1. L1 is a slide of a transverse section showing a tubular part of the digestive system of a mammal. You
are not expected to have studied this material.

a) Draw a large plan diagram showing only the features of the wall of the tube as in the sector shaded in
Figure 1.

Annotate your diagram to describe one difference between the innermost layer and outermost layer.

The figure below is a photomicrograph showing a transverse section through a part of the specimen on
slide L1.

b) Make a large drawing of the four cells, labelled P, on the figure. On your drawing, use a label line and
label to show one nucleus.

2. Figure 1 shows a stage micrometer viewed through a microscope with an eyepiece graticule scale. The
smallest measurement on this stage micrometer is 0.1 mm.

You are required to:


• find the area of the field of view, using Figure 1
• count the number of stomata in a quarter of the field of view, using Figure 2
• calculate the number of stomata per mm2.

a) i. Calculate the area of the field of view, using:


• the formula for the area of a circle πr2
• π = 3.14
• r = radius of the field of view.
You will lose marks if you do not show your working or if you do not use appropriate units.

Figure 2 is a photomicrograph of the lower surface of a leaf, with the same field of view as in Figure 1.

Figure 2 shows stomata on a leaf surface. There are too many stomata to count so the technique of
sampling may be used to estimate the number of stomata in the field of view.

A sample should be counted in a known smaller portion and then the result multiplied to obtain an
estimate of the number of stomata in the whole field of view. For example, if the number of stomata is
counted in a quarter of the field of view then this number would be multiplied by 4 to obtain the
estimate of the total number in the field of view.

ii. Count and record the sample number of stomata in the quarter of the field of view in Figure 2.
• Mark clearly on Figure 2 each of the stomata counted.
• Estimate the total number in the whole field of view.

Calculate the number of stomata per mm2. You will lose marks if you do not show your working.

Figure 3 is a photomicrograph of the lower surface of a different leaf, with the same field of view (using the
same lenses) as Figure 2.

b) Identify the observable differences between the surface of each leaf shown in Figure 2 and Figure 3.

3. The figure below is a photomicrograph of a stained transverse section of a stem of a different plant
species.

a) Calculate the actual length of the line A shown on the figure. You may lose marks if you do not show
your working.

b) A student observed a different plant of the same species shown in the figure. The student determined
the ratio of the radius of the stem to the length of one air space as 266:82.

However, a ratio may be simplified to the smallest possible whole number on each side. In this
example, both sides of the ratio 266:82 are divisible by 2, so the simplest ratio for the measurements
taken by the student is 133:41. The actual radius of the stem in the figure is 825 μm.

Determine the simplest ratio of the radius of the stem in the figure to the length of an air space (line A).
