Important Terms Related to Staining

Dyes – Chemicals used to add color to the cell.


Acidic Stain – are anionic, negative charge (Sodium, Potassium, Calcium)
Basic Stain – are cationic, positive charge (Chloride, Sulfate salts)
Staining – The process of adding color to a bacterial cell
Simple Stain- uses only 1 dye.

Differential Stain – uses 2 or more dyes

Crystal violet – Iodine crystal formation in cell
Gram + Alcohol dehydrates cell wall
CV-I, crystal formation is unable to leave cell

Gram – Alcohol dissolves outer membrane, leaving wholes
in wall, thus allowing CV-I to wash out
Heat Fix – fixes bacterial smear to slide, preventing smear from washing away during staining
Chromogen – Colored compound, not stain
