AMINO ACID AND PROTEIN METABOLISM

PART A. Understanding the Amino Acid Pool


1. List and describe the three main sources of the amino acid pool. List and
describe the ways amino acids are depleted from the amino acid pool.
2. Explain how amino acids from the amino acid pool contribute to the
synthesis of proteins in the body. Why is maintaining an adequate amino
acid pool crucial for muscle growth and repair?
3. Track your dietary protein intake for one day. List the main sources of
protein you consume and estimate the total intake in grams. Reflect on how
this dietary intake contributes to your body’s amino acid pool and
subsequent metabolic processes.
PART B. Protein Digestion
1. List and describe the main types of proteolytic enzymes, their proenzyme
forms, and their location. What distinguishes their mechanisms of action?
2. Explain how proteases recognize specific cleavage sites on protein
substrates. Provide an example of a protease and its typical cleavage site.
3. Why is it that enzymes are produced by organs in their proenzyme form?
Explain how the proenzyme forms in Part B No. 1 are activated.
4. Illustrates where the different peptidases (enzymes) would catalyze
hydrolysis of the peptide bonds.

PART C. Urea Cycle


1. Draw a labeled diagram of the urea cycle, showing all key intermediates
and enzymes involved (e.g., carbamoyl phosphate, citrulline,
argininosuccinate, arginine, and ornithine). (only the names)
2. Highlight which steps occur in the mitochondria and which occur in the
cytoplasm
3. Write a brief description (3-4 sentences) of each step of the urea cycle,
including the specific enzymes catalyzing each reaction and the locations
within the cell.
4. What is the main purpose of the urea cycle in the liver? Why is the removal
of ammonia essential for the body?
