The following values have been found for an enzym:

-Vmax = 10 mmol ml-1 min-1
a. At what substrate concentration will the enzyme operate at 25% of its maximal rate
b. What will the rate by at [S]= 0.0025 M (=0.5*Km); [S]= 0.01 M (=2*Km); [S]= 0.02
M (=4*Km)?
Question 2
Two reactions take place in cells that allow the transfer of amino groups from glutamate to a-
keto acids:

Glutamate + pyruvate ⇆ a-ketoglutarate + alanine DG°’ = -1.0 kJ/mol


Glutamate + oxaloacetate ⇆ a-ketoglutarate + aspartate DG°’ = -4.8 kJ/mol

In cells the molar concentrations of the reactants are:

[Pyruvate] = 10-2 M [Aspartate] = 10-2 M


[Alanine] = 10-4 M [Oxaloacetate] = 10-5 M

Does net synthesis of alanine and oxaloacetate from pyruvate and aspartate occur in these
conditions?

Side chain carboxylate of glutamate (pKa 4, so one negative charge at pH 6 and pH8)
Side chain imidazol of histidine (pKa 6, so uncharged at pH8; at pH 6 there are two
populations: 50% of the molecules is uncharged and 50% has one positive charge at
the imidazole group)
The side chain guanidine um group of arginine (pKa 12.5, so one positive charge at
pH 6 and pH8)
The C-terminal carboxylate (pKa 3, so one negative charge at pH 6 and pH8)
