Lab 2 Assignment

The lab assignment requires students to analyze absorbance data for methylene blue solutions, comparing results between two groups. Students must generate descriptive statistics, create tables, and construct a scatter plot based on their findings. The assignment emphasizes accuracy and precision in measurements, with specific calculations for concentration based on absorbance values.

Review Guidelines for Writing Assignments in lab manual introduction before

beginning. Table titles follow the same rules as figure titles, but they are found above the

table.

Please submit your assignment at the beginning of your next lab. Your pages should be in

the same order as below. You should include the following information:

Your name: Yatta Koroma

Your partner’s name: Romiel Richard

Your volume #: 4

Your group #: 1

Required volume of methylene blue to be pipetted: 800 µl

Required volume of water to be pipetted: 600 µl

PRECISION

1. Generate two tables containing descriptive statistics calculated on your data and on

the data of the other group working with the same volume.

2. Answer the following questions related to your descriptive statistics.

Descriptive statistics questions

a) Give the mean absorbance values obtained by your group and by the other group

working with the same volume. Give one or more specific reason(s) for the

similarity or difference in absorbance values.

• The mean of Group 1 I got 0.2766 while group 2 is 0.2614, Both groups are

working with the same volume of solution, so the basic conditions for

measuring absorbance are essentially the same. However, there may still be

differences in the results due to various factors. It's possible that the two

groups are using different instruments, or even the same instrument with

slight calibration differences, which could affect the readings. Additionally,

experimental error could arise from factors like variations in technique or

inconsistent reading of absorbance, leading to discrepancies in the results.

b) Give the standard deviations obtained by your group and by the other group

working with the same volume. Give one or more specific reason(s) for the

similarity or difference in standard deviations.

Group 1 = 0.012 while Group 2 = 0.014.

• The standard deviation indicates how spread out the absorbance values are
from the mean. A lower standard deviation means the values are closer to
the mean, implying greater precision in measurements. Group 1 is slightly
more precise with a lower standard deviation (0.012), while Group 2 has a
slightly higher standard deviation (0.014), indicating more fluctuation in
their measurements.

THE STANDARD CURVE (remember: a curve can be a straight line)

3. Include Table 6 from page 2-26. (2 marks)

Volume # Initial volume of Final concentration Average

1.4 mmol/L of methylene blue absorbance
methylene blue (µl) (mmol/L)
All water 0 0 0
Volume 1 Group 1 200 0.2 0.115
Volume 1 Group 2 200 0.2 0.100
Volume 2 Group 1 400 0.4 0.307
Volume 2 Group 2 400 0.4 0.339
Volume 3 Group 1 600 0.6 0.468
Volume 3 Group 2 600 0.6 0.398
Volume 4 Group 1 800 0.8 0.277
Volume 4 Group 2 800 0.8 0.261
Volume 5 Group 1 1000 1 0.732
Volume 5 Group 2 1000 1 0.86

4. Construct a scatter plot of average absorbance for each group as a function of final

concentration of methylene blue using the class data.

5. Give the absorbance of the unknown concentration of methylene blue solution on the

instructor's bench.

The absorbance value is around 0.3875.

6. Use the standard curve for the absorbance of methylene blue at 410 nm to answer the

following question: What is the concentration of the methylene blue solution on the

instructor’s bench?

Group 1 Group 2

Absorbance 1= 0.307, Absorbance 2= 0.468.

Absorbance = 0.3875

Conc 1 = 0.40 M, Conc 2 = 0.60 M.

Concentration = (Absorbance -Absorbance1/ Absorbance2 - Absorbance1) * (Conc 2

−Conc 1) +Conc1

Concentration = (0.3875-0.307/0.468-0.307) * (0.60-0.40) +0.40

Concentration = 0.10 + 0.40 = 0.50M

ACCURACY

7. Consider the standard curve and the results obtained by your group and the other

group working with the same volume. Determine which of the two groups was more

accurate.

• Group 1 was more accurate because their measured concentrations closely

matched the expected concentrations from the standard curve, with no

2-7

noticeable deviations. On the other hand, Group 2 had minor differences

between their measured concentrations and the expected values for some

absorbance readings.

What evidence did you use to determine this?

• The evidence for this conclusion is that Group 1’s concentrations were exactly

as expected, while Group 2 showed small discrepancies, indicating that Group

1’s measurements were more precise and accurate. Therefore, Group 1

demonstrated greater accuracy. Based on the trend of the standard curve.
