MDP fabrication method
To fabricate the micro-dome-shaped polymer (MDP), a commercial dimethylpolysiloxane
(PDMS) kit (SYLGARD®184, Dow Corning) was used. First, base and curing agents were mixed
with a weight ratio of 10:1 and then the mixed liquids were de-aired in a vacuum desiccator. A
small piece (approx. 4 mm × 4 mm) was cut from a commercially available PDMS sheet (PF-X4-
17 mil., Gel-Pak) and placed on a microscope slide. A droplet of the mixed liquid agents was
dropped with a toothpick onto the small PDMS sheet and cured at 130 °C for 5 min. This created
a large dome-shaped drop of PDMS. A second droplet of the mixed liquids was then dropped onto
the first one using a gold wire (diameter, 25.4 µm), and the sample was then cured at 130 °C for 5
min to create a mid-sized dome-shaped PDMS droplet on the large dome-shaped droplet. Finally,
another droplet of the mixed liquid was dropped with a tungsten needle (tip diameter, ~200 nm;
TP-0002, Micro Support Co., Ltd.) and the sample was subsequently cured once more. With this
sequence of dome formation steps, a size for the upper dome of ~30 µm (diameter) was achieved.

PVC-MDP fabrication method
A commercially available PVC sheet (RIKEN WRAP, Riken Fabro Corp.) was used to cover the
MDP to act as an adhesion layer. The procedure for covering the MDP with the PVC sheet is
shown in Figure S1. First, the PVC sheet is positioned on the microscope slide to obtain flat surface
[Figure S1(a)]. Next, four lengths of double-sided tape are placed on PVC sheet such that a square
shaped opening between them is generated [Figure S1(b)]. The PVC film is cut at the outside of
double-sided tape and both the PVC film and the double-sided tape are removed together from the
microscope slide [Figure S1(c)]. The PVC film with its double-sided tape window is placed on
MDP structure with the double-sided tape underneath [Figure S1(d)]. Thus, the PVC sheet is fixed
to the microscope slide while fully covering the MDP [Figure S1(e)]. Finally, microscope slide
with the PVC-MDP attached is mounted onto the optical microscope system [Figure S1(f)]; in the
