Polymerase Chain

Reaction
 The polymerase chain reaction (PCR)
is a technique to amplify a piece of
DNA very rapidly outside of a cell.
Process

- Buffer, polymerase Enzyme, oligonucleotide primers,

nucleotides,

 Denaturation temperature:
94°C

 Annealing temperature
54°C

 Extension temperature
72°C
Process
 25-30 cycles

2 minute cycles

 DNA thermal cycler

Collecting DNA evidence
 Blood  Old samples

 Semen  Small samples

 Tissue  Degenerated
samples
 Saliva
– Envelope, stamps,
cigarette butts
Advantages
 Quick results

 Small amount

 More stable
PCR Uses
 Diagnose genetic
diseases

 Find bacteria and

viruses

 Study human evolution

 Clone DNA of Egyptian

mummies

 Do DNA fingerprinting
Forensic Uses
 Can apply any biological technique

 Identify victim/suspect

 Paternity/maternity tests
Future Uses
 DNA as “id card”

 More studies
