CAUTION! After any firmware update, run the self test again and make a
new printout.
The DAVINCI QUATTRO is shipped in a wooden crate (1300 W x 1130 D x 1090 H).
It’s weight will be about 200 kg. The crate is palleted. A fork lift or some form of
pallet jack is required to move the crate.

The following steps should be followed and appropriate notes and printouts have to
be attached to the checklist as evidence that the procedures have been followed
during installation. A copy of this record can be made and left with the instrument
after installation.

5. Using a wrench (nº 13 - 1/2”), remove the two bolts from the Removable
Front Support Assembly. Set the Removable Front Support Assembly to the
side.

6. The instrument is ready to be lifted onto the cart or the workbench. Do not
pick up the unit by the outside skins/cover. Hand support the unit from
bottom frame. Slide the instrument out the front side of the crate. Do not pick
unit straight up, it will damage the support legs. Once the unit is out of the
back support area then the unit can be picked up.
7. When the instrument has been installed onto the workbench, remove the
transport protections. Remove the plastic bag from the instrument. Inspect
for any visible damage.
8. Open the blue cover slowly. Pull the pink foam on left side straight out.
9. Remove the accessory box on right side by tilting forward and sliding box
out.
10. Remove the arm pink support and box bottom support.

12. Remove the long white cover support from the front.
13. Using a bubble level, ensure that the system is level on the bench. If
necessary, level can be achieved by loosening the nut (wrench Nº 24) at the
top of the screwed-in legs and turning the legs (wrench nº 16) to make them
shorter or longer. After adjusting the legs, tighten the nuts again.
14. Verify that all modules are in place and secured.
15. Open the accessory box and check the contents (see Sales Package List).
16. Insert the washer module plate carrier into the washer module by securing
the two clamps on the bottom to the two railings that protrude from the rear
of the washer cavity. Try not to bend the railings or stress them too much. It
may be necessary to move the lever arm to the eject position to seat the
plate carrier on the railings.

17. Fit the dispense and aspirate tubing to the wash head. It is easier to attach
the small diameter dispense tubing first, then the larger aspirate tubing.
Mount the washer head onto the two pins on the carrier-arm in the washer.
The wash head is leveled with the screw on the top.
18. Secure the tubing in the associated clips so that they are out of the way and
don’t drag across the tops of plates. Ensure the smaller diameter tubing is
properly inserted into the pinch valve.
19. Rinse the wash buffer bottles with DI water and then fill them about halfway
with DI water. Tighten the lids. Handle the wash buffer bottles carefully.
20. Slide the wash buffer bottles into their slots, fasten the quick-disconnects,
plug in the electrical connections and insert the tubing into the pinch valve.
21. Connect the two waste bottle-tubing fittings. Ensure that the cap is tight.

The computer is delivered with the Time Zone GMT + 01:00.

− Select the proper "Dilution Plate", 1ml dilution plate (deep well plate) or
dilution strip plate (deep well dilution strips) according the customer needs.
− Click OK.

− The following screen appears. The test depth can be selected between
0 and 30.
Use the default setting of 10. Tip types, Tip racks and Target racks
need not be changed.
The settings have no influence on the test. Select “Go”.

− Load 108 sample Tips in Sample Tip Rack 1, 2, 3 and 4 (only 4 tips are
used).
Press the button to continue.
Ensure the tips are correctly picked up. Ensure the tip is centered in the deep well
plates or strips. The tip will not be centered to sample tubes and control vials.

− To start the test, select “System Manual Control” from the “Tools” pull-
down menu.
− Select “Yes” to continue when a warning appears concerning the use of
manual control.
− Click on “Testing” and choose “Sample rack scanning tests”. Select “Do
It”.
− Select any or all of the Sample racks (cycles to complete = 1). Select
“Go”.
The machine will sequentially pick up all racks and move them in front of the vertical
barcode reader. The readings are not displayed and no printout will be generated.
Visually watch and listen for smooth operation of the pick-up of the racks.
The scanned Sample ID’s can also be written to a file by activating the checkbox:

NOTE: The option “Write Engineering Test Results Document” is only active
when logged on in engineering mode.

− Enter the number of times to perform the Cycle Count and select “Go”.
At least 12 cycles should be chosen for a complete run. All modules will
be tested 2 times.

Each cycle involves the movement of the plate from the ambient drawer to another
module and back. Each movement should be smooth (i.e. no jerking, hesitation or
interference). Calibrate the modules if the movement is not smooth.
− Place a plate carrier in the front left drawer position (position 1).
NOTE: If the cover is not closed, an alarm will be generated and the following
error message will be displayed :
To mute this alarm and to continue the testing, select “Recovery Options” -
“Try to move the z-motor to 3240 again”. Select “Recovery Options” and
“Try to move the z-motor to 3240 again” again. After selecting this option twice, the
testing will continue.

both systems (if not, OD's of the verification plate can not be

Ensure the OPSYS plate is installed flat and in the correct orientation in the plate
carrier. Ensure that rows in row A and H are unobstructed and the plate glass is
clean. Make sure the filters are free of dust, oils, scratches and contamination.
Never touch the surface of the optical filter media. Keep the plate away from direct
sunlight.
• Select 96 samples.
• Press the Auto Assign Sample ID button and then press OK.
• Start the run by pressing the green triangle button.
• Press the ‘Fast Forward’ button.
• When the Lot Specific Data and Runtime Variables screen appears, press
the green ‘V’ button.
• When the Load Sample Rack screen appears, press the green ‘V’ button.
• Load the OPSYS calibration plate on the correct drawer position enter a
plate identifier in the entry box and then press the green ‘V’ button
• Check waste containers if necessary empty them and press OK.
• After the run is finished the results file can be found in the following folder
C:\Progam Files\bioMérieux\DAVINCI QUATTRO Program\WFV

When the reader verification test fails, the complete reader module has to be
replaced.
To check the channel to channel variation of the reader, run the C2C protocol.

To be able to calculate the results for the sample and reagent verification, a
reference plate is needed.
• Prepare the reference plate. Use a calibrated pipette to fill a Break-4 plate
with 80 µl low density in row 4-5-6, 160 µl in row 7-8-9 and 240 µl in row
10-11-12. Leave row 1-2-3 empty.

2.5.4.1 Principle
The manufacturer is using special tools (Artel reader, software and liquids) to
calibrate the delivered volumes of the pipette. The procedure takes several hours
and can not be done in the field. However, we encountered that the volume
dispensed by the calibrated pipette is 10 % lower than expected. The manufacturer
is using a calibration method based on water.
For serum, plasma or reagents the volume is a little bit lower (approximately 10 %).
Most of the pipette modules are using the same default calibration factors. These
values can be entered manually. To compensate for the lower volume, the
calibration factors have to be changed. This must be done during the installation of
the instrument. After entering the correct values, a pipette verification procedure
must be performed. When the pipettor verification fails, the calibration factors need
to be adapted and the pipettor verification has to repeated.

2. Enter the coordinates X=4000, Y=1000 and Z=2500 and press the button
“Show XYZ”.
The pipettor will move to this position.
3. Click on "Next".
4. Enter the "Actual Volumes" in the following screens. To do,
− Select the target volume.
− Click on "Edit" and press "OK".
− Then, the "Actual Volume" must be set to ** NOT CALIBRATED **
NOTE: To add or remove volumes, click on the "Add" or "Remove" button.
The choice of the Calibration Liquid is not important.
− When all "Actual Volumes" have been set to "NOT CALIBRATED", click on
"Next".

5. When having entered the previous screens, the following screen appears.
A. Change the "Number of cycles to average each volume" from the default
5 to just 1.
B. Tick the box to "Enter values manually".
C. Untick the box to "Remind the user to open / close the Artel lid".

6. Press “Finish” to continue. Now, you will be requested to enter the following
calibration factors for all sample and reagent volumes in single shot and
multiple shot. This should be done one by one.

We do not use the multiple shot function for sample tips. Therefore, we are not
going to change the default values.
For parallel sampling we are using single shot. The disadvantage is that it takes
some more time. The pipettor is moving with the same tip, two times to the sample
tube.

