PROCEDURE

THEORY:
Thin Layer Chromatography is a technique used to isolate
nonvolatile mixtures. The experiment is conducted on a sheet of aluminium
foil, plastic, or glass which is coated with thin layer of absorbent material.
The material usually used is aluminium oxide, cellulose, or silica gel.
On completion of the separation, each component appears as spots
separated vertically. Each spot has a retention factor (Rf) expressed as:

Rf = distance travelled by the sample/distance travelled by the solvent.

PROCEDURE:
1. Take a datura fruits and make it into pieces, keep it in the mortar and
crush it with the pestle.
2. Add distilled water to it until it comes to the paste format.
3. Now take a TLC paper and keep a drop of this paste on the TLC at least
a cm away from the base and wait for few minutes.

3. thermometer.

PROCEDURE

As per Bureau of Indian standards for petroleum products we are considering

following parameters:
1. Flash point and fire point
2. Specific gravity
3. Viscosity

THEORY: A good lubricating oil should not volatize under the working
temperature. Even same volatization takes place. The vapor formed should
not form inflammable mixture with oil air under the condition of other
lubricance.

FLASH POINT: It is the temperature to which a combustible liquid must be

heated to fire of sufficient vapors to form momentarily a flammable mixture
with air when a small flame is brought near the surface of liquid under
specified condition.

FIRE POINT: Fire point of oil in the lowest temperature at which it will give
enough vapor, which on raising will begin to produce a continuous flame
above the oil.

Page no: 3
Applied and Action Learning

PROCEDURE:
1. Thoroughly clean and dry all the parts of the cup and its accessories before
starting the test, being sure to remove any solvent which had been used to
clean the apparatus fill the cup with the sample to be tested to the level
indicated by filling marks.
2. Insert the thermometer, light the test flame and adjust it to 4 mm in
diameter.
3.The oil sample is further heated and the rate of about 1 degree centigrade
per minute and application of test flame is done after every 1-degree
centigrade raise in temperature of the oil. Sample gives a clear and distinct
blue flash is reached as the fire point of the oil. Say this as t2 degree
temperature.

Detection of low explosives of chemical/color test and TLC.

Material, Tools & Equipment Required (if any)

Sl.No. Name Specification Qnty.
1. Aluminium foil,
2. plastic or glass jar,
3. aluminium oxide,
4. cellulose or silica gel.

PROCEDURE

PRINCIPLE:
Like other chromatographic techniques thin layer chromatography (TLC)
depends on the separation principle. The compounds in the mobile phase move
over the surface of the stationary phase. The movement occurs in such a way that
the compounds which have a higher affinity to the stationary phase move slowly
while the other compounds travel fast. Therefore, the separation of the mixture is
attained. One completion of the separation process, the individual component from
the mixture appears as spots at receptive levels on the plates. Their character and
nature are identified by suitable detection techniques.

PROCEDURE:
1. The stationary phase that is applied to the plate is made to dry and stabilize.
2. To apply sample spots, thin marks are made at the bottom of the plates with the
help of a pencil.
3. Apply sample solutions to the marked spots.
4. Pour the mobile phase into the TLC chamber and maintain equal humidity, place
a moistened filter paper in the mobile phase.

5. Immerse the plate for the development remember to keep the sample spots well
above the level of the mobile phase. Do not immerse it in the solvent.
6. Wait till the development of the spots, once the spots are developed, take out the
plates and dry them. The sample spots can be observed under the UV light chamber.

To identify the physical examination of paint chips by microscopic method

Material, Tools & Equipment Required (if any)

Sl.No. Name Specification Qnty.
1. Paint chips,
2. slides,
3. core slips,
4. comparison microscope.

PROCEDURE

PRINCIPLE:
The paint as trace evidence is obtained in vehicular cases, hit and
run cases, theft etc. The paint is obtained on tools, structures, vehicles or
from clothing of victims from the crime scene. Paint can be defined as a
material used for the purpose of coating the surfaces. Forensic paint analysis
can be used to investigate damages, scrawling, burglary cases. Paint
residues are generally found on suspects, suspected burglary tools or other
devices used to carry out criminal activity.

PHYSICAL MATCH:
Physical match is the most conclusive type of examination that a
forensic investigator can perform on a paint sample. It involves the comparison
microscope of edges and surfaces between striae between samples. Both the
edges and stria in question must possess unique characteristics.

