SAS#7

This document outlines a student activity sheet for a biochemistry module on lipids. The activity involves solubility tests of different oils, spot tests to observe translucency, acrolein tests to detect odor, and tests for unsaturation using Hubl's solution. Students are instructed to record their observations and conclusions. They are also asked guiding questions about lipid characteristics, iodine number significance, lipid importance, and the definition of saponification of fats. The document concludes with a wrap-up asking students to identify the "muddies point" from the lesson.

Module #7 Student Activity Sheet

Name: _____________________________________________________ Class number: ____

Section: ____________ Schedule: _______________________________ Date: ___________

Lesson title: Lipids Materials: Pen, Paper, Module, Laboratory

Learning Targets: materials (See Main Lesson)
At the end of the module, students will be able to:
1. Explain the biological definition and function of lipids; References:
2. Know the occurrences and physical state of lipids; Josue, P.B. & Ocenar, M.M. (2014).
3. Determine the physical and chemical properties of lipids, BIOCHEMISTRY Laboratory Manual and
and Workbook for the Health Sciences. C&E
4. Explain the use of fats in saponification process. Publishing, Inc. Quezon City, Philippines

A. LESSON PREVIEW/REVIEW

GUESS THE IDENTITY OF THE SUGAR based on the following test results obtained.
Name of the sugar: _________________________
A. Upon addition of alpha-naphthol solution and H2SO4, the solution formed a violet ring at the junction of two layers.
B. Addition of Nylander’s reagent resulted into turbid solution
C. Color of the solution remains blue with Benedict’s reagent

B. MAIN LESSON

MATERIALS:
 Coconut oil  Hubl’s solution
 Vegetable oil  Cottonseed oil
 Water  Linseed oil
 Dilute HCl
 Litmus paper
 NaOH
 Alcohol  Test tube
 Chloroform  Filter paper
 Ether  Medicine dropper
 Potassium bisulfate

PROCEDURE:

Before
1. Use equipment only for its designated purpose.
2. Make sure you have complete materials and equipment. Wash them before use and check if everything is in good
condition.
3. To avoid mix-up and cross-contamination, organize and segregate the set of chemical reagents and equipment
needed in different procedure.
4. Make a label of the reagents or materials that you will use.
5. The acquired reagents should be put into an appropriate receiving and holding vessels such as test tube for liquids
and watch glass for solids.

During
NOTE: Results/Observation and Conclusions will be written down on the Check For Understanding Part.

A. SOLUBILITY TEST
 Put 2 drops of coconut oil in 1 ml of each of the following solvents contained in separate test tubes: water, dilute
HCl, dilute NaOH, ethanol, chloroform, and ether.
 Repeat the same procedure using Vegetable oil and Linseed oil instead of coconut oil. Tabulate the result on
the Check for Understanding Part.

B. TRANSLUSCENT SPOT TEST

Place 1 drop of coconut oil on a piece of paper. Do the same with vegetable oil and Linseed oil. Note the formation
of semi-translucent spot. Allow to evaporate spontaneously. Check the appearance of translucent spot.

C. ACROLEIN TEST
To 1 ml each of coconut oil, vegetable oil and linseed oil in their separate test tubes, add a pinch of Potassium
bisulfate. Heat gradually and note the odor produced.

D. TEST FOR UNSATURATION

1. Dissolve 5 drops of linseed oil 3 ml of chloroform.
2. Add Hubl’s solution drop by drop, shaking between additions, until a permanent brownish tinge is obtained.
3. Record the no of drops needed to obtain this color.
 No. of drops used: _________________________
4. Repeat steps 1 to 3 using cottonseed oil instead of linseed oil.

After
1. Dispose all waste materials into the laboratory waste bin. Observe proper segregation of hazardous and non-
hazardous chemicals in discarding them into the waste bin.
2. Clean all your laboratory apparatus and return them to the laboratory custodian or technician.
3. Clean your working area and sink.
4. Make sure to unplug all electronic devices used in the operation.