125 µl is the volume still present in the tip when dispensing 25 µl. The dispensed
volume is measured and entered in the screen e.g. 27.241 µl. The normalized factor
is calculated by the software and displayed on the screen e.g. 25 divided by 27.241
= 0.918.
8. After entering all values, the system will store the values into the EEprom in
the pipette module. Wait. After a few seconds, the main screen appears
again.

CAUTION! Before launching the calibration, make sure that the calibration
factors have been set to default values in the DAVINCI QUATTRO (see
Default parameters setting on page 2-35).

The offset range is from 0 to 100. Negative offset values are not allowed. This can
be critical for the z values. Change the Z home sensor position, if obtaining negative
values.

will appear. After verification or calibration this text will change to
"Pass" or "Fail" or “Calibrated”, depending on the outcome of the
action.
All test results will be logged and may be viewed, printed or saved.
When starting the washer software again, the status will again be
reset to "Not Tested" or “Not calibrated”. This is normal. It does not
mean that the pump has not been calibrated.
4. Select “Calibrate Head Offsets“ from the washer calibration menu. The
following screen appears. Insert the washer alignment fixture with the two
alignment grooves to the top and on the left and press OK.

After calibrating the offsets, we recommend to run the “Verify Head Offsets”.
1. Select “Verify Head Offsets“ from the washer calibration menu. The following
screen appears. Insert the washer alignment fixture upside down and press
OK.
2. The wash head needles must move above the calibration plate without
touching.
When finished the following screen appears. Press Yes or No.
CAUTION! After the new offset values are saved to the EEPROM, the
instrument must be powered off and back on to use those new
values.

The washer will dispense fluid into a low sided vessel (art. code 45688570) and the
operator weighs, or measures the volume of the fluid.
The program converts weight to volume. Two calibration points are used to generate
a linear equation that describes the calibration of a bottle. The calibration values for
a bottle include the pump speed, the slope and intercept of the line describing the
calibration.
When a balance is not available, you can also use a cylinder to measure the
dispensed volume.
The washer module will move the dispense head over to the middle of the fluid
reservoir, dispense, move slightly left and then return to dispense more fluid. All in
all, five fluid dispenses occur for each of the two calibration points in order to
minimize dispense-to-dispense errors.
The dispense speed is fixed at 190. The time that the pinch valve is open is
calibrated.
Each bottle has to be calibrated although only 1 pump is used. Due to a difference in
tubing length between the bottle and the pump, we observed that the dispensed
volume for bottle D is little bit higher (shorter distance). The influence of a different
pinch valve for each location is negligible.

Prerequisite:
− Calibrated gravimetric balance with 0.01g resolution (minimum range:
0.01 - 100.00g) or a 10 ml cylinder and a 50 ml cylinder to measure the
dispensed volumes.
− Deionized water or wash buffer.
− A low-sided vessel capable of containing approximately 50ml of fluid
without spillage (5 vessels are included in the sales package).
All fields must be entered before you can continue. Most information is only useful

To set the gain of the sensor (dispense gain / aspirate gain) for a specific volume,
the system will aspirate and dispense fluid from the first test sample. 5
measurements will be performed starting with a maximum gain of 256. When no
curve can be defined (ADC out of range), the next measurement will be done with
half of the previous gain (128, 64, 32, 16). After 5 measurements the gain for the
aspirate step and the dispense step has been defined. This gain will be modified
(increased or decreased a little bit) so that the curve values are in a specific
range. When after aspirating no curve can be obtained, the sample will not be
dispensed. This can occur when the gain is too high e.g. with first sample.
The system will then aspirate and dispense the required test volume to the
microplate.
When all samples are dispensed into the microplate, the user has
the possibility to mark some samples as invalid. Select the sample
and press the “Mark As Invalid” button. The results of these invalid
samples will be skipped to define the ESP curve.

The XXX represents a number 001, 002, 003, …, etc. The numbers begin
automatically with 001 and increment as files accumulate. The Quattro software will
continue to accumulate these files until they are deleted manually. Each crash event
will produce a pair of files with the same number designation. After the system has
been in use awhile, it is not unusual to see an accumulation of 5-10 pairs of these
files. The service engineer may routinely clear these files if desired. They will not
cause problems if they remain.
Usually, only the last pair of files is necessary. You may check the creation date of
the files and select the pair that was created when the specific crash event occurred.

3.4 DATABASE
This Microsoft SQL Server 2000 Desktop Engine (MSDE 2000) supports several
databases. (SQL = Structured Query Language)
The SQL Server 2000 database is using 2 types of files (secondary data files are not
used):
• Primary data files :
The primary data file is the starting point of the database and points to the
other files in the database. Every database has one primary data file. The
recommended file name extension for primary data files is .mdf.
• Log files :
Log files hold all of the log information used to recover the database.
There must be at least one log file for each database, although there can
be more than one.
The recommended file name extension for log files is .ldf.
During the software installation you will be requested to install the database
software.
The 4 system databases will be installed in the folder C:\ Program Files \ Common
Files \
Dynex Technologies \ Databases \ MSQL$DYNEXLOCAL \ Data. The master
database contains all the default settings for tips, plates, bottles, … and also the
default login account “admin”.

• master
The master database records all of the system level information for a SQL Server system.
It records all login accounts and all system configuration settings. master is the database that
records the existence of all other databases, including the location of the database files.
master records the initialization information for SQL Server.
• tempdb
tempdb holds all temporary tables and temporary stored procedures. It also fills any other
temporary storage needs such as work tables generated by SQL Server. tempdb is a global
resource; the temporary tables and stored procedures for all users connected to the system are
stored there. tempdb is re-created every time SQL Server is started so the system starts with a
clean copy of the database. Because temporary tables and stored procedures are dropped
automatically on disconnect, and no connections are active when the system is shut down,
there is never anything in tempdb to be saved from one session of SQL Server to another.
By default, tempdb autogrows as needed while SQL Server is running. Unlike other databases,
however, it is reset to its initial size each time the database engine is started.
• model
The model database is used as the template for all databases created on a system. When a
CREATE DATABASE statement is issued, the first part of the database is created by copying in
the contents of the model database, then the remainder of the new database is filled with
empty pages. Because tempdb is created every time SQL Server is started, the model
database must always exist on a SQL Server system.
• msdb
The msdb database is used by SQL Server Agent for scheduling alerts and jobs, and recording
operators.

specific setting:

The default settings are managed by the startup code of the QUATTRO software.
Every time the software starts, the default settings are rewritten to the database.
You are able to delete the default settings. But, when restarting the software the
default settings are reloaded.
NOTE: By removing the DAVINCI QUATTRO software or Microsoft SQL
Server Desktop engine both files ; RevelationDSX_Data.mdf and
RevelationDSX_Log.ldf will not be deleted. This database can be
deleted (e.g. when user names and passwords are lost) by following
this procedure :
1. The database service has to be stopped first before you can delete the 2
files.
Select Start - Settings - Control Panel - Administrative Tools - Services -
MSSQL$DYNEXLOCAL. Press the “Stop” button to stop this service.
The service status will change from ‘started’ to ‘stopped’.

the programmed target volume in the assay protocol + the extra fluid that is asked to
the pipette module + some extra fluid to be absolutely on the safe side (13/14 µl).
Therefore, enough fluid will be present in the bioMérieux control bottle if the volume
is loaded that is asked by the application software.

Dual detect
The option Do Dual Detect has been enabled in the Runtime Options.
Dual detect divides the level detection phase into 2 zones, the upper zone and the
lower zone. Dual detect was created for users who use more than 50% of the
volume in the tube. The reason this was done is because the system is a little faster
if it does not need to pull the syringe plunger all the way up to detect liquid in the
upper zone. In case the liquid is accidentally in the lower zone, the syringe will be
pulled up again
If you use less than 50%, you should leave this option unchecked. This is also
faster. The syringe is pulled up completely to perform the liquid detection in one
step. There is enough air inside the syringe to perform a liquid detection on an
almost empty tube.
Dual detect splits the level detection phase into 2 zones, the upper zone of a bottle
and the lower zone.

