1.0 Purpose
1.1 To ensure that seed doesn’t devalue due to the presence of mixtures and impurities by determining
the proportion of pure seed, inert matter and other seeds (weed seed).

2.0 Scope
2.1 This standard operating procedure will run from mixing and dividing of submitted sample to the
reporting of results.

4.0 Definitions
4.1 QAM: Quality Assurance Manager
4.2 HOST: Head of seed testing
4.3 DHOST: Deputy Head of seed testing
4.3 SSU: Seed Services Unit

5.0 Responsibilities and Authority


5.1 QAM
 Supervising and monitoring seed purity testing procedure and staff
 Ensuring that all seed purity testing processes, procedure and reporting are done
according to ISTA standards in order to meet clients needs.
 Ensuring that all equipments required for seed purity test are available and in good
condition.
 Approving seed purity test results.

5.2 HOST
 Supervising seed analysts as described in seed quality manual
 Ensuring that seed analysts are using all relevant documents when conducting seed purity
test.
 Providing leadership guidance to the staff working in the seed testing laboratory.
 Ensuring a safe work environment in the laboratory.
 Production of certificates of analysis to provide timely and accurate results to the clients.

5.3 DHOST
Works hand in hand with the Host by ;
 Supervising seed analysts as described in seed quality manual
 Ensuring that seed analysts are using all relevant documents when conducting seed tests.
 Providing leadership guidance to the staff working in the seed testing laboratory.
 Ensuring a safe work environment in the laboratory.
 Production of certificates of analysis to provide timely and accurate results to the clients.

5.4 Seed Analysts


 Analyse the purity of all seed species submitted to the labolatory through the guidance of
ISTA standards
 Recording purity analysis findings on pre-printed form.
 Making calculation on seed purity results.
 Checking accuracy of required equipment.

6.0 Activities
6.1 Obtaining a working sample.
6.1.1 Ensure that the submitted sample is of appropriate size.
6.1.2 Ensure that the working area is clean.
6.1.3 Ensure appropriate working documents are at hand.
6.1.4 Ensure the equipment used are in good condition and well calibrated
6.1.5 All the necessary equipment required must be gathered for testing as follows
 Seed divider
 Weighing balance (precision and analytical)
 Scalpels
 Petri dishes
 Calculators
 Pens
 Containers
 Magnifiers and working bench lights.

6.1.6 Have knowledge of the prescribed working sample weight for different species.

6.2 Separation of impurities


6.2.1 Understand preliminary knowledge of plant species and seed structure.
6.2.2 Must have the current version of ISTA rules.
6.2.3 Be able to understand and apply ISTA rules on seed testing.
6.2.4 Impurities to be separated are pure seed, inert matter, other crop seed, weed seeds.

6.3 Component identification


6.3.1 Be able to apply the principles of pure seed definition.
6.3.2 Be familiar with the botanic structures of the seed for easy identification.
6.3.3 Be able to use seed identification reference materials like;
 Microscope
 Internet
 Reference books
 Seed identification handbook.
6.3.4 Other seeds, weed seeds, inert matter and pure seed are put in separate petri dishes
6.3.5 The kind of inert matter are recorded on the working card in the appropriate spaces provided
6.3.6 Other seeds are examined and each kind found is identified.
6.3.7 The identified species are reported using botanical names and number of seeds provided.
6.3.8 The components are transferred in separate containers and carried on a tray to avoid dropping

6.4 Weighing of the components


6.4.1 Seed analyst must be weighed in grams to the specific minimum number of decimal places.
6.4.2 Seed analyst must have knowledge of the type of weighing balance depending on size and
sample weight.

6.8 Rounding procedure


6.8.1 Check if all fractions are within tolerance.
6.8.2 If the fractions are within tolerance add the weights together, calculate percentage and round the

6.9 Reporting results


6.9.1 Percentage of all components must add up to 100.
6.9.2 Purity analysis results must be given to one decimal place.
6.9.3 Report all the components that are less than 0.05% as Trace or ‘TR’.
6.9.4 Report the percentage of pure seed, other seed and inert matter in the spaces provided on the
analysis report.
6.9.5 If the results of analysis is nil it must be shown as ‘0.0’
6.9.6 Indicate the scientific name of the specie of pure seed, scientific name of each specie
Of other seed and the kind of inert matter according to the current stabilized plant names.

7.1 Working cards.
7.2 Copies of certificate.
7.3 Equipment calibration data.
