BACTERIOLOGY MTAP (PART 2)

MEDIA USE
Blood agar medium  Enrichment/Differential
Mannitol Salt Agar  Selective (7.5% NaCl)
 Differential (Mannitol)
Columbia CAN agar  Colistin and Nalidixic Acid
 Selective for G+
Phenylethyl Alcohol (PEA) Agar  Used for determination of hemolytic reactions since
atypical reactions
Vogel-Johnson (VJ)  Selectivity (potassium tellurite, lithium chloride and
glycine)
 Inhibit nonstaphylococcal organisms
Chapman Stone Agar  Selective (5.5% NaCl)
 Differential (Mannitol and Gelatin)
Baird-Parker Agar  Selective (tellurite, lithium chloride)
 Demonstrate proteolytic activity

STAPHYLOCOCCUS

STAPHYLOCOCCUS
 Lactose Fermenter
 Halotolerant

KEY TESTS FOR IDENTIFICATION OF

STAPHYLOCOCCUS SPECIES

1. COLONY PIGMENT
 S. AUREUS (+) YELLOW
6. PYRROLIDONYL ARYLAMIDASE/ PYRase
2. STAPHYLOCOAGULASE TEST
 S. AUREUS (+)  S. AUREUS (-)
 S. SCHLEIFERI (+)
3. CLUMPIG FACTOR 7. OTHERS IN THE TABLE ABOVE
 S. AUREUS (+)
 S. LUGDUNENSIS (+)
 S. SCHLEIFERI (+) mimics tests  If the sample is from sterile site,
for staphylococcus aureus perform SUGAR TEST (the ability of the
organism to ferment sugar)
4. HEAT STABLE NUCLEASE
COAGULASE (+) STAPH DIFFERENTIATION
 S. AUREUS (+) Staph spp. VP PYR
 S. SCHLEIFERI (+) S. aureus + -
S. schleiferi + +
5. ALKALINE PHOSPHATASE S. hyicus - -
 S. AUREUS (+) S. intermedius - +
 S. SCHLEIFERI (+) S. delphini - +
BACTERIOLOGY MTAP (PART 2)
Cannot be differentiated in VP and
PYRase test

STREPTOCOCCUS

LANCEFIELD GROUP: Grouped according to the presence of sugar in the cell wall which serves as the antigen

COCOBACILLUS: Streptococcus pneumonia

Cultural characteristics:

1. Stab and Streak Method

 performed to assess the hemolytic reaction of the microorganisms when streptococcus is suspected
 Hemolysis is enhanced in anaerobic condition
2. Colonies with Depressed center
 Characteristic of S. PNEUMONIAE
 Because the organism produces autolytic enzyme
3. Mucoid colonies
 S. PNEUMONIA can produce capsul

OPTOCHIN : Enhances the organisms ability to produce AUTOLYSIS AMYDASE that lyses itself