Bubbles are generated during the liquid detection and the “kiss-off”.
The liquid detection is only performed the first time the sample is aspirated from the
tube. During the detection the system is blowing air into the sample liquid.
The “kiss-off” occurs when a used tip is called to make a new sample aspiration
(when dispensing samples in parallel mode, single shot – multiple shot is not
allowed for samples). It ejects any remaining liquid from inside the tip before
beginning the aspiration. The purpose ofthis operation is to prevent a small droplet
of liquid from flying up and contaminating the spigot. During this process the system
is again blowing air into the sample liquid.
Despite the generation of bubbles in the sample liquid, the R&D study showed that
as the original liquid level is correctly tracked, the tip does not dispense these
bubbles in the wells. Therefore, make sure that the settings of the used sample
tubes are correctly defined in the Tubes Database (Tools - Edit Databases - Tubes
Database) to avoid incorrect liquid tracking. Especially, the internal diameter of the
tube is very critical and needs special attention. A small deviation might be enough
to have incorrect liquid tracking. Most sample tubes are tapered. The lowest
measured internal diameter (mostly just above the U-part) must be used in the tubes
database.
NOTE: It is not possible to permanently modify the bioMérieux predefined
sample tubes and sample tube caddies. After restarting the database,
the original predefined settings will be given. In case one of the
predefined sample tube settings can’t be used, a new sample tube
and a new sample tube caddy have to be created.
2. Controls

3. Reagents
Because reagents are dispensed in multi-shot mode, the pipette will aspirate an
excess of fluid. It will immediately perform a “pre-shot” of the fluid back into the
bottle. It will then go on to dispense multiple aliquots as required, followed by a
“post-shot” of excess liquid back into the bottle. Both the pre- and post-shot
dispense can cause bubbles.
Some reagents with protein or detergents can produce quite a layer of foam that can
interfere with future liquid level detection operations. The bubbles may be seen as
the liquid level with the consequence of dispensing air.
To keep the pipette tip immersed in fluid when it is performing an aspiration, there is
a process in the software that can force a Liquid Level Detection to occur. The
accuracy of dispenses have been defined at 3.3 %. Each time it dispense an aliquot,
it adds 3.3% of that volume to the tally. When the accumulated total reaches a
volume that is 50 % of the fluid volume defined by the tip submersion depth, the
software will force another Liquid Level Detection to make sure the tip remains
submersed to the required depth.
To obtain a safe operation, we have to exclude more than 1 Liquid Level Detection
on the reagent bottle.

Actual measurements are from a table that the plate is resting on (= 0) to the bottom

Dispense height
Default this value is 1 mm above the Top of well height. An optimum value was
found by increasing the Dispense height by 0.6 mm resulting in a height of 16.10
mm.
Aspirate height
Default this value is 1 mm above the bottom of the well and is therefore defined by
Height in Plate Characteristics and Height in Well Characteristics of the Define Plate
dialog box. An optimum value was found by decreasing the Aspirate height by 0.7

Sweep height
Raises the aspiration pins on so that the aspiration pins can be moved back and
forth in the well while the fluid is being aspirated. Default this value is 2 mm above
the bottom of the well and is therefore defined by Height in Plate Characteristics and
Height in Well Characteristics of the Define Plate dialog box.
An optimum value was found by decreasing the Sweep height by 0.7 mm resulting
in a height of 4.90 mm, which equals to a distance of 1.3 mm above the bottom of
the wells
Sweep stroke
Specifies the amount of deflection the aspiration pins move from the center of the
well.
The well diameter of the Break-4 is 6.4 mm. By default this value is 3 mm.
An optimum value was found by decreasing the Sweep stroke to 2.5 mm.
When there are accuspheres in the wells of the microplate and the pipette module
has to dispense small volumes the tip will be too close to the accuspheres, which
can cause splashing. Of the assays that are going to be run on DAVINCI QUATTRO
the following ones contain accuspheres:
− Vironostika HIV Uni-Form II plus O
− Vironostika HIV Uni-Form II Ag/Ab
These assays start with a 100 µl dispense step (sample / control or specimen
diluent).
The application software calculates the target liquid level and then adds 2 mm as
dispense height for the tip.
In the accusphere Microplates of these assays, it has been described that the
maximum diameter of the accuspheres is 4.75 mm. It has been decided that the tip
on the pipette moduleshould dispense also at a height of 3.5 mm above the
accusphere, which is 8.25 mm above the bottom of the well. The maximum diameter
of the accusphere is 4.75 mm and this is 1.65 mm (4.75 - 3.1) larger than the liquid
level in the well for 100 µl. Therefore a plate file must be created with a well height
that is 1.65 mm ( 1.7 mm) smaller. This has been done in Well Characteristics of
the Define Plate dialog box.

When the imaginary height of the well has been decreased it is necessary that the
height of the well is still the same for the washer.
The software has automatically adjusted the Offset of Aspirate height and Sweep
height with 1.7 mm downwards to keep the Actual Aspirate height and Sweep height
respectively 3.90 mm and 4.90 mm. In this way the washer shall use the actual well
height and the pipette module the imaginary well height.
NOTE: It is necessary to create a new name for the second plate file
because else conflicts will occur in the system database of the
application software. When a validated bMx protocol has been run
without accuspheres and afterwards a validated bMx protocol with
accuspheres is loaded, the question appears on the screen if the
operator wants to use the plate file of the database version or the
imported version (protocol that is loaded). When the operator
chooses for the database version the accuspheres will get pierced !

3.8 BACKUP
On regular base the customer can make a backup of the patient results. The OD
results are stored in the folder C:\ Program Files \ bioMérieux \ DAVINCI QUATTRO
Program \ Plates.
The Sample ID’s are stored in the folder C:\ Program Files \ bioMérieux \ DAVINCI
QUATTRO Program \ SID.
The dat files and the sid files can be copied to a CD or DVD. This can be done from
the windows explore screen by dragging all the files to the CD or DVD.
The communication between computer and modules will be logged in a file. When
DAVINCI QUATTRO Software is started a new log file will be created storing all
these commands. The log file is saved in the folder : C:\ Program Files \ bioMérieux
\ DAVINCI QUATTRO Program \ Logs.
This file can be useful to investigate a hardware problem. The description of the
commands can be found in the manual “Appendix Manual Control Codes”. However,
this manual is not up-to-date. Some commands are missing.
Log files can be opened in notepad or workpad. Another possibility is to use the
viewer ViewLog.exe. With the viewer you can select between “view all events” or
“exclude commands”.
The viewer can be found in the folder C:\ Program Files \ bioMérieux \ DAVINCI
QUATTRO Program.

Dual detect divides the level detection phase into 2 zones, the upper zone of a bottle
and the lower zone. This allows taller bottles and tubes to be level detected at any
liquid level whether the bottle is full or near empty. It should always be left on as
turning it off will prevent low volumes from being detected in tall bottles and tubes.
If you use less than 50%, you should leave this option unchecked. This is also
faster.
The syringe is pulled up completely to perform the liquid detection in one step.

horizontal and vertical barcode scanner are set up and the cover lock is tested. The

The print command can be used to print a copy of the initial self test
results (3 pages).

• There is some degree of control over the shake speed used by the
incubators. A set of registry settings may be used to control the shaking. It
is imperative that the end user validates their assays with the final shake
settings selected.

Under rare circumstances the connector on the heater flex cable from the plate
carrier may heat up and eventually singe the board causing temperature related
problems. The plate carrier movement causes the connector to move causing
resistance, which in turn will heat up the connector and eventually fail.
Resolution : Secure the cable to prevent movement of the connector. tions below.
Cut two Foam Tape. Peel the backing off of one of the foam tape strips and place it
along the edge of the slot in the chassis, centered under the heater flex cable. Place
the flex cable against the foam tape and apply the other piece of foam tape directly
on top of the existing piece, securing the flex cable between the foam strips. See
Figure.

