The pentose phosphate pathway occurs in the cytosol and consists of an oxidative and nonoxidative phase. The oxidative phase generates NADPH which is used for reductive biosynthesis. The nonoxidative phase interconverts 5-carbon sugars to produce ribose-5-phosphate for nucleotide biosynthesis and glycolytic intermediates. Key enzymes in the oxidative phase include glucose-6-phosphate dehydrogenase and 6-phosphogluconate dehydrogenase. The pentose phosphate pathway provides NADPH for fatty acid, cholesterol and neurotransmitter biosynthesis as well as glutathione reduction. Tissues with active pentose phosphate pathways include the liver, adipose tissue and red blood cells.

 Occurs in the cytosol
 Meets the need of all organisms for a source
of NADPH to use in reductive biosynthesis
 consists of two phases:
Oxidative generation of NADPH when glucose-6-
phosphate is oxidized to ribose-5-phosphate
Nonoxidative interconversion of 3-, 4-, 5-, 6- and 7
carbon sugars to synthesize five-carbon sugars
for two purposes:
 Biosynthesis of nucleotides
 Production of excess into glycolytic pathway intermediates
