I. INTRODUCTION
DNA extraction can be considered as one of the breakthrough in field of molecular
biology. This discovery gives way to different advancement that helps people. DNA
extraction makes it possible to study the genetic causes of diseases and for the
development of diagnostics and drugs. DNA extraction also becomes an essential
process in field of forensic science. Sequencing genome, detecting bacteria, and viruses
in the environment and for detecting paternity become possible with the help of DNA
extraction.
In 1869, the first method of extracting and purifying DNA was performed by Friedrich
Miescher. Then as time goes by, the method of extracting DNA become specialized and
easier. Such changes have both sped up production and increased the yield of DNA. One
good example is the simulation of DNA extraction in banana.

1. Place 1 tablespoon of 70% Isopropyl alcohol in a clean vial. Then prepare a ice bath by
putting crushed ice into a plastic container. Put the vial into the ice bath. Set it aside.
2. Obtain a fresh piece of peeled banana and place it in a plastic up. Then place ½ cup of
distilled water and mash it using a stainless fork. Do it until the banana is completely
mashed. Set it aside.
3. In another plastic cup, place ¼ tablespoon salt, 1 teaspoon dishwashing liquid, and 2
tablespoon water. Stir it using a coffee stirrer until the salt is dissolved. Make sure that
there is no bubble formed when mixing the solution.
4. Add 2 tablespoon of the banana mixture that you prepared earlier into the solution that
you prepared in step 3. Mix it again and make sure no foam is formed.
5. Get a coffee filter and wet it with water. Put the coffee filter on top of the plastic up.
Pour down the mixture that you prepared in step 4 and let it pass through the coffee
filter.
6. Add the filtrate that you collected inside the vial that you prepared in step 1. Stir it
slowly. Observe the goo-like white small particle formed that is floating on top of the
alcohol layer. That is the DNA from the banana.
7. Make sure the DNA is fully formed before you scoop it using the coffee filter. If you can’t
scoop it, return the vial into the ice bath and let it cool down before scooping the DNA
again.