Remove the Incubator Module from the system by rotating the two Allen
screws counter clockwise ¼ turn and pulling the module from the system.
Install the new module and perform the plate alignment.
• Position Home sensor
The position home sensor (SP 45688549) is mounted on the base as
shown in the figure and is removed by unscrewing the two M3 x 6mm
Socket Cap screws and lock washers.
All other incubator problems (e.g. damaged heater pads, Incubator PCB, defective
eject sensor, plate carrier, belt, motor, ... ) can only be solved by replacing the
complete incubator module. The defective module must be returned to Dynex for
repair.
Special tools and Dynex Test Software are needed to repair the module.

Lamp PWM : The duty cycle is defined during the self test at 405 nm. This duty
cycle is used for all filters. The 100% light value and dark values are checked
against limits before each plate read to ensure each read is valid. If the 100% value
is out the lamp will be adjusted to bring it back within the limits if possible. If it is not
possible then you will get a lamp related error.

Troubleshooting : Self test fails (bad lamp - focus problem (adjust lamp position))
The duty cycle is an indication for the status of the reader optics. Dust or any other
lightblockage will increase the duty cycle. By sending the command A0LPD, the
value of the duty cycle will be returned. Normal value 85 - maximum value 140.
When the LPD value is 140, a lamp margin failure will be displayed.
LAMP CALIBRATION - This option is responsible for calibrating the lamp duty cycle
for filter position 1 (405 nm). This is accomplished by assigning the lamp duty cycle
an arbitrary number, moving to filter position 1 and adjusting the lamp duty cycle
until the minimum channel reads 170,000 counts above the dark reading and the
maximum channel reads less than 275,000 counts above the dark reading.

− Reader Module
Remove the Reader Module from the system by rotating the two Allen screws
counter clockwise ¼ turn and pulling the module from the system.
The new module will come with the following filters: 405, 450, 490 and 620 nm.
Probably, some filters from the defective module (e.g. 690 nm) must be tranferred to
the new module. Reinstall the module. Enter the correct filter settings in the setup
screen and perform the pipettor plate alignment.

− Fan
• Remove the fan from the optics cube by removing the 4 M3 x 25mm flat
head screws that secure it to the cube (see figure).
• Carefully cut away the RTV around the fan wires and remove the
connector from the wires.
• Pull the fan out of the cube.
• Mount the fan in the optics cube using the four M3 x 25mm flat head
screws and Loctite 242.
• Install the fan to blow into the cube with the wire going out the access hole
on the right. Attach the 2 mm 2-pin connector to fan wires using 2 contact
crimps.
Pin 1 connects to the Black wire and Pin 2 connects to the red wire. The
connector has a small notch that indicates the position of Pin 1. The wires
should be twisted 1 turn per inch before termination. The final length of the
terminated wires should be 5.25 inches.
− Lamp
• Remove the Reader Module from the system by rotating the two Allen
screws counter clockwise ¼ turn and pulling the module from the system.
• Remove the Filter Access panel on the rear of the Reader Module to
expose the optical assembly. The bulb is mounted in the upper portion of
the optical assembly.
• Grasp the lamp with plastic tweezers and pull the lamp out of its
receptacle.

• Grasp the lamp tines of the new lamp with a pair of needle nose pliers and
align the tines with the corresponding holes in the lamp socket as shown
in the figure and firmly insert the lamp into the socket.
NOTE: If you see any fingerprints or foreign material on the lamp, remove
them using a lint-free cloth saturated with methanol.
• Reinstall the Filter Access Panel and reinstall the Reader Module in the
system.
• Run the selftest. When it fails, adjust the lamp position. To adjust the lamp
position a power extension cable might be useful to avoid the reader has
to be installed several times. (the low level command STSa can be used).
− Filters
• Remove the Reader Module from the system by rotating the two Allen
screws counter clockwise ¼ turn and pulling the module from the system.
• Remove the Filter Access Panel on the rear of the Reader Module to
expose the optical assembly. The filters are mounted on the filter wheel as
shown in the figure.
• Locate the filter that is to be removed. Firmly grasp the exterior filter
housing with a pair of needle nose pliers and pull the filter out of the
spring-loaded slot.
Note: The filter 405 nm must be installed on position 1.

• To install a new filter, firmly grasp the exterior filter housing with a pair of
needle nose pliers and push the filter wheel into the spring-loaded slot.
• Reinstall the filter access panel and reinstall the Reader Module in the
system.
• When adding a filter, the new wavelength has also to be entered in the
software. Go to DAVINCI QUATTRO Software - Tools - Configure system
- Absorbance Reader. Enter the new wavelength.
NOTE: The bottom groove of the filter must be firmly seated in the filter
wheel.
If the groove is aligned with the spring on the filter wheel, the filter has
been installed incorrectly and will result in invalid instrument
performance.
All other reader problems can only be solved by replacing the complete reader
module. The defective module must be returned for repair. Special tools and
software
(Dynex test software) are needed to repair this module.
• After the replacement of the PCB, the module needs to be calibrated and
tested using the Dynex Test Software.
• Replacement of the eject sensor requires calibration using Dynex Test
Software.
• Replacement of the optics, fiber, plate carrier, motor, belt needs special
calibration tools and Dynex Test Software.

• The X-Drive Microswitch is mounted on the left plate. To remove the
microswitch, remove the two M2 x 12mm Pozi Panhead screws, which
mount the microswitch to the plate..
− Wash Z Microswitch
• The Z Microswitch is a mounted on the Z-drive assembly in the upper left
corner.
• To replace the Switch, disconnect the two M2 x 12 mm Pozi Pan screws.
• Remove the solder the wires from the switch and re-solder to the new
switch.
• Secure the switch using the two screws removed in Step 1. Push the
switch upwards when tightening the screws.

− Wash head
• The wash head can be removed from the washer by lifting the manifold
and disconnecting the tubes.
• When a new wash head is installed, adjust the horizontal position of the
needles by the screw on top of the manifold.
− Vacuum switch
• The vacuum switch is connected to the left side of the unit as shown in the
figure. To have access to the vacuum switch, remove the reader module.
• To remove the vacuum switch disconnect the silicon tubing from port A on
the vacuum switch, disconnect the wires, and remove the switch.
• Attach the silicon tubing to port “A” on the new vacuum switch.
• Recalibrate the new vacuum switch. During calibration, the waste bottle
must be empty and the washer module with the wash head must be
present.

− Dispense pump
• The dispense pump is connected to the left side of the unit as shown in
the figure.
• Open the left side panel by removing the 4 screws.
• Disconnect the dispense pump from the tubing.
• Disconnect the power connector.
• Reinstall the new pump and calibrate the pump for the 4 bottles.
− Aspirate pump
• The Aspirate Pump Assembly is located at the bottom of the system as
shown in the figure. To remove the pump assembly, unscrew the four M4
x 8 SKT cap screws.
• Disconnect the tubing and remove the pump from the housing.
• When replacing the pump, secure the screws with Loctite 222. Ensure that
the wires and the tubing from the straight barb fitting on the pump are
passed thru the grommet.

Temperature sensing :
Sensor at the rear of the module. But, sensor is calibrated at the middle of the
drawer using the Dynex Test software. That means temp sensor and the PCB can
not be replaced in the field.

firmware are identical. There is a command to set the operating mode

module name will become S0 or S1 depending on the number of
modules present in the system. The command "SOMD" will set your
module to Drawer mode. The module name will become D0 or D1

Remove the Drawer Module from the system by rotating the two Allen screws
counter clockwise ¼ turn and pulling the module from the system.
After installing a new module calibrate the drawer and the plate coordinates.
− Sensors
• Unfasten the sensor by removing the two socket head screws and lock
washers.
• Disconnect the sensor cable from the ambient drawer main circuit board.
• Remove the cable through the access hole, noting the position of the
access hole. Note where the cable is secured with a tie wrap.
• Secure the new sensor into position, using the two socket head screws
and lock washers.
• Route the connector through the access hole noted in Step 2.
• Replace any tie wraps.
• Plug the connector into the PCB connector.
All other drawer problems can only be solved by replacing the complete drawer
module. The defective module must be returned for repair.
Special tools and software (Dynex test software) are needed to repair this module.

