Pre-Laboratory Activity 1
The Compound Microscope

Pre-Lab Questions
1. What is the importance of microscopes in Forensic Chemistry?
The importance of microscopes in Forensic Chemistry is maintaining the
prominent position as an important tool for examination of trace amounts of evidence.
The microscope is used by forensic scientists to locate, isolate, identify, and compare
samples. Because of its low magnification, wide field of view, large working distance,
and stereoscopic vision, the stereomicroscope is used for preliminary evidence
evaluations. It also allows for comparative examinations for evidence such as paint,
physical matches, questioned documents, and impression evidence and can also be
used to identify and compare forensic samples.
2. Label the part of a Microscope.

1. Revolving Nosepiece 5. Light Source 9.Coarse Adjustment

2. Objective Lens 6. Eyepiece 10. Stage Control

3. Stage Clips 7. Arm 11. Base
4. Diaphragm 8. Mechanical Stage

3.What are the two types of diaphragm used as a part of a microscope?

Two basic of diaphragm are utilized in a microscope: The Field Diaphragm

Control - The base of the microscope contains the field diaphragm. This controls the size
of the illuminated field. The field diaphragm control is located around the lens located in
the base. And the Condenser Diaphragm- This diaphragm controls the amount of light
entering the lens system. This feature is useful for viewing unstained biological
specimens that are translucent. Reducing the amount of light improves contrast, making
the specimen "stand out" against the background.
