Lab Activity 1: Enzyme Activity
Enzymes are very important proteins in every living cell because they speed up chemical
reactions. They work at an optimum temperature and pH. Any changes in these will affect their
activity.
The enzyme catalase present in both plant and animal cells breaks down hydrogen
peroxide. In this lab activity, you will see the action of the enzyme catalase on hydrogen
peroxide and see the effect of temperature and pH on such action. (question)
Prepare the following:
Potato
Hydrogen peroxide (the one used for cleaning wounds)
Vinegar
5 uniform small jars or glasses
Syringe or droppers or small measuring cups for measuring liquids
Knife
Candle, burner or stove
Ruler
Water
Chopping board
Marker
Peel the potato and cut 4 pieces of about ½ inch cube each. Set aside
Boil for 5 minutes 1 cube of potato with enough water to submerge it. Remove it from the water
and cool it to room temperature. Set aside.
Cut one cube of potato into smaller pieces. Set aside.
Soak one cube of potato in a jar or glass with vinegar for 5 minute, making sure the potato is
submerged in the liquid.
Prepare 4 clean small glasses or glass jars, labeling them A, B, C, D.
Add an equal amount of hydrogen peroxide into each of the four glasses or jars, making sure
the liquid will be enough to submerge the potato cubes.
Add different cubes of potatoes to each of the glasses or jars of hydrogen peroxide as follows:
A - cube of potato
B - chopped potato
C- boiled cube of potato
D- potato soaked in vinegar
Observe the reactions by taking note of the bubble formation on the surface of the tissues.
Document your activity with pictures.
REcord your observations in your own table.
Make the Lab Activity REport following the format given.
Answer the following questions:
1, From your observations, which jar or glass showed the greatest activity of catalase?
2. What is the reaction and what is the equation?
3. How does the chopping of the potato differ from the whole cube set-up? How would you
account for the difference?
4. Explain how the boiling in water and soaking in vinegar affected the activity of the enzyme
catalase.
Write your conclusion.