6.6 EPOD
This module supplies the power to the system (Power supply), performs the
communication between PC and the various modules (Communication PCB) and
contains the XY Drive PCB.
The workspace calibration is stored on the XY Drive PCB and the module plate
coordinates are stored on the Communication PCB.
Power Supply

The calibration of the power supply has been done by the manufacturer. No
adjustment is necessary at installation.
Each voltage V1, V2 and V3 can be adjusted by a potentiometer. The adjustment is
done without load. The voltage is measured at the power supply.

EPOD
Field repair : The following parts can be replaced : Fan, Power Supply, XY Drive
PCB, Communication PCB and Communication PCB Firmware.
If needed, other parts like capacitor, inlet switch, inlet filter can be ordered after
contacting GCS.
• Remove the line power cord and the communications cable to the
personal computer from the EPOD.
• Remove the drawer module.
• Remove the racks from the right side of the workspace and then remove
the plate on the right rear side of the workspace.
• Push the EPOD from the rear to release it from the back plane board.
Considerable force is required to release the connectors from the socket in
the back plane board.
• Once the EPOD is freed from the back plane board, continue pushing it
toward the front of the unit and angle it up to remove it from the system. It
may be necessary to open the front extension to allow enough room for
the EPOD to be removed.
− Fan
The fan and fan grill are fitted on the EPOD housing.
• The fan is secured using four M3 X 35mm Pan Head screws with four M3
Flat washers and four M3 Nyloc Nuts. The red wire from the fan is
attached to V3 positive (+) and the black wire from the fan is attached to
V3 negative (-) on the power supply.
• When reinstalling the fan, feed the wires of the fan through 175mm of
Shrink Wrap Jacket.

− Power supply
• Open the EPOD assy
• Remove the four M4 X 6mm screws and four flat head washers that attach
the power supply to the enclosure.
• Remove the Power Supply Unit (PSU) filter harness to the AC input
terminals
(see figure) of the power supply as follows :
− Blue wire ring terminal to the Neutral terminal on the PSU
− Brown wire ring terminal to the Live terminal on the PSU
− Green/yellow ring terminal to Earth terminal on the PSU

• Remove the backplane harness and PSU harness from the DC output
terminals of the Power Supply Unit (see figure).
• To reinstall a new power supply, connect the Back Plane harness, PSU
harness and DC output terminals as follows :
1. Attach two double red wires of harness to V1 Positive (1 ring terminal to each
screw).
2. Attach black wires of harness to V1 Negative (double wires on the left and
single wire on the right).
3. Attach double orange wires of harness with the short blue harness, and
yellow wire end of harness to V2 Positive.
4. Attach double grey wires of harness and small lug end of blue wire to V2
Negative.
5. Attach double brown wires of harness and red wire from fan to V3 Positive.
6. Attach other end of the short blue harness and black wire from fan to V3
Negative.
7. The single yellow wire from the backplane harness and the free end of the
yellow arness with the diode should be attached to the positive (+) terminal
of the capacitor. When the backplane harness is installed, make certain that
it is secured in a manner so that is not in contact with Crystal (X1) on the
Communications PCB.
8. The free end of the single blue wire harness with the large lug should be
attached to the negative (-) terminal of the capacitor. Use two terminal
screws and two lock washers supplied with the capacitor (the lugs are placed
on top of the lock washers, tighten to 12 in/lbs with a torque driver.
• Place the PSU in the enclosure and use the four M4 flat washers and four
M4 x 6 button head screws to attach the PSU to the body. Use Loctite 222
to secure the screws.
NOTE: Before closing the Power Supply Unit, make certain that the ground
wires from the filter, the power input and the power supply filter
harness are firmly connected to the ground.

• XY Drive PCB
The XY Drive PCB is used in the movement of the X motor and the Y motor
(either motor can be driven individually or both can be driven together). The board
is mounted on top of the communications board as shown in the figure.
• Communication Board
The Communication PCB receives information from the host computer and transmits
it to the various devices on the DAVINCI QUATTRO (e.g. the incubator).
In addition, the communication board receives information from each of the various
devices and transmits it to the host computer.

The pipette module must be aligned on site when replacing the pipette
module, by loosening the 2 adjustment screws, accessed through the
controller board.
To align the pipette to the base plate, the Pipette alignment fixture (45688566) is
necessary. The Pipette base plate should be fitted on the jig as shown in the figure.
The Pipette alignment fixture should be positioned on the workspace in the reagent
rack location, using the fool-proofing.

Pipette Module
• Gently move the X arm, Y arm, and the pipette until the pipette is
inserted flat inside the jig. The pipette base plate should fit perfectly in
the jig before proceeding to the next step.
• Tighten the two clamps to secure the pipette firmly to the jig.
• Remove each of the two adjustment screws (one at a time), then re-
install and tighten each screw using Loctite 222.
• Release the clamps, and ensure that the pipette bottom plate is
parallel to the fixture. If the pipette bottom plate is not parallel to the
fixture, readjust as described above.
• Remove the fixture from the system.
After installation of a new pipette assembly: align the pipette (see above), perform the
pipette collar calibration, the E-sensor calibration, the workspace calibration, the drawer
calibration, the plate alignments, enter the volume calibration factors and the ESP
curves (if used). Repeat the pipettor verification.

• Remove the eight M5 X 10mm BHCS screws that attach the back
cover to the system.
• Disconnect the fan cables from J25 and J26 on the back plane of the
system while removing the cover.

Removal of the system covers
• Place the cover assembly in the open position (the gas springs are fully
extended in this position).
• Remove the three M8 X 25mm screws that connect the cover to the hinge
assembly on each side of the cover.
• Spread the cover assembly slightly to clear the hinge assemblies, then lift
the cover off of the unit (2 persons are needed).
• Hold the cover in the open position while spreading the cover assembly
slightly to clear the hinge assemblies.
• On one side of the cover assembly, align the Outer Cover Disc with the
holes in the cover. Loosely install three M8 X 25mm BHCS screws and
three M8 flat washers. Repeat for the other side.
• Gently push the cover up as high as it will go. Hold in this position while
tightening the six screws.
• If there is a gap between the cover assembly and the front extrusion when
the cover is closed, repeat Step 3, then check again. Ensure that the cover
lock operates properly after the installation.

The tension of a belt can be adjusted by using a tension meter (example: Brecoflex
Model SM4). However, some experience to measure the belt tension is required.
Get the average by measuring the tension 10 times.
Specifications :
X = 25 ± 1 Hz
The tension is measured in the middle of the non-carriage side of the belt.
The position of the arm can be either end (completely left or right).

• Remove the lower cover for the Z-Drive (refer to the Replacing of the
Pipette).
• Remove the 4 M 3 x 30 flat head screws that attach the cover to the
mechanism.
• Remove the two M3 x 12 Posi screws.
NOTE: When reassembling the Spring Assembly housing, rotate the cover
eight times counter-clockwise before attaching the screws.
CAUTION! When removing the spring from the housing, be aware that the
spring will expand rapidly and may cause injury.
NOTE: When reinstalling the spring assembly, make sure that the eye lines
up with the groove in the shaft and take care to ensure that the spring
remains inside the housing.
The Z-Drive Assembly
• Carefully remove the Rotary Spring and place a new one on the
housing as shown in the figure, allowing the retainer to slide off while
pushing the spring into the housing fully. Discard the retainer.

XYZ drive
• Move the Z drive block to the end of the rail closest to the motor.
Ensure it remains there while winding the spring.
• Place the spring assembly onto the Z assembly, ensuring that the eye
lines up with the slot in the Z idler shaft and being careful not to push
the spring out of the housing. Orient the housing so that the two screw
holes on the side of the housing are facing towards the end of the Z
assembly extrusion.
• Attach the cover to the housing using two M3x12 flat head screws.
• Rotate the spring assembly eight complete revolutions counter-
clockwise
(as observed from the spring assembly). Secure the assembly to the
extrusion using four M3x30 flat head screws.

