BLOOD SMEAR AND
STAINING (BLOOD FILM)
A blood film or peripheral blood smear is a thin layer of blood
spreaded on microscope slide and then stained to allow
various blood cells to be examined microscopically.
AIM OF BLOOD SMEAR
- Examined to investigate hematological problems (disorder of
the blood).
-To look for parasite within blood such as malaria and filaria.
-To look number and shape of blood cells, and any change in
the appearance of blood cells.
-Provide report rapidly and at low cost, useful information
about the patient’s condition.
Three basic steps for preparation of blood film:
1- Preparation of blood smear 2- Fixation of blood smear 3-
Staining of blood smear
MATERIAL AND REAGENTS
Compound microscope.
Glass slides(Spreader and stationary)
Cotton.
Sterile finger pricking lancet.
Detol or alcohol
Leishman´s stain.
Buffer or D.W.
Cedar wood oil.
PREPARATIONS OF BLOOD
SMEAR AND STAINING :
Fill a capillary tube three-quarter full with the anticoagulated
specimen.
A drop of blood in the center of a clean glass slide 1 to 2 cm
from one end.
Place another slide (spreader) with smooth edge at an angle of
30 to 45° near the drop of blood.
Move the spreader backward so that it makes contact with drop
of blood.
Then move the spreader forward rapidly over the slide.
A thin or thick peripheral blood smear is prepared.
Dry it and then stain it
STAINING OF BLOOD
SMEAR
Blood cells have different structures, which take different
stains. Some are basophilic; others are acidophilic while some
cells accept neutral stain. Therefore, the stain used is
combination of these three. Such stains are called as
Romanowsky stain.
The three different types of Romanowsky stains are commonly
in use.
Leishman stain Giemsa’s stain Wright’s stain.
Each of the stain contains acidic stain, basic stain and buffer
solution. Generally, methylene blue are basic stains, and Eosin
are the acidic stains in use.
PROCEDURE:
Put the smeared slide on the rack.
Put 10 drop of Leishman stain on the blood smear & leave for 1
– 2 min.
Add double drop amount (20 drop) of buffered water or D.W.
leave it for 8 – 10 min.
Wash the slide with D.W.
Clean the back of the slide by filter paper.
Leave the slide to dry in air.
Exam the slide under microscope using oil immersion.
To obtain best smear stain Leishman stain used , because
there is no water in its content