PROCEDURE:

1. Take the paint chips and two slides and cover them with the cover slips.
2. Keep these under the comparison microscope and observe them.
3. Mark them as "A" and "B".

1
Page No.............
Applied and Action Learning
Observations, Ca cu ations, Graph
1. Both the paint chips are of different pigments.
2. Both of them have different patterns.
3. The paint in "A" is thinner and paint chip "B" is thicker.
4. The paint in "A" slide have designs on it where as the paint on the
slide "B" is plain without any designs.
5. There is a scratch mark on the slide "A" and no scratch mark on the
slide "B".
6. As the paint chips on the slide "B" is thicker than the paint on slide "A" it
can be made of many layers than the other one.

Safely Precaution: 1. As the paint is too small, they must be handled with care.
2. note down the observations carefully
Results:

As there are many differences between the paint chips it can be concluded
that the paint is obtained from different materials.

To identify the physical matching of broken glass pieces.

Material, Tools & Equipment Required (if any)

Sl.No. Name Specification Qnty.

1. Glass pieces,
2. any hard substances for breaking the glass.

PROCEDURE

Glass is an inorganic solid material that is usually transparent or translucent as

well as hard, brittle and impervious to the natural elements. Forensic analysis of
the glass evidence to determine whether they have common source to the
exclusion of all other sources may only be done if they can be physically matched
together. They don't degrade like biological evidence and don't alter over time.
Every glass analysis starts with the identification of the glass by physical and
optical properties such as amorphous structure and isotropism and hardness.
Initial stages of analysis involve physical observation such as color, surface
feature, fluorescence and thickness of the glass sample. Physical examination
plays an important role in the comparison process of glass evidence recovered
from the scene of crime and the questioned sample.

PROCEDURE:

1. Take a glass material and also a hard substance.

2. Beat the glass to a hard substance so that the glass breaks.
3. Now observe the glass pieces and note down the observations.

Page no. 3
Applied and Action Learning

1. The nature of the glass fragments obtained are in the chip forms.
2. All the fragments are of same color and they are so thick.
3. All resemble to be transparent.
4. The texture of all the glass pieces are similar.
5. The fluorescence is absent but they are all in white color which is
colorless.
6. The pigment distribution is all similar.
7. By physical matching we can say that they are made of many layers.

Safely Precaution: 1.Be careful with the glass pieces.

2.Have gloves to your hands before handling with the glass.
Results: .3.Make a well examination so that no criteria is missed

From the above observation we can know that these fragments are made of
same material.

To identify the density of the glass floating method.

Material, Tools & Equipment Required (if any)

Sl.No. Name Specification Qnty.

1. A measuring glass jar,

2. glass pieces,
3. solvents like glycerin & methanol

PROCEDURE

THEORY:

Glass is an inorganic solid material that is usually transparent or

translucent as well as hard, brittle, and impervious to the natural elements.
Glass analysis is applicable to wide range of forensic cases. In burglary or
theft, window pane maybe broken and these glass pieces are analyzed
and in assaults and rape cases glass bottles maybe analyzed. Glass
analysis is also applicable in motor vehicle cases particularly in hit and run
cases, as the glass from the headlights can be analyzed. Density is the
mass of unit volume of a material substance. The formula for density is
d=M/v, where d is the density, M is the mass and v are the volume. Density
is commonly expressed in units of gram per cubic centimeter. The flotation
method of bulk density determination is based on the simple principle: a
sample immersed in a liquid is in a state of mechanical equilibrium, i.e.,
neither sinking now floating occurs, when its density is equal to the liquid.
This state is called as floating equilibrium.

Page no: 5
Applied and Action Learning

PROCEDURE:

1. Take a glass measuring jar.

2. Add both the solvents which we have taken glycerin and methanol in the
same quantity.
3. As they are of different densities these solvents float.
4. Now add our glass pieces in the solvents.
5. Observe whether they float or sink.
When we add the solvent the glycerin sinks and methanol floats. After
adding glass pieces, they float in glycerin and sinks in methanol.

Safely Precaution: 1. Clean the glass measuring jar before you use it.
2. Be careful with the glass pieces.
Results: 3. Take accurate results without any errors

As glycerin is of higher density it sank and methanol is of lower density

it floats on the methanol. Also, the glass is of less dense than the
glycerin and highly denser than the methanol. As a result, their floating
in the glycerin and sinking in methanol.
