Unit 1 - Activity 4 - Factors Affecting Enzyme Lab

The document summarizes an online lab activity on factors affecting enzyme reaction rates. The maximum reaction rate occurred at pH 7. The optimal substrate amount was 2.0 g and pH level for lactase enzyme activity. Increasing the amount of lactase enzyme or raising the temperature could increase the reaction rate by providing more opportunities for collisions between substrate and enzyme.

Task: Complete the online lab and answer the following analysis questions.

Analysis Questions:
1. The maximum reaction rate occurred at what pH level?
pH7

2. The substrate amount needed to achieve the maximum reaction rate was
_2.0 g_?

3. What does the green line represent in the graph?

pH7

4. The optimal pH for lactase enzyme activity is _pH7___ according to the

graph.

5. Why was there no increase in the reaction rate with 8.0 g. of substrate as
compared to 4.0 g. of substrate? What would you need to add to see an
increase in the reaction rate with 8.0 g. of substrate?
It’s because the lactose material in the tube was insufficient to react with
that much lactose.

6. Observing only the results of the experiment conducted with 0.5 g of

lactose.
a. The pH level is the (independent variable or dependent variable)?
It’s a dependent variable.
b. The number of molecules of product formed per minute is the
(independent variable or dependent variable)?
It’s a dependent variable.
7. The maximum rate of this reaction is 350 molecules product/minute. List
two changes you could make in the experimental conditions or variables
that would increase this reaction rate. Explain why each change you listed
will increase the reaction rate.
The amount of lactase substance in the container could increase the
reaction rate. There would be more lactase for the lactose to react with.
Another condition is the increasing temperature. If the temperature
increases, the molecules movements would become faster and the
reaction rate would increase.
