A molecular biologist interested in investigating the DNA concentration of two
fruits namely strawberry and banana. Plan and design an experiment to isolate and
compare the DNA concentration of named fruits.
Hypothesis: The DNA concentration of the strawberry will be determined to be
higher than that of the banana due to its’ cellular characteristics and/or genetic
make up
Aim: To determine the DNA concertation of the strawberry and banana by
comparing the volume of their precipitate
Materials/Apparatus: bananas, strawberries, dishwashing detergent, table salt,
Isopropyl alcohol(chilled in freezer), two ziplock bags, coffee filter, test tube,
spoon,electronic balance , knife , beaker, clean container, graduated cylinder
Method :
1. Ensure all apparatus are thoroughly cleaned before starting the experiment
2. Peel and cut the banana into small pieces using a knife
3. Place individual pieces of cut banana on the electronic balance until a weight
of 7 grams is obtained
4. Discard the remaining slices and place the 7 grams of the fruit into a ziplock
bag
5. Use a beakerto measure 70 ml of water
6. Add a pinch of salt and the 70 ml of water to theziplock bag
7. Use a spoon to mash the fruits in the ziplock bagto a slurry
8. Measure 3ml of dishwashing liquid using the beakerand add it to ziplock bag
9. Close the ziplock bag, shake it rigorously then let it sit for 7 minutes
10.Place the coffee filter over the test tube then pour the liquid from the
ziplock bag into the test tube to separate the liquid from the solid debris
11.Transfer the liquid in the test tube into a clean container
12.Measure 70ml of isopropyl alcohol
13.Slowly layer chilled isopropyl alcohol on top of the liquid then observe the
formation of a white, stringy precipitate ( DNA ) at the interface of the
alcohol and fruit extract
14.Use a spoon to scoop out the precipitate from container and place in a
graduated cylinder to obtain the volume
15.Record the observations made in a table
16.Repeat steps 1 to 14 for the strawberry

Variables:
Controlled – the amount of water, dishwashing liquid and grams used for each
fruit. The time given for each fruit to sit in the ziplockbagsand the amount of
isopropyl alcohol used on each fruit
Independent –the fruits used (banana and strawberry)
Responding – the volume of the precipitate obtained
Precautions:
Ensure the graduated cylinder is read at eye level on a flat surface
Ensure the fruits used were at relative levels of ripeness
Sources of error:
Inadequate amount of water or alcohol being used on each fruit
The precipitate of each fruit was not properly extracted resulting in
inaccurate values
Expected results: The volume of the precipitate obtained from the strawberry will
be greater than that of the banana confirming the proposed hypothesis.
