AIM: To study the different routes of drug administration in laboratory animals.

PRINCIPLE:
The onset duration of action of a drug depends on the route of administration. For example, when a drug
is given by intravenous route the effect is seen instant as compared to oral administration of drug when it
takes up to one hour to show the effect. Before performing this experiment, the students are advised to
have the back ground of various routes of administration their relative merits & demerits.
Pentobarbital sodium is chosen for the present experimental because it shows onset of action and point
is also clear when the animals recover from sleep moreover its action can be seen when administered.
For the present purpose, three routes are chosen i.e. intraperitoneal. Intravenous and oral route for the
administration of drug.
1. Oral route:
Care should be taken that the administered material should not enter the respiratory tract.
Accidental entry of the material in respiratory tract is traced by appearance of material in nasal cavity
and violent straining by the animal.
For mouse, rat and guinea pig:
A eustachian catheter or a hypodermis needle) may be used.
A hypodermis needle if suitable gauze (15-16 for rat and guinea pig and 18-20 for mouse) is blunted
and a ball of solder is put around the tip to avoid any injury to the inner surface of mouth and
esophagus.
The needle is bent by 20-30° at around 2 cm distances from the tip.
The animal is grasped by the rope of the neck by one hand and thus making the animal to open the
mouth.
The feeding cannula is inserted through the intra dental space and gently pushes into esophagus.
Once the desired position of the cannula tip is obtained, the drug is gently pushed with the help of
syringe.
For rabbit
A no. 12 French catheter or a no. 9 soft rubber catheter (5 μm diameter) is used and is inserted behind
the incisors of the animal. While removing the tube, outer end is occulted.
2. Parenteral routes:
a) Intravenous:
Injection of drug solution is made directly into the systemic circulation.
In this route, the bioavailability of the drug is100%
b) Intraperitonial:
Injection is made into the peritoneal cavity i.e. the cavity between the abdominal wall and visceral
organs.
The cavity of absorption in this route is maximum.
c) Intramuscular:
Injection is made into muscle.
In this type of administration, the amount of drug administered is small and also the rate of absorption is
steady, small and rate of drug absorption is slow.
