Data Collection

Subject Selection
Subjects will be isolated from ten pools, five from each Public and Private pools in Metro
Manila. The colony of Pseudomonas aeruginosa having abundant growth on
Cetramide agar will be the subject of the study.
Data Collection
Data will be obtained from public and private swimming pools in Metro
Manila. Physical and chemical analysis requires a liter of sample to be used
and inoculated in Centrimide agar and to be performed in Water Laboratory
Inc. (water testing laboratory). The samples will be collected in a sterile
bottle to avoid contamination or errors on the results. Proper handling and
sampling techniques must be performed. Contaminations must be avoided.
Unwanted contaminations must be removed by cleaning it with distilled
water and drain it before use. Sample containers should be sterile and should
be filled with the desired volume. It also must be properly labeled and it
should be kept cooled or refrigerated.

By using aseptic technique, grasp the sample container around the


lower half to reduce the chance of finger contamination.

Carefully remove the cap from the bottle and be careful not to touch
the inside of the sterile bottle or cap with your fingers.

Plunge the container into water (lip of bottle first) until container is
approximately 1 foot below surface of the water.

Turn open end of bottle into the direction of current and allow container
to fill. If there is no current, create one by slowly moving bottle in a
direction away from hand.

Fill it to the shoulder or to about 1/4 inch from the top.

Remove container from water. Screw the cap on the bottle and dont
touch the inside of the cap.

Pack the sample. Samples should be placed on ice/ice packs during


transit to laboratory. The temperature of the sample must be kept or
maintain below 10 degrees Celsius.

Data Processing and Analysis Multistage sampling will be used in collecting


twenty (10) water samples from five (5) public and five (5) private pools in Metro
Manila ,the water samples collected will be isolated and inoculated using
Cetrimide agar to observe for the growth of Pseudomonas aeruginosa. A
presence of blue green pigment on the medium will show a positive result of the
organism.
The growth of the colonies of Pseudomonas aeruginosa on the Cetrimide
agar, a plate count will be conducted using the automated colony counting
chamber to count the growth of the organisms. The results will be recorded and
will be analyzed to compare which of the water samples coming from the 5
public and 5 private pools in Metro Manila has the highest concentration of
Pseudomonas aeruginosa. Statistical test, will be used to detect significant
difference between the concentration of the Pseudomonas aeruginosa present in
public and private pools in Metro Manila.
