1.Intr What is protein ?
Protein is one of three most important
macronutrient that body needed to survive
and growth. Moreover, it is essential to helps
the body buid and repair cells and tissues.

It is made up of long chains of amino acids


and they differ from one another primarily in
the sequence of amino acids.
WHERE PROTEIN
CAN BE FOUND?
Protein are commonly found in animal
product such as meat and also present in
other sources, poultry, seafood, beans, eggs,
nuts and seeds.

HOW TO ANALYSIS?
Kjeldahl method was consumed to analysis the total
amount of nitrogen content in food then protein can
be calculated also. The Kjeldahl method involves a
three- step approach to the quantification of
protein: digestion, distillation, and titration.
Objective
The aim of this experiment is to determine the
protein content in black sesame by using
Kjeldahl method where it is the most frequently
used methods for measuring protein content in
foods.
2.Material and method
Material and Reagents
kjedhal apparatus Black Sesame
Heating a block Concentrated H2SO4
electronic balance Standard H2SO4 (0.5N)
glass tube digestion NaOH (35%)
erlenmeyer H3BO4 4%
pipettes Indicator Kjeldhal
pro pipettes Catalyst (K2SO4 and CuSO4)
burettes.
There are 3 steps to measure the amount of
Nitrogen in sesame seeds 3.Method

Step1: Digestion
Firstly, weigh 1.0284g of black sesame seeds in electronic balance and put it
in glass tube digestion then 7grams of K2SO4 was added and CuSO4 as a
catalyst.
Next, 20ml of concentrated H2SO4 was added
in glass tube digestion and
bring the glass tube digestion and mixture to a "rolling boil" by using a
heating a block at 370 oC for 2 hours. Until the reaction is complete, our
sample turns to aqua. Turn on the scrubber to eliminate H2SO4 odors that
emit into the atmosphere.
Step 2: Distillation
3.Method
Basically, prepare a new Erlenmeyer with
60ml of H3BO4 4% and add 3 to 4 drops of
indicator Kjedhal and it will turn to purple.
Next, put the glass tube digestion and solution
H3BO4 into the Kjedhal apparatus.

Then, turn
on the refrigerant which contain enough
NaOH 45% and water for distillation in order
to separate the ammonia which is a gas and
straightened by an Erlenmeyer contained
H3BO4 with an indicator to catch ammonia
Step 3: Titration
3.Method
Titration: Put stirrer bar in Erlenmeyer and stir
it with magnetic stirrer during titration with
H2SO4 (0.5N) until it reaches to equivalent
point and turns to purple. Note the volume
titrated.

Total nitrogen was

Protein was calculated by
0.014*N*V*Seedsame Factor*100

Result and Discussion
After determining, we got total of nitrogen
equal 0.0371 grams and protein equal 19.11
percentages. Which it is such a nice amount
of protein contain in black sesame.
4.Conclusion
After finished this experiment, we have fought out that
our experiment, which is to quantification the protein by
using the Kjeldahl method is really worked, and
according to the result and discussion, we also could
assume that one gram of black sesame is exactly
contained around 0.2g of protein.
