild type (WT) and Omicron variants, as well as spike-specific IgG levels, were measured at two time points (May 2022 and December 2023). Medical records and questionnaires were used to gather participant information. Results: A total of 26 HD patients were enrolled, including 3 on immunosuppressive therapies. A total of 8 patients had COVID-19 during the first sampling, and 19 during the second. The results showed that sVNT levels for WT decreased over time, though positivity remained at 100% during both sampling periods. In contrast, sVNT levels for Omicron increased significantly, with positivity rising from 46.2% to 75.0% (p < 0.05). Spike-specific IgG levels also increased, with positivity improving from 96.2% to 100%. Patients on immunosuppressive therapies had significantly lower sVNT levels for both WT and Omicron in the second period (p < 0.05), though no significant differences were observed during the first period. Conclusion: HD patients, particularly those on immunosuppressive therapies, showed reduced and declining neutralizing responses over time. A meta-analysis of HD patients seems necessary to determine whether all dialysis patients need COVID-19 booster vaccinations, similar to the hepatitis B vaccine, highlighting the need for targeted vaccination strategies. The sensory acceptability rate of pasteurized orange juices stored at different temperature levels was evaluated using a large-scale experiment with untrained consumers and an accelerated shelf-life testing method. Pasteurized orange juice samples were stored at 10, 20, 30 and 40 °C and tested for acceptance or rejection at consecutive time points using a reverse product preservation plan. From a total of 504 samples and the participation of 72 consumers, 155 failures and 109 passes were recorded. Due to the large number of samples (28) selected for testing from each consumer, a balanced incomplete block design was adopted to facilitate a very small number for evaluation (7 for each consumer) with minimal reduction in the reliability of the experiment and results. As expected, there was a gradual increasing trend of rejections at all temperature levels by time. Also, it was observed that pasteurized orange juices tended to be rejected by consumers at approximately 3 days before they would be microbiologically unsafe for consumption. Also, it was shown from the risk rate trend that higher storage temperatures are linked with very early perceptiveness of sensory defects during nearly the first 100 h of preservation. The present work constitutes essentially a very useful tool for the industry because it enables the proper adjustment of shelf life including both sensory and microbiological parameters, which results in fewer returned and deteriorated products.
