Calibration and Verification of Volumetric Glassware

1. Purpose
To establish a standard procedure for the calibration and verification of volumetric
glassware in compliance with ISO 8655 and ISO 4787 to ensure accuracy and reliability in
laboratory measurements.

2. Scope
This procedure applies to all volumetric glassware (pipettes, burettes, volumetric flasks,
graduated cylinders, dispensers) used in the laboratory for quantitative analysis.

3. Responsibility
- Laboratory personnel are responsible for performing calibration and recording data.

- The Quality Assurance (QA) department ensures adherence to this SOP and reviews
calibration records.

4. Definitions
- **Nominal Volume:** The volume indicated on the glassware.

- **Measured Volume:** The actual volume delivered, determined through calibration.

- **Deviation:** The percentage difference between the measured and nominal volume.

5. Materials and Equipment


- Deionized water (temperature 20°C ± 2°C).

- Analytical balance (sensitivity ±0.1 mg).

- Thermometer (accuracy ±0.1°C).

- Clean and dry volumetric glassware.

- Calibration logbook.
- ISO 4787 water density table.

6. Procedure

6.1 General Guidelines


1. Ensure all glassware is clean and dry before calibration.

2. Perform calibration in a temperature-controlled environment (20°C ± 2°C).

3. Document all readings and calculations in the calibration logbook.

6.2 Calibration Steps for Pipettes


1. **Preparation:**
- Zero the analytical balance with an empty collection vessel.

2. **Filling the Pipette:**


- Fill the pipette with deionized water up to the mark (meniscus level).

- Avoid air bubbles during filling.

3. **Dispensing Water:**
- Dispense the water into the pre-weighed collection vessel.

- Ensure complete drainage according to the pipette type (e.g., 'To Deliver' or 'To
Contain').

4. **Weighing:**
- Weigh the collection vessel with the dispensed water.
- Record the gross weight.

5. **Volume Calculation:**
- Use the formula:

Volume (mL) = Weight (g) / Density (g/mL)

- Refer to ISO 4787 for the density of water at the measured temperature.

6. **Deviation Calculation:**
- Calculate the percentage deviation using the formula:

%Deviation = ((Measured Volume - Nominal Volume) / Nominal Volume) × 100

7. **Acceptance Criteria:**
- The deviation must be within ±0.3% for pipettes or as per manufacturer specifications.

6.3 Calibration Steps for Other Glassware


1. Follow the same procedure as pipettes for burettes, dispensers, and graduated cylinders.
2. For volumetric flasks, fill the flask with water to the calibration mark and weigh the
contents to determine the actual volume.

7. Documentation
1. Record the following data in the calibration logbook:

- Glassware type and nominal volume.


- Gross and net weights.
- Calculated volume and deviation.
- Date and initials of the personnel performing calibration.

2. Retain calibration certificates for future reference.

8. Frequency of Calibration
Perform calibration annually or more frequently if:
- Glassware is heavily used.
- Suspected damage or inaccuracy.
