Practical Sheet Enzymes

The document outlines a practical experiment to investigate the effect of temperature on enzyme activity using catalase from potatoes. It details the equipment needed, measurements to be taken, and a step-by-step method for conducting the experiment. Additionally, it includes sections for recording results and answering questions related to the experiment's findings.

Practical 3: Enzymes

AIM

Investigate how temperature affects enzyme

activity using catalase in potatoes

EQUIPMENT MEASUREMENTS TO BE TAKEN

 3 test tubes  Height of the ramp (low, medium, high).
 Test tube rack
 Fresh potato (cut into equal-sized  Distance traveled by the toy car in
cubes, ~1 cm³ each) centimeters.
 15 ml of 3% hydrogen peroxide  Time taken for the car to reach the
solution (5 ml per test tube) stopping point.
 3 water baths set to 5°C, 20°C, and
40°C EQUATIONS TO USE
 Thermometer
 Stopwatch
 Calculating mean
 Measuring cylinder (for precise liquid
measurement)
 Forceps (to handle potato cubes)
 Marker pen (to label test tubes)

METHOD
 Using a marker, label three test tubes as 5°C, 20°C, and
40°C.
 Place the test tubes in a test tube rack to keep them
stable.
 Prepare Potato Cubes:
 Cut three equal-sized potato cubes (~1 cm³ each).
 Use forceps to place one cube into each test tube.
 Add Hydrogen Peroxide:
 Use a measuring cylinder to measure 5 ml of 3%
hydrogen peroxide solution.
 Carefully pour 5 ml into each test tube containing the
potato cube.
 Start the Stopwatch:
 As soon as the hydrogen peroxide is added, start the
stopwatch immediately.
 Observe and record the time until bubbles stop
forming
 Record your results on a results table

QUESTIONS (2 MARKS)

1. Explain how temperature affected the enzyme activity in this experiment. Use your results to
support your answer.

2. Why was it important to use equal-sized potato cubes and the same volume of hydrogen
peroxide in each test tube?

3. If the experiment were repeated at 70°C, what would you expect to happen to the enzyme
activity? Explain your reasoning using scientific concepts
