L2 Cell Parts and Its Structures

This document provides instructions for a biology lab activity to identify cell parts and structures of plant and animal cells under a microscope. Students will prepare slides of onion peel and human cheek cells stained with different dyes to observe and compare plant and animal cell structures. The objectives are to identify cell parts and functions, differentiate between plant and animal cells, and draw and label the cells. Questions at the end ask students to list cell structures and functions, explain the importance of cells, discuss discoveries from cell study, and draw and label plant and animal cells.

Laboratory Activity 2
Cell Parts and It’s Structures

INTRODUCTION

The cell is the basic unit of living things. Cell structure is one of the ways in which organisms differ
from each other. For example, animal cells contain structures that are not found in plant cells. Muscle cells
contain structures not present in blood cells. No matter where a cell comes from, however, there are certain
features common to all, or most, cells. While each part of a cell has its specific role to perform, all work
harmoniously to keep an organism alive.

OBJECTIVES
a. Identify the parts and functions of a typical plant and animal cell.
b. Differentiate between plant and animal cell.
c. Draw a plant and animal cell and label its parts correctly.

MATERIALS
• Compound light microscope
• forceps
• glass slide
• cover slip
• Glycerine
• Safranin stain solution
• Methylene blue stain solution
• watch glass
• clean toothpick
• cheek cell
• onion bulb

PROCEDURES
A. To Prepare Stained Temporary Mount of Onion Peel
1. Take an onion and remove its outermost peel.
2. Cut a small part from an inner scale leaf with the help of a blade.
3. Separate a thin, transparent peel from the convex surface of the scale leaf with the help of forceps.
4. Keep these peels in a watch glass containing water.
5. Add two drops of Safranin stain in the watch glass to stain the peel.
6. Take a clean slide and put a drop of glycerine in the center of the slide.
7. Carefully mount the peel on the slide. Glycerine prevents the peel from drying up.
8. Carefully cover it with a cover slip and avoid any air bubble from entering he coverslip.
9. Remove any excess glycerine with a filter paper.

Cell Parts and Its Functions

10. Observe the prepared mount of the peel under low and high magnification using a compound microscope.
11. Draw and write your observations.

B. To prepare Stained Temporary Mount of Human Cheek Cells

1. Gently scrape the inside of your cheek with a clean toothpick.
2. Smear toothpick onto clean glass slide.
3. Add one drop of Methylene blue stain.
4. Place cover slip over stain.
5. Under the compound microscope, locate and observe your cheek cells under low and high magnification.
6. Draw and write your observations.
Precautions:
1. Over staining should be avoided.
2. Folding of the peel should be avoided.
3. Clean and dry glass slide and coverslip should be used.
4. Coverslip should be put carefully avoiding any air bubbles.
