Method: Microscope number three (3) was removed from its rightful storage place and
placed on top of the table. The cord was unwrapped and plugged into the nearest
available socket. Using the lens cleaner, all lenses of the microscope were cleaned
thoroughly. The light intensity control knob was turned to ‘4’ and the field iris diaphragm
was opened. The light switch was then turned to the on position. The razor blade was
used to cut out a single letter ‘Y’ from the news paper and then one drop of water was
dropped on to the microscope slide in order to prepare a wet mount. Using the forceps,
the letter was placed on the drop of water. This was then covered with the cover slip.
being careful not to trap any bubbles beneath it, by placing the cover slip unto it at an
angle. The slide was then locked in place on the stage using the specimen holder. The
scanning lens was clicked into the 4X place and the light intensity control was set to four
(4). The stage control knobs were used to center the letter in the middle of the light and
directly below the lens. I looked through the ocular lenses at the image, which appeared
fuzzy at first but became clearer when adjustments were made using the course and fine
adjustment knobs. After doing a few adjustments, a clear image could be seen of the
letter that was on the slide. The specimen was then viewed under low power, (10x) and
then under high power (40x). The hair strand with the root follicle at the end was then
taken up using the forceps. Some water was sucked up using the dropper and this drop of
water was placed onto a microscope slide. The hair was placed on the drop of water and a
cover slip was used to cover the hair strand. The previous slide with the letter ‘Y’ was
removed and this slide with the hair strand was placed into the specimen holder. The
strand was then viewed under 4x, 10x and 40x magnifications respectively.
Another slide was then prepared, using a drop of the pond water as the specimen. Some
pond water was sucked up using the dropper, and a drop was placed onto a microscope
slide. This was covered with a cover slip and the slide was subsequently placed into the
specimen holder and viewed under 4x then 10x and finally 40x magnifications.
