Exercise 2: Preparation of the standard curve for betacyanin

Once you have obtained the absorbance at 525 nm (A 525; column 6 of
Table 1), draw the standard curve on the graph paper provided. Make sure
you properly label the axes and include the figure number and a figure title.

Exercise 3: Effect of temperature on membrane integrity

Table 2: Absorbance of betacyanin leaked from beet root discs exposed to


different temperatures. Column 3 to 5 are suggested times for the
experiment.

**Using the standard curve and the absorbance values obtained (column 6),
determine the concentration of betacyanin in the water.

Using the graph paper provided, draw a bar graph showing the relationship
between the concentration of betacyanin released and temperature.

1. According to your experiment, which temperature(s) were the most


damaging to the membrane?

2. According to your experiment, which temperature(s) had minimal or no


effect on membranes?

3. How do high temperatures damage membranes? Briefly explain your


answer, incorporating theory of the Fluid Mosaic Model. (Answer
before the lab as part of the lab assignment)

4. How does freezing damage membranes? Briefly explain your answer,


incorporating theory of the Fluid Mosaic Model. (Answer before the
lab as part of the lab assignment)

5. Are your experimental results in accordance with your answers in


questions 3 and 4? Please comment.

6. Throughout the experiment, the spectrophotometer was set at 525
nm. Explain why this wavelength was used.

7. Your TA gives you a solution of chlorophyll, the green pigment found in


plant cells. Can you use the standard curve for betacyanin to
determine the concentration of chlorophyll in this solution? Briefly
explain why or why not.
