Information sources

[edit]

Much of the information from this article is culled from http://plavix.com and http://www.sanofi-synthelabous.com/products/index.html . This includes the incidences of TTP and GI bleeding.

Three of the four links to plavix.com at the end of the article are broken.

I took care of the dead links (by deleting them). Why keep the phonetic pronunciation in the first line of the article? it's not in most of the other drug articles. Ksheka 20:34, 31 October 2006 (UTC)[reply]
The single trial in the trials section is basically a copy-paste of the conclusion of the trial. Can someone do better? Important clopidogrel trials include Caprie [1] and CURE [2].Ksheka 20:43, 31 October 2006 (UTC)[reply]

grapefruit juice?

[edit]

I understand that clopidogrel is metabolized by CYP2C19... However, does grapefruit juice affect CYP2C19 as it does with CYP3A4? Does this affect the metabolism of clopidogrel? --128.227.62.57 (talk) 13:52, 19 February 2010 (UTC)[reply]

Das absolute Plavix

[edit]

Ann Intern Med this week: PMID 17371888. JFW | T@lk 16:32, 21 March 2007 (UTC)[reply]

http://jama.ama-assn.org/cgi/content/abstract/299/18/2164 - reduces absolute risk of dialysis fistula thrombosis by 7% (NNT 14) but does not lead to a larger proportion of them becoming suitable for dialysis. Hence: not likely to be taken up due to bleeding risk unless concomitant indication (e.g. coronary stents). JFW | T@lk 12:53, 14 May 2008 (UTC)[reply]

Pharmacogenetics

[edit]

From TIMI: reduced CYP2C19 function leads to reduced response and increased rates of "adverse cardiovascular events" (MI/stroke etc.) doi:10.1056/NEJMoa0809171/doi:10.1056/NEJMoa0808227. Will add this to the article myself if nobody beats me to it. Fvasconcellos (t·c) 13:41, 23 December 2008 (UTC)[reply]

PK/PD

[edit]

"Platelet inhibition can be demonstrated two hours after a single dose of oral clopidogrel, but the onset of action is slow, so that a loading-dose of 300–600mg is usually administered" needs clarification - if there is detectable action after 2h this is not particularly slow in most circumstances. Does this mean that (a) onset of clinically relevant [however defined] effect is slower than 2h and is accelerated with a higher loading dose (b) there are some circumstances (e.g. an acute coronary event) where 2h is too slow to be acceptable? --mman444 (talk) 12:17, 25 August 2010 (UTC)[reply]

Hello fellow Wikipedians,

I have just added archive links to one external link on Clopidogrel. Please take a moment to review my edit. If necessary, add {{cbignore}} after the link to keep me from modifying it. Alternatively, you can add {{nobots|deny=InternetArchiveBot}} to keep me off the page altogether. I made the following changes:

severe neutropenia

[edit]

I've removed "severe neutropenia" as an adverse effect. There's no reference for it happening frequently. The only thing like it happens rarely (I'm no doctor: Deficiency of Granulocytes a Type of White Blood Cell). The rare serious side effects should not be in the top section of the article because they're too many. See: http://www.webmd.com/drugs/2/drug-5190/clopidogrel-oral/details/list-sideeffects — Preceding unsigned comment added by 212.78.165.181 (talk) 14:08, 2 February 2016 (UTC)[reply]

Allergy

[edit]

A freind of mine reports an allergic reaction using Plavix. An internet search finds frequent occurances. Would somebody here knowledgable like to add this in the Adverse Effects section? Theosch (talk) 07:35, 12 October 2017 (UTC)[reply]

Done, thanks for bringing this up. These are probably not true allergies but hypersensitivity reactions. They are uncommon by percentage, but as clopidogrel is prescribed very frequently, events add up. --ἀνυπόδητος (talk) 09:02, 12 October 2017 (UTC)[reply]
