SYLLABUS OBJECTIVE: Module 1 Obj. 4.3 – To explain the effects of pH, temperature,
enzyme concentration and substrate concentration on enzyme action.

APPARATUS: Stopwatch, test tubes, water baths (0oC, 20oC, 40oC, 60oC, 80oC and
100oC), cork borer, knife, white tile, ruler, soap detergent, 1.0M Hydrogen
Peroxide, potato
METHOD:
1. Use the cork borer to obtain potato cylinders. Cut potato pieces 2mm in length.
2. Incubate six 10 mL test tubes containing 10 mls hydrogen peroxide at different
temperatures: 0oC, 20oC, 40oC, 60oC, 80oC and 100oC for five minutes.
3. Add one drop of soap detergent to each tube.
4. Add 3 cubes of potato to the six test tubes.
5. Measure the height of foam after 1 minute.
6. Carry out the procedure out three times at each temperature and calculate the average.
7. Tabulate the results and record what is observed as temperature is increased.
8. A graph of Height of Foam (mm) against Temperature (oC) was drawn.
