Intended Use
The LightCycler® 480 Instrument is intended for performing rapid, accurate polymerase chain reaction (PCR)
in combination with real-time, online detection, enabling Absolute or Relative Quantification of a target nucleic
acid, as well as post-PCR analysis of the amplified nucleic acid by Melting Curve analysis.

The LightCycler® 480 Instrument is intended for life science research only and must be used exclusively by
laboratory professionals trained in laboratory techniques and having studied the Instructions for Use of this
instrument. The LightCycler® 480 Instrument is not for use in diagnostic procedures.

About LightCycler® 480 Service Pack SP3

The LightCycler® 480 Service Pack SP3 is designed for LightCycler® 480 Instruments, Versions I and II. Owners
of the LightCycler® 480 Software Version 1.5.1 can download this service pack to update their software.

The LightCycler® 480 Service Pack 3 Software implements compatibility with the new hardware configuration.

The Service Pack SP3 can be installed on both the LightCycler® 480 control unit and client PC. The update
for the LightCycler® 480 Software, Version 1.5.1 SP3 is free of charge and includes all updates introduced with
Service Pack SP1 and Service Pack SP2.

How to Get the LightCycler® 480 Service Pack SP3

The latest LightCycler® 480 System software packages, version 1.5.1 and Service Pack SP3 can be downloaded
by instrument owners at www.lightcycler.com.

To download LightCycler® 480 System software packages, first register by completing the registration form on
the Internet (www.lightcycler.com) under the tab Software. Under Application, select “LightCycler 480”. After
successful submission of all information required in the form, an email with the respective download link will
be provided.

For further assistance regarding Service Pack SP3, please contact your local LightCycler® 480 Instrument spe-
cialist.

Prerequisite for Installing the LightCycler® 480 Service

Pack SP3
The LightCycler® 480 Service Pack SP3 is applicable for the LightCycler® 480 Software Version 1.5.1 only;
ensure that one of the following software versions is installed on the control unit (or client PC):

c LightCycler® 480 Software Version 1.5.1

c LightCycler® 480 Software Version 1.5.1 SP1
c LightCycler® 480 Software Version 1.5.1 SP2

Installing the LightCycler® 480 Service Pack SP3

LightCycler® 480 Software, Version 1.5.1 Service Pack SP3 is provided as a ZIP file: LC480SW_1.5.1_SP3.zip.
The file size is approximately 6.7 MB.

Use these Simple Steps to Install LightCycler® 480

Software Version 1.5.1 Service Pack SP3
1 Unzip the LightCycler® 480 Service Pack SP3 and save the extracted files on a memory stick for transfer to
the LightCycler® 480 Instrument control unit (or client PC).

2 Make sure that the LightCycler® 480 Software Version 1.5.1 is installed on the LightCycler® 480 control unit
(or client PC).

3 Restart the control unit and log in with the user name LCinstall and password LCinstall.
Do not start the LightCycler® 480 Software.
To update with Service Pack SP3, it is not necessary to shut down the Exor Database Server.

4 Turn on the LightCycler® 480 Instrument and wait until the left-side LED turns to green.

5 Insert the memory stick into the LightCycler® 480 control unit (or PC).

6 Double-click the LightCycler480_Software_SP3_Setup file to start the installation.

7 Click Yes to confirm the security warning: “Do you want to allow the following program from an unknown
publisher to make changes to this computer?”

8 Click Next to confirm the installation, as shown below.

9 Click Finish when the installation is completed.

10 Start the LightCycler® 480 Software Version 1.5.1 SP3 and log in.

11 After logging in, the software will automatically download the new firmwares on the connected instrument
(this may take up to 10 minutes), then the instrument will reinitialize.
Please wait until the initialization of the instrument is completed. Do not turn off the control

unit or the instrument during this step.

12 Open the LightCycler 480 Software and make sure that “LightCycler 480 Software release 1.5.1.62 SP3” is
displayed in the Program Bar, as shown in the picture below (left from the red arrow).
