Introduction to purine
• Purine is a heterocyclic aromatic organic compound that consists of a
pyrimidine ring fused to an imidazole ring.
• It is water-soluble.
• Purines occur in all organisms as the bases of DNA and RNA
• Examples of purines include caffeine, xanthine, hypoxanthine, uric acid,
theobromine, and the nitrogenous bases adenine and guanine
Salvage pathway for purine Synthesis
• Introduction :-
• The pathway involved in the conversion of free purine to nucleotide
is called salvage pathway
• Free purine are salvaged and used over again to remake purine
nucleotide in which purine ring system is assembled step by step on
ribose -5-phosphate .
• Salvage pathway is much simpler and requires far less energy than
does de novo synthesise
Salvage pathway for purine synthesis
• Salvage reaction :-
• In salvage rxn ribose phosphate moiety of PRPP is transferred to the
purine form. There are two salvage enzymes with different specificities.
• 1)Adenine phosphoribosyl transferase :- catalyse the formation of adenine
nucleotide (AMP) from adenine
• 2)Hypoxanthine Guanine phosphoribosyl transferase (HGPRTase) :-
catalyse the formation of ionosine from hypoxanthine and guanine.
Introduction to pyramidine
• Pyrimidine is an aromatic heterocyclic organic compound similar to
pyridine
• One of the three diazines (six-membered heterocyclics with two nitrogen
atoms in the ring), it has the nitrogen atoms at positions 1 and 3 in the
ring.
• Pyrimidines include cytosine, thymine,
and uracil whereas purines include adenine and guanine.
Salvage pathway for pyramidine synthesise
• Similar to purine pyramidine are also recoverd from derivative
Intermediate of nucleic acid such as DNA and RNA
• The recovery of pyramidine is catalyse by the enzyme pyramidine
Phosphoryboslytransferase.
• Pyramidine phosphoribosyltranferase enzyme utilise PRPP as the source
of ribose -5- phosphate.
Significance of salvage pathway
• Salvage pathway provide purines nucleotide for tissues which are
incapable to synthesize purine nucleotide by De novo pathway , e. g
human brain, erythrocytes, and polymorphonuclear leukocytes
