DNA EXTRACTION

FROM CHEEK
CELLS
MATERIALS
Clean Plastic cups (clear)

Table salt

Distilled water

Dishwashing liquid

Ethyl Alcohol, Ice cold

Test tube /vial / any small clear  container


PREPARE 1. SALINE SOLUTION- Add half pinch salt to
a glass full of distilled water
THE
2. DISHWASHING LIQUID SOLUTION- 1 par t

FOLLOWING dishwashing liquid and 3 parts water  

3. Ice cold ethanol / alcohol- refrigerate

SOLUTION
Part 1: CELL HARVESTING

Do not eat or drink 15 minutes before sample collection.


• Pour saline solution into a clean plastic drinking cup until half full.
• Pour the saline solution into your mouth and “swish” the water around  between
your cheeks and gums for 60 seconds.
• You may “scrape” the inside of your cheeks with your teeth to help slough  off
cheek cells to increase DNA yield.
• Spit all of  the water back into the empty plastic cup. You may transf er the
solution into a test tube or vial if you have one.

• Your plastic cup now contains cells from your cheeks, called buccal
cells.  These cells still contain an intact nucleus, where the DNA is
found.
Part 2: CELL LYSIS

• Add 40 drops dishwashing liquid solution


to the collected saline solution 
• Stopper the test tube , when using
different container mix by inverting it 
gently
• Do not shake the test tube.  Excess
shaking can break the long strands
of DNA, making it harder to see and
collect when precipitated.​
Part 3: PRECIPITATION

• Holding the test tube at a slight angle,  carefully  add


25mL of the ice-cold ethyl alcohol down the side of the
test tube so it forms a layer over the  cheek cell
mixture.  Do not mix the water and ethanol layers.
• Hold the test tube /vial upright for 60 seconds and
obser ve what happens at the  inter face between the
ethyl alcohol and the cheek cell solution.