− Motors
X-Motor
• To have access to the X-Axis motor, remove the top cover, the rear
cover, the blue system cover and the right side cover of the system
(see procedure 6.8).
• Remove the cover of the EMC box (X transition board) - 4 screws.
• Disconnect the X-motor connector J1 from the X-transition board.
• Remove the three M4 x 16 SKT cap screws that attach the motor to
the motor mount. The motor axis is positioned ± 7 mm inside the
bearing (under the pulley). The motor can be lifted by moving the
motor from left to right and puling.
After a while the motor axis is coming outside the bearing.
• Remove the X-belt from the pulley and remove the motor from the
system.
• Remove the X-motor Pulley from the old motor by removing the 2
setscrews. You need some force to remove the pulley from the axis.
Be careful not to damage the pulley.
• Fit the pulley to the new X-motor using new M4X6 Setscrews and
Loctite 222. Discard the old setscrews. The distance between the
pulley and the motor base must be 3.8 mm.
• Reinstall the X-belt and the motor. Secure the screws using Loctite
222. Ensure that the motor wires are facing to the rear of the system.
• Connect the X-motor connector J1 to the X-transition board and close
the EMC box.
• Reinstall the covers.

Y-Motor
• To be able to have access to the Y-motor, remove the top cover and
the rear cover of the system (see procedure 7.8).
• The Y-Axis motor is attached to the Y-Axis extrusion via two M3 x 8
mm cap head screws and two M3 x 10 mm cap head screws as shown
in the figure.
Remove the 4 screws.
• Disconnect the connector from the encoder.
• Remove the cover of the EMC box (Y transition board) - 3 screws.
• Disconnect the Y-motor connector from the Y-transition board.
• Remove the Y-belt from the pulley and remove the motor from the
system.
• Remove the encoder from the old motor and place it on the new motor
(See procedure). Ensure that the connector is facing to the right of the
system.
• Remove the Y-motor Pulley from the old motor.
• Fit the pulley to the new Y-motor using 2 new M4X6 Setscrew and
Loctite 222. Discard the old setscrews. The distance between the
pulley and the motor base must be 3.2 mm.
• Reinstall the Y-belt and the motor. Secure the screws using Loctite
222.
Ensure that the motor wires are facing to the left of the system.
• Connect the Y-motor connector to the Y-transition board and close the
EMC box.
• Connect the encoder.
• Reinstall the cover

• To have access to the motor, remove the 3 z-drive covers (top, mid
and bottom).
• Disconnect the flexi cable from the top of the Z-transition board. Be
careful not to damage the connector.
• To remove the Z-motor assembly, unscrew the four M3 x 30 Flat head
screws.
• Disconnect the Z-motor connector from the Z transition board.
• Remove the Z-belt from the pulley and remove the motor from the
system.
• Remove the Z-motor Pulley from the old motor by removing the 2
setscrews.
• Fit the pulley to the new Z-motor using new M4X6 Setscrews and
Loctite 222. Discard the old setscrews. The distance between the
pulley and the motor base must be 2.2 mm.
• Reinstall the Z-belt.
• When reattaching the motor, use Loctite 222 on the screws and ensure
that the wires are facing to the bottom of the system.
• Connect the motor connector to the Z-transition board.
• Connect the flexi cable to the top of the Z-transition board.
• Reinstall the covers.

NOTE: The Drive Motor Pulleys and Idler Pulley must be properly positioned
on their respective shafts. Otherwise, the X-, Y-, or Z- Drive Belt may
contact the edge of one or both pulleys during operation.
• Observe the Drive Motor Pulley while manually moving the drive back and
forth along the drive axis.
• If the drive belt contacts the edge of the Drive Motor Pulley while the drive
is moved back and forth, stop at the point where the drive belt begins to
contact the edge of the pulley. Then, loosen the two M4 x 6mm cone
screws that secure the Drive Motor Pulley to the shaft, slide the pulley 2
mm away from the edge of the belt, and retighten the two screws.
• Perform an Arm self test by selecting “DSX Manual Control” from the
Tools menu, responding “Yes” to the prompt, and then selecting “Arm” in
the left panel and “Self-Test the XY” or “Self-Test the Z” (as appropriate) in
the right panel. Then, select “Do It” to begin the test. During the test,
observe the drive belt to determine if it rides near the center of the pulley
through its travel.
• Repeat steps 2 and 3 until the belt rides near the center of the pulley. If
the results are satisfactory, replace the M4 x 6mm cone screws with new
screws one at a time, replacing and tightening the first screw before the
second is replaced. Use Loctite 222 on the new M4 x 6mm cone screws.
• Repeat Steps 1 through 4 for the Idler Pulley.

sink on the underside of the X-transition PCB to the right frame panel.

the standoffs.

button head screws.
• When you replace the board, slide insulation between the motor driver
IC plates and the Y extrusion and secure the IC mounting plates to the
extrusion and assemble in reverse order.

The Y Opto interrupter is mounted as shown in the figure. To remove the Y sensor,
remove the Y-drive cover and remove the two screws. When you replace the
sensor, make certain that the Y-Flag travels through the center of the sensor.

The Z Opto interrupter Assembly is mounted on the Z Sensor Plate.
To remove the sensor assembly, remove the Z drive covers, remove the two M3X6
Cap Head Screws and two M3 Shakeproof washers. When you replace the
assembly, ensure that the Z sensor Flag passes through the middle of the sensor.

− Remove the top cover and the rear cover to have access to the fans (see

− When reinstalling the fan, it should be installed so that the air is forced out of the
interior of the system and the wires are toward the rear.

Other parts
The interlock switch is used to prevent operation of the machine unless the cover is
closed or the override key is inserted. The switch is located adjacent to the right
hinge.
− Remove the top cover, the rear cover, the blue system cover and
the right side panel (see procedure 6.8).
− Remove the harness terminals after noting their location.
− Depress the mounting tabs on the sides of the switch to remove it
from the bracket.
These parts are not available as regular spare part. Contact GCS for more
information when these parts need to be replaced. To replace the gas damping
spring a special tool is needed.
Procedure :
Install the spring first on the lower mounting screw and screw the nut a little bit so
that the spring can’t fall down anymore. Now you have to compress the spring so
that it can be installed on the upper mounting screw. By using the washer calibration
tool plate it’s a little bit easier to compress the spring and to put it on the screw. Be
careful, do not bend the spring. If the spring is in the correct position, tighten both
nuts.

The workspace calibration will also set the 3 motor encoders to zero. That means
that the workspace calibration has influence on the drawer calibration and the
module plate calibration.The coordinates of the drawer calibration and the plate
calibrations are related to the zero position. A new workspace calibration can result
in a new zero position. So, it’s recommended to repeat the Drawer calibration and to
check at least the module plate calibration. Especially, a deviation in the Y
coordinate can result in plate pickup failures.
The calibrated coordinates are stored on the XY PCB in the EPOD (not on the
computer).
The workspace file (bioMérieux Workspace.wsf) contains all the coordinates of the
sample / reagent / control positions, the sample rack pick up positions, the sample /
reagent tip positions, the deep well plate positions in relation to expected workspace
calibration settings.
This workspace file is defined on a “golden standard instrument” and is the same for
all the instruments.
When the computer has to send a command to the pipettor module to move to a
certain position, the computer will use the correction factor to adapt the fixed
coordinates from the workspace file.

Calibration Jig (part number WAFIX-003) onto the workspace at the left-hand
end. Make sure that the holes and pins align. The pillars on the plate should

It is performed if a new drawer module is installed, if the tip does not center

eject and the arm will position itself over the drawer. The program will prompt

to ensure proper positioning. The program will prompt you to “Please move
the arm to the front left drawer calibration hole and click OK when done”.
5. Manually move the arm and pipette to the hole of the calibration fixture.
When moving the arm you should always grasp the points at which the arm
and pipette are secured to prevent bending / flexing of the arms. You should
also be cautious not to bump or move the drawer out of its proper position.
Carefully position the tip of the pipette spigot until it fits into the hole of the
Calibration Plate. While holding the pipette in the calibration hole, press OK
when complete. The program will prompt you to “Please remove the
Calibration Plate from the Front left drawer”. Press OK when done.
The drawer will move to the in position.
The plate coordinates are saved on the Communication PCB in the EPOD
(not on the computer).

