Hanging Drop

The document outlines the procedure for demonstrating bacterial motility using the Hanging Drop method, detailing steps for preparation and observation under a microscope. It also describes different arrangements of flagella in bacteria, including peritrichous, polar, monotrichous, lophotrichous, and amphitrichous configurations. Additionally, it defines bacterial motility as a purposeful and directional movement from one location to another.

Procedure for Hanging Drop

Preparation:

1.Take a clean cover slip apply a

minimal amount of Vaseline to the
four corners with a thin glass rod, or
an applicator stick.

2. Using a 2mm loop place a loopful of

the given suspension at the centre of
the cover slip.
3. Invert a cavity slide over the cover
slip so that the drop of suspension is
4. Quickly and carefully turn the slide
over so that the coverslip is
uppermost with the drop hanging
from the same into the cavity

5. Focus the edge of the drop under

low power of the microscope after
reducing the light using the
condenser.

6. Turn the high power objective ,

identify the edge and carefully focus
the edge with fine adjustment.
