KOMPLEMENT

The complement system consists of over 30 proteins that activate in a cascade to destroy microorganisms via three pathways: classical, lectin, and alternative. The classical pathway is initiated by antibody-antigen reactions, while the lectin pathway involves mannose-binding lectin. The alternative pathway activates spontaneously on microbial surfaces. All three pathways converge at C3 cleavage and generate C5 convertases that cleave C5 to initiate the lytic pathway. This inserts the membrane attack complex (MAC) into microbial cell membranes to cause cytolysis. Complement activation leads to inflammation, phagocytosis, and microbial killing.

C2b Prokinin; cleaved by plasmin to yield kinin, which

results in edema
C3a Anaphylotoxin; can activate basophils and mast
cells to degranulate resulting in increased vascular
permeability and contraction of smooth muscle cells,
which may lead to anaphylaxis

C4b C4 binding protein (C4-BP) and Factor I; C4-BP
facilitates degradation of C4b by Factor I; C4-BP
also prevents the association of C2a with C4b thus
blocking formation of C3 convertase