3. Select “Get Plate” while observing the positioning of the pipette plate clamp
in relation to the carrier. The pipette plate clamp area should pass freely and
evenly into the carrier in each X, Y and Z-axis. If the pipette clamp is not
properly positioned the coordinates can manually be changed by highlighting
the coordinate value in the manual adjustment dialog box and increasing or
decreasing it to realign the pipette clamp. The process may take several
attempts until the optimal results are achieved.

The values in the manual adjustment Dialog Box for the three axes represent
approximately 0.1 mm in travel. The exact Y & Z step count multiplication factor is
0.0625 and the X is 0.0600. The home position for the X is the farthest left position
or 0, the Y is farthest inward position or zero and the Z is the up most position or
zero.
For example the value reported in the figure for Z is equivalent to the step count
(3560), multiplied by 0.0625 would provide the distance in millimeters from the Z
home position
(= dividing the number of motor steps by 16).
Depending on the particular situation you can either increase or decrease the
number to achieve the optimal position. Try small increases or decreases around 5
until you gain a thorough understanding.
After calibrating the workspace, drawer or modules perform the Cycle Test
and the Tip Check Test.

This calibration is done by the pump manufacturer. It needs not to be done in the
field. However, when the dispense pump can not prime empty lines, the pump can
be calibrated according to this procedure.

To prime the line if the dispense pump is not working at all, you can
connect a large syringe to the outlet of the dispense pump. Using the
syringe, you can support the dispense pump in sucking up the liquid
from the container.
4. When the system is primed properly, ensure the manometer and the tubing
are connected according the diagram above. Turn the dispense pump on
using the command W0DPCA250. The dispense pump will run continuously
with maximum speed. The pinch valve at the wash head will stay closed. The
pressure inside the lines is monitored by the manometer.
5. The PSI reading should be between 3.5 and 4.235 PSI. If it needs adjusting,
see instructions below on how to change the pressure setting.
6. Turn the dispense pump off by sending the command W0DPCA000.
Do not leave the pump on for a long time.
7. Reinstall the dispense pump mount into the left side enclosure. Reinstall the
left panel.

The results are not displayed.
If an error occurs during scanning, you will get a System Sample ID
Error. Verify that the barcode is facing the SID and then select
Recovery Options and choose Try to Rescan this Barcode. You
may also choose the Move This Barcode Slightly and Try to
Rescan. If the error persists, select Recovery Option and choose
Skip Reading This Barcode and continue.
The system has a search algorithm that will move the rack back and forth to locate
the barcode. If all sample barcodes cannot be read verify if the type of barcode label
is enabled. If using a correct barcode label then an adjustment must be made.

For the plate barcode scanner the default settings can be used.
In case the settings are not giving the correct readings, perform the following
adjustments.
b) Record the default Sample Rack Scan Position, Plate Scan Position and L/R Pos
Offset numbers. Next make any adjustment in either X, Y or Z.
− By decreasing the Y value the beam length in the Vertical direction
will decrease, by increasing the beam it will lengthen and loose
focus.
− Decreasing the Z value will raise the reading position of the rack,
increasing the number will lower it.
− Increasing the X value will move the rack to the right, decreasing it
will move it towards the left.
In most cases the Y and Z values for the sample barcode scanner will provide the
broadest coverage and an adjustment in the X will be all that’s needed.
The Plate Scan position is adjusted in the same manner with the likely adjustment
being in the Z direction. The L/R position offset determines the number of steps to
move off center to read plate barcodes that are placed to the left or right.

The pipetting system of the DAVINCI QUATTRO instrument includes ESP software
for automatic detection of clots, foam, bubbles or low volume when pipetting
samples.
The detection is done with a pressure sensor, which gives an analogue signal
dependant upon the pressure inside the barrel of the pipette.
The pipetting signature observed when pipetting the sample, is compared to nominal
pipetting signatures obtained during system configuration. If the pipetting signatures
does not fall within the normal range, the system reports an error.
The emphasis has been put on identifying good valid samples rather than bad
samples.
A sample may fail but nothing has been done to identify why it has failed.
The minimum sample volume for ESP is 5 µl. However, a fail rate of 10 % has been
observed for 10 µl. For the customer it might be not convnient to use ESP with
volumes lower than 50 µl.
ESP will report a volume failure when the volume is 9% to 13.55% lower. There is
no overlap in curves for 50 µl and 100 µl.
The pressure curve depends on tubing / sensor / type of sample : serum - plasma /
volume / viscosity of the sample / dispense profile (use always 4).

ESP (Electronic Signature Pipetting)
ESP description
Assuming that only the sample volume changes, A and B will change. This will bring
an obvious change in the area under the curve. This shape is modified with an
oscillatory waveform caused by the spring damper effect of the air column. The
shape and probably area will be modified further by any transients caused by
bubbles or clots interfering with the flow of medium into the pipette tip. Given that
the shape and area for a given sample size are known then any changes from this
ideal pattern can be detected.
The following information is sent by the pipette module and are used to define
several algorithms to evaluate the sample :
Area : The pipette pressure is measured 3 times per µl of sample. The area under
the curve, the integral, can be calculated. In order to remove any contamination of
the result by any bias of the signal towards positive or negative pressure (an offset),
the first pressure reading is subtracted from all subsequent ones. The area value of
7 digits is sent by the pipette module.
Curve Length : This is the perimeter length of the curve (6 digits). It can easily be
calculated by adding the absolute differences between consecutive pressure
readings.
Area Peak : For a curve there is a maximum pressure point theoretically as the
deceleration ramp starts. Any bias has been removed. (3 digits)
Pressure before air gap (4 digits) and pressure after the air gap (4 digits) is also
defined during the aspirate step.
The final column pressure (4 digits) is the first pressure before dispensing.
NOTE: When viewing the ESP detection data, the “area length” is also
displayed.
This value is a calculation of the Area divided by the curve length.
This value is not sent by the pipette module. The ratio varies
proportionally to the shape of the area.

ESP (Electronic Signature Pipetting)
ESP description
The signal from the pressure sensor as received by the ADC is from 0 to 5 volts.
Where 2.5 volts represents zero pressure, while those above and below represent
positive and negative pressure respectively. The output from the pressure sensor
has been filtered, amplified and level changed to achieve this. The gain can be
dynamically changed under software control. The gain depends on the volume and
is defined during the ESP calibration. The gain will be lower when the volume is
higher. The aspirate needs a higher gain than the dispense.
The ADC gives a 10 bit output allowing values from 0 to 1023. The half (511)
represents zero pressure. In order to protect the pressure sensor from excess
internal pressure, the pipette is stopped in its movement if the ADC output exceeds
1000 or reduces to 0.
An ESP error (e.g. clot) might result that the sample (+ clot) will be dispensed back
into the sample tube rather than risk dragging the clot all around the workspace.

0276 = Motor steps, position of P motor.
0005078 = The summation of all pressure readings during the aspirate. The pressure

• In the “Select Module to Upgrade” pull down list, select the module to be
updated, among the bold ones below :

Firmware troubleshooting
When downloading firmware to a module, you have always to select the correct
module. However, by mistake it can be possible that the wrong module was
selected.
e.g. drawer firmware was downloaded to the reader
This is not a problem. In this case you can always download the reader firmware to
the reader again.
But, be careful. When the wrong firmware was downloaded to the Communication
PCB, the communication between the instrument and computer will be lost after the
download. Downloading firmware is no longer possible.
1. You can order the IC, firmware for the communication PCB – spare part
reference 45688584
2. Cheaper and faster, use a second instrument and downloading the
communication firmware to one of the other modules. Preferable, choose
incubator 1 (= I0).
Select the communication firmware file and select "port 10 - I0" as module
to upgrade.
After the download, remove the IC firmware component from the incubator
PCB.
Install it on the communication PCB.
Change the label.

