Analytical procedure for testing iodine in salt by WYD

1. Principle:-

KI can be oxidized to I2 by KIO3 in the sample under acidic condition and blue iodine-starch compound is
formed in the solution, the concentration (mg/kg) of iodine was directly read out on the LCD of WYD
iodine checker.

2. Required calibration Standard solution, Chemicals and reagents


2.1 Calibration solution

2.1.1 KIO3 standard solution (1000ppm):- weigh 0.8432g KIO3 which was dried at 100-110℃for
3 hours and dissolve this in 500ml distill water.

2.1.2 Iodine working standard solution (10mg/ml - 20% NaCl ) take 5.0ml KIO3 standard
solution and add 100g NaCl and 0.5g Na 2CO3 a, add distill water to make 500ml, and shake the
solution thoroughly. This solution can be used within 6 months. 5.0 ml of solution equals the
concentration of 50mg/kg in the iodized salt sample.
2.2 Solution A and B
2.2.1 KI-starch solution (A):-

 Make solution a in a sterilized container


 Into 200 ml of boiling distilled water, add 2.0 g of soluble starch and stir thoroughly. Do not
add starch into distilled water until it is boiling vigorously, and then maintain boiling for two
minutes (heat to above 100ºc).
 After cooling, add 1.0 g KI (potassium iodide), 20.0 g K2HPO4.3H2O (di-potassium hydrogen
orthophosphate trihydrate) and 1.0 g Na2B407.10H2O (di-sodium tetra borate) and mix
thoroughly.
 This solution can be stored in a bottle for 6 months. If possible, store under refrigeration or
in a temperature range of 4-20 ºc. In warm weather store in the fridge. Solution A is the
primary reagent used, and proper storage and quality is essential for accurate results.
2.2.2 1 mol/L Sulphuric acid-H2SO4 (solution B)

 Mix 11 ml H2SO4 (sulfuric acid) with 190 ml distilled water evenly.


 Please note that when preparing the sulfuric acid solution, it is critical that the acid be
added to the water slowly.
 As sulfuric acid is a hazardous material, handling and proper disposal is necessary of this
chemical plus any leftover reagents, standard solution and sample specimens.
3. Equipments, glass wares required
 WYD iodine checker
 Analytical balance
 Glass or plastic test tube 50 ml
 Pipette 5 and 10ml

4. Analytical Procedure

4.1 Preparation of standard solution


 Transfer 5.0ml iodine working standard solution (10mg/ml) into 50ml tube
 Add 2 ml KI-starch solution (A) and 2ml H2SO4 (B)
 Mix the solution evenly, and add distill water to make 50 ml
 Shake the tube thoroughly, measure this solution, this solution equals concentration of
50 mg/kg salt.

4.2 Calibration of the WYD


 Turn on the WYD checker and warm it up for at least ten minutes
 Remove the 'blank" sample and fill the cell with the standard solution that was prepared
before. Once again wipe any excess moisture with a tissue paper.
 Place the cell with standard solution into the cell holder in the same way as done for the
blank sample. Close the lid
 Adjust the reading until 50.0 appears on the LED display by suing the Δ and ∇ keys.
 To verify the reading, insert 'gray' glass in order to calibrate the machine and record the
value for later use
4.3 Preparation of sample solution
 Weigh 1.0 g of well-mixed salt. You can use a 1.0 g plastic spoon to approximate the
amount in case there is no balance.
 Transfer the 1.0 g salt from the spoon to a 50 ml plastic test tube
 Add 10 ml of distilled water to the 50 ml plastic tube
 Shake the test tube thoroughly to dissolve the salt as much as possible
 Add 2.0 ml (one plastic pipette) of solution A into the 50 ml plastic test tube
 Add 2.0 ml (one plastic pipette) of solution B into the 50 ml plastic test tube
 Mix thoroughly
 add distilled water to the 50 ml plastic test tube till the mark (line)
 Shake the tube again thoroughly to mix
 Fill a clean cell (cuvette) with the solution containing the iodized salt-be careful to wipe
any excess moisture
 Place the cell in the wyd checker, close the lid and read the lcd panel for iodide
concentration
 Record the results an clean glass wares and all equipments

5. Measurement on WYD iodine checker

 Firstly zero point of this instrument is calibrated by distill water.


 Secondly, span calibration is performed by a standard solution (or gray glass).
 Fill a clean cell (cuvette) with the solution containing the iodized salt-be careful to wipe
any excess moisture
 Place the cell in the WYD checker, close the lid and read the LCD panel for iodide
concentration
 Record the results an clean glass wares and all equipments
