Problems

1. For a single point standard addition, show that

Sx Cs x V s
Cx = S −S x Vx
(5pts)
T x

Where:

Sx is the signal due to analyte

ST is the total signal
Cs is the concentration of standard added
Vs is the volume of standard added to the solution
Vx is the volume of sample taken from the original solution
Cx is the concentration of analyte in the original solution

2. Show that for a multiple point standard addition method, (5pts)

b C std
Cx = m x V (5pts)
x

Where:
b is the y-intercept
Cstd is the concentration of standard added to the sample
m is the slope of the line
Vx is the volume of the sample taken for analysis

3. The concentration of phenol in a water sample is determined by separating the phenol from
nonvolatile impurities by steam distillation, followed by reacting with 4- aminoantipyrine and
K3Fe(CN)6 at pH 7.9 to form a colored antipyrine dye. A phenol standard with a concentration
of 4.00 ppm has an absorbance of 0.424 at a wavelength of 460 nm using a 1.00-cm cell. A
water sample is steam-distilled, and a 50.00-mL aliquot of the distillate is placed in a 100-mL
volumetric flask and diluted to volume with distilled water. The absorbance of this solution is
found to be 0.394. What is the concentration of phenol (in parts per million) in the water
sample?

4. AAS was used to analyze the calcium content of milk. Solution 1 was prepared by mixing
10.0mL of milk with 5.00mL of a 1.00x10-4M of Ca2+, then diluting to 100.0mL. Solution 1 gave
an absorbance of 3 units. Solution 2 was prepared by diluting 10.0mL of milk to 100mL and
gave an absorbance of 2.0 units. What’s the concentration of Ca 2+ in the original milk
sample? (5 pts).
5. The iron content of municipal water supply was determined via AAS. The absorbance of the
water sample after a five-fold dilution, was 0.646. A standard solution prepared by
dissolving0.1483g of pure iron in acid, diluting to 250.0mL and then making 100-fold dilution,
had an absorbance of 0.813. What is the Fe content of the water sample? (5pts)

6. Nitrite is commonly determined by a colorimetric procedure using a reaction called the

Griess reaction. In this reaction, the sample containing nitrite is reacted with sulfanilimide
and N-(1-Napthyl) ethylenediamine to form a colored species that absorbs at 550 nm. Using
an automated flow analysis instrument, the following results were obtained for standard
solutions of nitrite and for a sample containing an unknown amount:

Solution Concentration, µM Absorbance at 550nm

2.00 0.065
6.00 0.205
10.00 0.338
14.00 0.474
18.00 0.598
Unknown 0.402

Determine the concentration of nitrite in the sample .

7. The following volumes of a standard 10.0 ppb F - solution were added to four 10.00-mL
aliquots of a water sample: 0.00, 1.00, 2.00, and 3.00 mL. Precisely 5.00 mL of a solution
containing an excess of the strongly absorbing Al-acid Alizarin Garnet R complex was added
to each of the four solutions, and they were each diluted to 50.0 mL. The fluorescence
intensity of the four solutions were as follows:

Vol. of std. added, mL Meter reading

0.00 68.2
1.00 55.3
2.00 41.3
3.00 28.8

Calculate the concentration of F- in the sample in ppb.

8. In a chromatographic analysis, a solution was prepared with 0.0837M Octane (A) and
0.0666M nonane (B). Each one produced a peak area of AA =423 and A8 =347.
10.0mL of 0.146MNonane was added to 10mL of an unknown sample, and the sample
diluted to 25 mL. The peak areas obtained were : A A =553 and AB =582. What is the
concentration of Octane in the unknown sample? (5 pts).

9. Compounds C and D gave the following HPLC results. A solution was prepared by mixing 1.23
mg of D in 5.00mL with 10.00mL of analyte C and diluting to 25.00mL. Peak areas of the
samples were obtained: 3.33cm2 and 2.22cm2 for C and D respectively. What is the
concentration of C in the sample? (5pts)
Compound concentration Peak areas
C 236ug/mL 4.42 cm2
D 337ug/mL 5.52cm2
