1. Describe in details how to create a transgenic mice (8)
Firstly, a cloned DNA (gene) is injected into the nucleus of fertilized egg. The inoculated
fertilized mouse eggs are held under suction at the end of a pipette and the transgene is
microinjected into one of the pronuclei with a fine needle. The transgene integrates into the
chromosomal DNA of the pronucleus and is passed on to the daughter cells of eggs that
survive the process. The eggs then are implanted in the oviduct of pseudopregnant females,
and the transgenic pups are born after 19-20 days of gestation. Some of the offspring derived
from the implanted eggs carry the cloned gene in all of their cells. Animals with the cloned
gene integrated in their germ line cells are bred to establish new genetic lines.

2. Define vectors and provide one example of a vector (2)


A vector is a DNA molecule in which foreign DNA can be inserted or integrated and which is
further capable of replicating within host cell to produce multiple clones of recombinant
DNA.
E.g. Plasmid

3. Why are vectors used to clone genes? (2)


Because vectors are used as vehicles to artificially carry foreign genetic material into another
cell, where it can be replicated and expressed. Furthermore, vectors allow the exogenous
DNA to be inserted, stored, and manipulated mainly at the DNA level.

4. Define (6)
a) Transgene ~ A gene that is taken from the genome of one organism and introduced into
the genome of another organism by artificial techniques.
b) Chimeric ~ An organism or tissue that contains at least two different sets of DNA, most
often originating from the fusion of as many different zygotes (fertilized eggs).
c) Wild-type ~ A term used to describe a gene when it is found in its natural, non-mutated
(unchanged) form.
