ACT 11 - Isolation of DNA

1. DNA was extracted from a banana as a white clump of string-like strands. Tests found inorganic phosphate, purines, and deoxyribose in the extract, confirming it was DNA. 2. The document discusses the structure of purine and pyrimidine bases in DNA. Purines adenine and guanine contain two carbon rings fused to an imidazole ring, while pyrimidines cytosine and thymine contain a single ring with two nitrogen atoms. 3. Formation of a nucleotide involves an ester bond between a purine or pyrimidine base and deoxyribose sugar. Nucleotides are bound together in DNA through phosphodiester bonds between

 GUIDE QUESTIONS
 1. Name and write the structure of the purine ad pyrimidine
bases present in DNA .
 Purine A heterocyclic aromatic organic compound made of
pyrimidine ring with an imidazole ring fused with it. It has two
carbon rings and nitrogen atoms. Adenine (A) and Guanine (G)
are purines.
 Pyrimidine A heterocyclic aromatic organic compound having a
single ring structure and two nitrogen atoms at position 1 and 3
of the ring, Cytosine (C) and Thymine (T) are pyrimidines.

PYRIMIDINE
 2. Explain for the formation of precipitate in the test for
purines.
 The gelatinous white precipitate formation in the purines
test is due to the NH4OH which break the beta glycosidic
bond between deoxyribose sugar and purine bases, it
causes a reaction with the silver nitrate thus realizing the
formation of white precipitate indicating the presence of
the purines.
 3. What bonds are present in the formation of
nucleotide and the binding of nucleotide to another
nucleotide in DNA? Show the bonds.
 Ester bond and glycosidic bond are present in the
formation of nucleotide whereas, phosphodiester
bonds and hydrogen bonds are present in the
binding of a nucleotide to another nucleotide.
