YEAST EXTRACT MEDIA
➢ PREPARATION OF MEDIA
REQUIREMENTS
• Conical flasks
• Yeast extract
• Agar
• Glass rod
• Distilled water
• Hot plate
• Cotton
PROCEDURE
1. First take 1000 ml of distilled water in the conical flask
2. Add 23 gm of yeast extract in the water.
3. Add 7 gm of agar agar into the flask
4. Now heat the flask on the hot plate until yeast extract and agar agar gets
dissolved in the water
5. After complete boiling ,sterilize media by autoclaving at 15lbs (121℃) for
15 min
LEAF DISC METHOD
REQUIREMENTS:
• YEAST EXTRACT MEDIA
• LAMIAN AIR FLOW
• YOUNG LEAVES
• ANTIBIOTICS
• SPIRIT LAMP
• ETHANOL
• PETRI PLATES
PROCEDURE:
1. First on the laminar air flow before starting the experiment and on
the uv light
2. Take the young leaves and wash them with the distilled water and
dry
3. Now add the autoclaved media in the petri dishes and leave it to dry
for one minute.
4. Now cut the leaves in the smaller pieces so that the pieces can be
placed in triangular format.
5. After placing all the leaves pack the petri dishes in such a way that
there will no way to contaminate the media.
6. Store this in the incubator for the 6 hours.
7. Now remove all the leaves and pack it and keep in the incubator
Result
The next morning you can see the yeast grown in the petri dishes
By this we can understand that yeast is present in the leaves
Visualizing the yeast under microscope
REQUIREMENTS:
• MICROSCOPE
• YESAT
• SLIDES
• COVERSLIP
PROCEDURE
1. Take the slide and wash it properly
2. Take the yeast from which the yeast was extracted from the leaf disc
method
3. Now place the coverslip in which there should be no air bubbles on
the slide
4. Now do observe on the microscope
OBSERVATION:
PREPARATION OF SLANTS
REQUIREMENTS:
• Yeast extract media
• Test tubed
• Laminar air flow
• Inoculation tubes
• Lamp
PROCEDURE
1. Take the sterile test tubes from the autoclave to avoid the contamination
from bacteria
2. Pour the yeast extract media in the test tube in such a way that through the
walls of the test tube
3. Now leave 2-3 min to dry
4. Now make the lines in each of the test tubes
5. Now pack the all the test tubes in the polythene bags cover it properly.
6. Now keep it in incubator for 6 hrs. and then observe
Result
Now you the see the yeast grown on the media from which we have inoculated.
MOLASSES
REQUIREMENTS:
• Molasses
• Urea
• Conical flask
• Water
PROCEDURE:
1. Take the 3 conical flasks
2. Now add 500 ml of water in each of the flask
3. Now add 2gm ,3gm and 4gm molasses in conical flask 1,2and 3
respectively
4. Then start heating on the hot plate so that the molasses get mixed with the
water completely
5. Again now place in the autoclave for the 15 min at 15 lbs.
6. Now place the stants in the incubator for 4-6 hrs.
Result
After incubation now you can see the yeast growth on the bottom of the flask
