Hitachi U-2000 Spectrophotometer –Basic Operation


These directions are for a normal wavelength scan, using the keypad (No computer). For
further details, or other applications, consult the manual.

1. Sign the logbook.


2. Turn on the POWER switch, located under the keypad.
3. After the self-test is complete (about 1 min), press AUTO ZERO.
4. Insert sample, and reference if desired. Plastic cuvettes can be used with aqueous
solutions in the visible range (>340 nm). Use quartz for UV, glass or quartz for
organic solvents.
5. General menu navigation. Use the up and down arrow keys to move the
highlight. Use ENTER to select a parameter group. Use the keypad, then
ENTER to change the value of a parameter. The range of values for each
parameter is given at the bottom of the screen. The RETURN key returns to the
previous menu. MAIN MENU returns to the Main Menu.
6. From the Main menu, select W L SCAN, then ENTER. Use the arrow keys to
view groups of parameters. If you wish to change any, highlight the group, then
ENTER. Highlight the parameter, then use the keypad and ENTER to change the
value. Be sure the PRINT parameters under INSTR SETUP are set as you want
them. For a wavelength scan TEXT PRINT is normally OFF, GRAPH PRINT
normally ON. BASELINE is normally set to SYSTEM. If set to USER, you
should run a new user baseline each time you change scan parameters.
7. When all parameters are set to desired values, press FORWARD, then START to
start the scan. The scan will display on the screen. When the scan is complete, it
will be printed, if GRAPH PRINT is ON.
8. After the spectrum prints. The PRINT PARAMS button can be used to print the
scan parameters, if desired.
9. When you are finished, turn the power OFF, and remove your samples.
10. The instrument manual and extra printer paper are located in the drawer
under the instrument.

Hitachi U-2000 Spectrophotometer –Computer


Controlled Operation
These directions are for a normal wavelength scan, using the computer. Computer
control gives more attractive printed output, as well as files that may be exported to other
applications.

1. Sign the log book.


2. Turn on the computer, monitor, and printer if needed. Wait until the computer
boots.
3. Turn on the spectrophotometer. From the main menu, choose REMOTE
CONTROL, then ENTER. Do not touch the instrument keyboard again.
4. From the computer keyboard, type LC to start LabCalc.
5. General LabCalc menu navigation. Press F2 to bring up the menu. Use the
keys to highlight a menu choice, the keys to highlight a sub-menu
choice, ENTER to choose. If a numerical value is requested, type it, followed by
ENTER. ESC will back up one level.
6. From the LabCalc menu, choose Collect. Then choose Wavelength Scan. Current
parameters will be transferred to the U-2000. The U-2000 screen will react as if
you were entering values manually.
7. To change parameter values, choose a parameter from the menu line, ENTER,
type a new value, then ENTER.
8. To start the scan choose Go. The file will be automatically saved to the disk in
the directory C:\LC. A different directory can be specified by choosing
Environment/Directory from the menu.
9. To retrieve existing data, choose File/Retrieve from the menu. Use the
keys to highlight the file name, then ENTER.
10. To change the wavelength limits of the displayed spectrum, use the +/- keys.
11. To plot, choose Plot/Graphics Printer from the menu.
12. To export data to a text file, choose File/Export from the menu.
13. When finished, choose Quit.
14. Turn off the computer, printer and spectrophotometer, and remove your samples.