Firmware troubleshooting
Afterwards, you have to install the IC coming from the communication PCB
(instrument nº 1) into the Incubator module (instrument nº 2) and you have to
download the incubator firmware to this module. The incubator module will not be
recognized by the flash program when selecting the “MAP” button. But, this
unrecognized module is located on Port 10. This is the normal location for incubator
1 (= I0). To program the incubator firmware, select port 10.
NOTE: Remember when one of the incubators is not recognized e.g. in this
case incubator 1, incubator 2 becomes I0, incubator 3 becomes I1
and incubator 4 becomes I2. When pressing the MAP button, you will
found I0, I1 and I2.
Troubleshooting : You can use also this method when you can not download the
firmware to a module for any reason.
e.g. downloading new firmware to the washer is not possible
download the washer firmware to incubator 1 and replace the IC’s

8. Inspect the Pipette Collar Mechanism for wear and debris between Tip Body
and Clamp Body. Clean the Pipette collar Mechanism using the cotton-tipped
applicators and isopropyl alcohol. Do not apply alcohol to the lead screws of
the two motors.
To clean the tip body, move the E motor down by means of the following command :

9. Clean the dispense tip. If dirty, use a small cleaning wire to clean it out.
10. Reinstall the covers for the X, Y and Z drives and the Pipette module. Install
the Z-arm middle cover first, ensuring the ferrite core is still next to the
connector at the top of the Z-transition PCB when the cover is in place.
11. Remove and insert all modules again to have good contacts.
12. Inspect the spring clips of the 4 microplate holders for damage. Replace,
repair as necessary.

13. Remove the 8 way wash head, submerge it in 70% or greater Isopropyl
alcohol and clean both aspirate and dispense pins with cleaning wires. Then
remove the four cleaning port setscrews on the wash head. Flush with DI
water. When finished, install the four setscrews. Do not over-torque these
screws because the wash head can be cracked easily.

14. Using the tubing diagrams on the following pages, assemble the replacement
tubing for both dispense and aspirate tubing. Leave the 4 reducer fittings, 42
mm PVC tubing and elbow fittings off the dispense lines until assembled in
the front extrusion.

15. Using the newly-assembled tubing, replace the existing tubing and reconnect
it exactly as removed. Removing the Washer module will help facilitate the
replacement of the vacuum switch tubing. Use existing clips to secure tubing
and secure it is not pinched or compressed in any way that would impede
flow. Replace any unsecured or damaged clips. Reinstall the Washer module
if removed. Reinstall all panels and ensure no tubes are pinched off.

1. Replace the tubing and related parts using the kit and the diagram below.
2. For units with a vacuum trap, do the following:
a. Cut the 300 mm Tubing (T1) to 220 mm.
b. Switch the positions of the 220 mm Tubing (T1) and the 650 mm Tubing (T2).
c. Connect the Vacuum Trap in the position of the Straight Fitting (F1).
d. Discard the Straight Fitting (F1).

• Switch off the instrument. Disconnect the manometer and multimeter and
reassemble the tubing and sensor back into the system. Visually check
that the vacuum sensor is tightly secured to the sidewall. Check for kinks
or pinching of the tubing.
25. Remove sample tubes and the control rack otherwise the tips might hit the
tubes and bottles during the next test. Select “Tools”, “System Manual
Controls”, “Testing”, and “Tip Alignment Test” and click “Do It”. Observe the
movement and ensure the tip is properly positioned into each area. A
workspace calibration needs to be done if there is a problem with the tip
pickup. Then, run the Tip Alignment test again. Exit the program. (if a
workspace calibration is performed, the drawer calibration must be repeated
and all module coordinates must be rechecked)

26. Run the WASHDISP assay to fill a microtitre plate with 300 µl DI water to run
the Plate Movement test. Check the dispensed volume. Clean the wash head
if necessary.
27. Select “Tools”, “System Manual Controls”, “Testing”, and “Plate
Movement”.Click “Do It”. Enter cycle count of 12 and place a carrier with the
filled microtitre plate into the #1 ambient drawer position. While running the
test, watch the plate pick-up at each position. If necessary, choose “Tools”,
“Configure System” and manually adjust the module for proper pick-up.
28. Perform the Instrument qualification.
NOTE: During the verification, check for leaks in the installed dispense
tubing. Check and make sure that the head is not leaking from the
cleaning ports.
29. Reinstall the cover lock key and ensure the lock works. From the software
select “Tools”, “System Manual Controls”, “Arm”, “Lock cover” and “unlock
cover”. If it is not working properly adjust the cover position. Loose the 6
screws of the cover and push the cover up. Then, tighten the 6 screws.
Repeat the test.
30. Remove all consumables used for the test and clean the workspace.
31. Delete all the log files from the folder

PROCEDURE
The system is constructed from materials that are resistant to chemical attack.
Spills should be cleaned up as soon as possible. The service engineer should clean and
decontaminate the system before servicing.
Only the following modules can be returned for repair ; reader, drawer, barcode scanner,
incubator, pipette and washer. The IVT and OPSYS tools must be returned for
recalibration on a regular basis.
Fill out the form CFO0142 in use and send this document to bioMérieux
France, Global Supply Chain to receive the bioMérieux Return
Authorization Reference and a new module. The correct fax number can
be found on the document. The fax number can be found on the
document.
− For bioMérieux subsidiaries there is no warranty.
− For distributors there is a warranty period of 1 Year.
After receiving the CFO0142 form Logistics France will send you a “new
module” from stock. Keep in mind that the modules on stock might be
looking as “being used”. There is no difference between new manufactured
modules and repaired modules.
After receiving the Return Authorization Reference from France, fill out the
"Repair Sheet” (see next pages). Modules without such a form or without a
decontamination certificate will not be accepted !! (Do not forget to indicate
the bioMérieux Return Authorization Reference. Leave the Dynex Return
Authorization Number blank.)
Indicate the fault condition clearly on this paper.
Send the repair sheet to Global Supply Chain, bioMérieux France.
Send the defective module to the bioMérieux central warehouse.
NOTE: This procedure means that the defective module will immediately be
replaced by another module. You have not to wait until the module has
been repaired by Dynex.

Cleaning
IMPORTANT! Due to some local custom rules in some countries (e.g. Brazil) a module
can only be sent for repair without high export costs, if the same module
(same serial number) is returned after repair. These countries have to wait ±
2 months until the module has been repaired. For the pipette assembly it
will take even 4 months. So, these countries have to organize a larger local
stock to service the instruments.
12.2 CLEANING
Clean all external surfaces with a cloth moistened with a mild laboratory detergent. If
necessary, dilute the detergent according to the manufacturer’s instructions before using.
12.3 DECONTAMINATION
Wipe all surfaces, plate carriers, pipette spigot, module plate carriers with a cloth
moistened with a disinfectant cleaner (e.g. Mucasol or Extran) or a 70 % (by volume)
solution of Isopropyl alcohol.
The manifold of the washer needs to be decontaminated very well. The manifold can be
soaked in a bottle of disinfectant cleaner (e.g. Mucasol or Extran) for 30 minutes.

13.1 PRINCIPLE
The LIS - Link software (version 1.06) based on the ASTM standard, is included with
the instrument. The software and documentation can also be downloaded from the
GCS Portal.

Once the installation and the configuration are complete follow these procedures to
process test runs.
NOTE: Make sure scanning and using LIS are selected during the worklist
creation.
2. Press OK. Select the last sample and Load the Sample Caddy. Then, scan
the sample ID’s.

3. After scanning the sample IDs, a query will be sent to the HOST. The HOST
will send the joblist.
4. After the Quattro has received the joblist, automatically the next screen
appears. Press “New Plate” and select the assay(s) to be run. Automatically,
the samples will be flagged for the correct assays.
5. Press OK to start the run. Perform required operations, loading fluids, etc…
6. Allow the test runs to go to completion.
7. Review the plate data.

9. Click on the name of the assay that was run and review all results returned
by the QUATTRO software.
NOTE: Make sure Send is selected for the results to be transmitted to the
LIS host computer.

10. After the review is complete, transmit the results to the host computer.
