INSTRUCTIONS: In this format, you must complete each step with the information provided in
class, the guide uploaded by the teacher in the Aula Virtual and your own research on the topic.

Laboratory Exercise 1

1. INTRODUCTION
Each living being on our planet is made up of cells, which at its core is DNA, which allows
them to store, share and duplicate their genetic information, thanks to this it is possible to
share or duplicate the genetic information of some A living being can also pass from one body
to another, making the new organism inherit the previous genetic information from the past
organism.
Thanks to the broad concept that DNA has, there are organisms which share similarities in
their DNA without the need to be of the same species, such as bananas, which have 60% DNA
similar to that of human beings, thanks with similarities like these, we can study other
organisms, with similarities or differences to ours, to see what their result is.

2. OBJECTIVES
- To be able to make the DNA of a banana visible.
- Learn the chemical reactions to perform in order to obtain this result.
- Observe the result and verify that everything has gone correctly.
3. MATERIALS AND EQUIPMENT.
 Lab Coat
 Banana
 1 mortar
 Detergent
 Distilled water
 Salt
 Funnel
 Filter paper
 95% ethylene
 2 beakers
 1 rack
 1 test tube
 1 pipette

4. PROCEDURE
1. Crush half a banana with a mortar and add water.
2. In a beaker, add a solution of: detergent (2 Tbsp), distilled water (15 cc) and a pinch of salt.
3. Add to step two and the mashed banana, then stir for 5 minutes.
4. The test tube is placed in a rack and a funnel is placed on top with the filter paper and the. Add
the preparation from step 3.
5. Add 15 ml of cold alcohol to the filtered solution and wait for your result.

5. RESULTS
After filtering the solution and adding the cold alcohol, it was only a matter of minutes to see
the result clearly, as will be shown in the following photos. Thanks to the clean filtering that
was obtained, just adding the alcohol made the DNA visible, causing it to float above the
solution, leaving it in the background and protruding this above.

6. CONCLUSION
We obtained a very satisfactory result, verifying that in a few steps it is possible to clearly see
the DNA of a banana, we had a very clear result, since we were very careful when filtering to
achieve a cleaner filter and when adding the alcohol do not mix these two. This experiment left
us shocked to see its good result that was achieved and thanks to this we verified the fact that
this experiment does work.
7. QUESTIONS
1. What purpose was the banana mashed and the pineapple juice obtained?
To make it easier to break cell walls and disperse cell.

2. Why was the solution made with detergent?


The detergent together with the saline solution better dissolved cell and nuclear walls and
membranes, which helped to release the DNA.

3. What was the need to do the filtration in the above steps?


It was necessary to filter the proteins, lipids and solids being removed from the aqueous DNA
solution.
4. What was the function of the alcohol in the experience?
What the alcohol did was that the DNA separated from the water, and after the DNA separated
from the aqueous solution, this makes it visible on the surface.
