Instructions:

1. Use this manual as reference during your lab session weekly (refer to lab schedule below).
2. You will be given experiments’ links based on the weekly experiments to watch.
3. You may use other reliable sources that available in online regarding the experiments to
assist you.
4. You should form a group consisting of 4-5 students before join the lab session. Every
student must be responsible to watch the videos provided and discuss among their
group members to complete the written lab report (GROUP).
5. Complete and submit the lab report to their respective lecturers via online platforms
before the due date. 1 lab report submission from each group together with datasheets
(all members from each group should complete and attach their datasheets together
with the lab report after the reference part).
6. Should you need any further clarification about the lab report, please consult with your
lecturer. 1 lab report only.
7. DO NOT alter this workbook.

Experiment: Microscopy and the Cells

Objectives

At the end of this lab session, students able to:


1. Describe the parts and functions of the compound light and dissecting microscope.
2. State the steps in proper order for bringing the cell’s image into focus with the compound
light microscope.
3. Calculate the diameter of field and the total magnification of the Cell’s image.
4. Identify the differences between prokaryotic and eukaryotic cells.
5. Identify the differences between animal and plant cells.

Materials

1. Compound light microscope


2. Stereomicroscope/ dissecting microscope
3. Slides

Procedure

1. Click the link stated in the lab schedule and watch the videos related to experiment:
Microscopy and the Cells.
2. Watch the experiment videos and write up ONE lab report for this experiment and submit
as Group lab report to the respective lecturer before the due date. Even though group lab
report, all the students in a group should submit the datasheets together with lab report. Can
refer other reliable online sources/books/e-books/journals to support the results and
discussion of your experiment.
3. Watch the week 5 lab demonstration video for discussion of objective No. 1.
4. Watch the week 7 lab demonstration video for discussion of objective No. 2.
5. Do hands-on experiment in the virtual lab using link provided (COMPULSORY: Install
adobe flash player before perform this experiment) for discussion of objective No. 3, 4 and
5.
6. Can choose any cell either onion root tip/cheek smear cell from the virtual lab to calculate
the total magnification of the cell’s image and the diameter of field.
7. Total magnification is calculated by multiplying of the Ocular lens (eyepiece) by the
magnification of the objective lens.
8. The diameter of the field (the circle visible through the lens) is the length of the field. This
can be measured by the provided ruler on the stage in the microscope in the virtual lab
while viewing through the eyepieces with different objectives lenses.
9. Refer the guidelines of the lab report.
10. Students should discuss the objectives of the experiment and relate to the results of
the experiment in the DISCUSSION part of the lab report. You may use other reliable
sources that available in online regarding the experiments to complete the lab report.

*Observe the bacterial cell, animal and plant cell slides in the Virtual Lab. By referring these
slides’ image, search diagram/microscopic photos from the online resources or can attach own
drawing photo based on the microscopic photos)

Note:
1. Choose either cheek smear cell or onion root tip cell from the virtual lab to complete this
table.
2. Total magnification of cell’s image can be calculated based on the Procedure No. 7.
3. Diameter of field can be measured based on the Procedure No. 8.
4. Use the ruler on the stage in the virtual compound microscope to measure the diameter of
field under the magnification of objective: 4X for the selected slide either onion root tip or
cheek smear cell.
5. Based on the diameter of field under the magnification of objective: 4X, calculate and
identify the diameter of field under the magnification of objectives: 10X and 40X.
6. The unit in the ruler is mm.
