Pre-Lab Review:

(Enzyme Lab)
Learning Objectives:

By the end of this class you will be able to :

- To observe the breakdown of hydrogen
peroxide toxin by potato’s enzyme catalase.
- describe the specific action of the enzyme
catalase
See what things can denature an enzyme and
what happens when it does get denatured

2
Do you remember?

- What is an Enzymes?
Enzymes are proteins that act as
catalysts in living organisms,
speeding up chemical reactions
without being consumed in the
process.
What does the Shape provide?
The shape of an enzyme provides
a specific active site where the
substrate can bind, allowing the
enzyme to catalyze the reaction
with precision.

3
Enzyme:
● Protein that speeds up
chemical reactions.

● Enzymes allow
reactions to occur at
the rate necessary for
life.

4
Enzyme:
★ In animals, an important function of enzymes is to help
digest food.

★ Denaturing a protein changes its shape and so it can no

longer function properly.

5
Can We damage Enzymes?

- Enzyme shape is everything (destroy

the shape destroy the enzyme).

- Two things can destroy/denature an

enzyme

Two things that can denature an enzyme

are high temperatures and extreme pH
levels.me?

6
Hydrogen Peroxide?

- Is a chemical compound -
- H2O2 is damaging
internally, it's a waste
product of our organs
(livers, kidneys) if it
builds up in our bodies
its toxic.

7
Catalase:

★ Common Enzyme
found in nearly all living
organisms exposed to
oxygen.
★ Catalase Breaks down
hydrogen peroxide into
non toxic chemicals.
★ (Potato)

8
Let's think about this??

- What would you expect If catalase is present, you would

to see if….. expect to see the breakdown of
hydrogen peroxide into water and
- Catalase is present? oxygen. If catalase is not present,
- If catalase is not you would not observe the
present.? breakdown of hydrogen peroxide.

9
Clean up :

- Dump your solutions into

the red bucket.
- Rinse out the test tubes.
- Dry your test tubes/ trays
- Place trays back with all the
materials.

10
Let's Start the Lab!!!!

11
Lets get set…

Tube # What’s in it? What do you expect to see?

Test tube 1
Does catalase breakdown
hydrogen peroxide?

Test tube 2
Does acid denature catalase?

Test tube 3
Does heat denature catalase?
